Bitter taste, an organoleptic characteristic of virgin olive oil, has been related to phenolic compound composition. The usual method to assess this attribute is by a sensorial panel of tasters, while in the laboratory; methods based on physicochemical properties have been assayed as K225, the most widely used one. However, a direct determination of bitterness in virgin olive oil is useful for quality-control purposes. The proposed method is supported by the observable spectral change undergone by the compounds responsible for bitterness as pH varied. This measurement was carried out directly in the oil, without prior isolation of bitter analytes. The difference of absorbance between alkaline and neutral medium showed a highly significant correlation (r = 0.988, p < 0.0001) with the conventional parameter (K225). The method was rapid, required a small sample, allowed direct determination of bitterness in virgin olive oil, and could be easily automated. 相似文献

Accumulation of beta-carotene and trans-cis isomerization of ripening mango mesocarp were investigated as to concomitant ultrastructural changes. Proceeding postharvest ripening was shown by relevant starch degradation, tissue softening, and a rising sugar/acid ratio, resulting in a linear decrease (R (2) = 0.89) of a ripening index (RPI(KS)) with increasing ripening time. A modest accumulation of all-trans-beta-carotene and its cis isomers resulted in a slight pigmentation of the mango chromoplasts, because ambient temperatures of 18.2-19.5 degrees C provided suboptimal ripening conditions, affecting color development and beta-carotene biosynthesis. The ultrastructures of chromoplasts from mango mesocarp and carrot roots were comparatively studied by means of light and transmission electron microscopy. Irrespective of the ripening stage, mango chromoplasts showed numerous plastoglobuli varying in size and electron density. They comprised the main part of carotenoids, thus supporting the partial solubilization of the pigments in lipid droplets. However, because different pigment-carrying tubular membrane structures were also observed, mango chromoplasts were assigned to the globular and reticulotubular types, whereas the crystalline type was confirmed for carrot chromoplasts. The large portions of naturally occurring cis-beta-carotene in mango fruits contrasted with the predominance of the all-trans isomer characteristic of carrots, indicating that the nature of the structure where carotenoids are deposited and the physical state of the pigments are crucial for the stability of the all-trans configuration. 相似文献

Nitrogen (N) by form of nutrition, ammonium (NH4+) or nitrate (NO3?), affects metabolic and physiological processes of plants. In general, a high proportion of N in NH4+ form results in poor growth. Nonetheless, a number of species exhibit optimum growth when high levels of NH4+ are provided. In the present study, lisianthus [Eustoma grandiflorum (Raf.) Shinn] was grown in rockwool cultures and irrigated with nutrient solutions containing 15 mM N with varying proportions of NH4+ and NO3?. The results showed that an increase in NH4+-N form increased plant height, number of flowers and leaves, leaf area, and shoot, stem, and leaf dry weight. The proportion of NH4+ also affected leaf concentration of phosphorus, potassium (K), calcium (Ca), and magnesium (Mg), although leaf N concentration was unaffected. Potassium leaf concentration was higher when a low proportion of NH4+ was supplemented in the nutrient solution; however, plants exhibited a decrease in leaf K concentration and a decrease in leaf Ca as the proportion of NH4+-N increased. Shoot dry weight was higher with low leaf K whereas high leaf Ca was associated with high shoot dry weight. Net photosynthesis rate was higher in plants irrigated with solutions containing 75% of total N in NH4+ form than in those irrigated with solutions of 0 or 25%. Phosphorus (P) and zinc (Zn) are important determinants of plant productivity, particularly in the tropical grasslands of Brazil. Nutrient deficiency is one of the most important factors limiting plant productivity, decreasing photosynthesis efficiency and plant development. The present study investigates in Brachiaria brizantha (Hochst. ex A. Rich.) Stapf. cv. ‘Marandu’: 1) the gas exchange measurements; 2) the total leaf area development; and 3) the dry matter production due to P and Zn nutrition. Plants of B. brizantha cv. ‘Marandu’ were grown in nutrient solution under five rates of P (0.1, 0.6, 1.1, 1.6, and 2.1 mmol L?1) and five rates of Zn (0.00, 0.75, 1.5, 2.25, and 3.00 μmol L?1), in a fractioned factorial. Plants were harvested two times. Phosphorus supply increased carbon dioxide (CO2) assimilation and stomatal conductance, and decreased intercellular CO2. The interaction P rates x Zn rates were significant for the total leaf area variables and shoot dry matter in the second growth period. The nutrition of P and Zn interfered in the B. brizantha productivity by changing the grass photosynthesis and leaf area. 相似文献

The seed storage protein composition of one collection of cultivated einkorn wheat (Triticum monococcum L. ssp. monococcum) of Spanish origin has been analysed by SDS-PAGE and A-PAGE. Three allelic variants were detected for the Glu-A1m, whereas up to six alleles were detected for the Glu-A3m. For the gliadins, 7 and 14 alleles for the Gli-A1m and Gli-A2m were found between the evaluated accessions. Internal variability was detected in some of these materials, which could be
related to the landrace nature of them. Up to 48 different genotypes based on the origin and seed storage protein composition

The antiproliferative effects of 100% methanol crude extract and of Toyopearl and silica gel fractions from the seed coats of black Jamapa beans (Phaseolus vulgaris L.) were evaluated using HeLa, human adenocarcinoma cells, and HaCaT, human premalignant keratinocytes. The 100% methanol crude extract [172.2 microM equiv of (+)-catechin] increased adhesion of HeLa cells; however, 3- and 5-fold higher concentrations decreased the number of cells attached as a function of the treatment time. The highest concentration tested diminished the cell adhesion until 40% (after 24 h) to almost 80% (after 72 h). The IC50 values showed that the 100% methanol crude extract was the most effective inhibitor of HeLa cell proliferation, even when it was dissolved in dimethylsulfoxide (DMSO) [34.5 microM equiv of (+)-catechin] or in medium [97.7 microM equiv of (+)-catechin]. The Toyopearl 5 (TP5) fraction and silica gel 2 (SG2) fraction inhibited 60% of the HeLa cell proliferation. The IC50 was 154 microM equiv of (+)-catechin of the 100% methanol crude extract on HaCaT cells. Toyopearl fractions TP4 and TP6 significantly inhibited HaCaT cell proliferation, but the silica gel fractions did not have a significant effect. The 100% methanol crude extract (35 microg of dry material/mL) decreased the number of HeLa cells in the G0/G1 phase from 68.9% (for control cells) to 51.4% (for treated cells) and increased apoptosis (2.9 and 21.2% for control and treated cells, respectively). The results indicated that black Jamapa beans could be a source of polyphenolic compounds, which have an inhibitory effect toward HeLa cancer cells but are less aggressive on HaCaT premalignant cells. 相似文献

Andean potato varieties are cultivated in the northwest of Argentina and constitute the most important staple food for the
local farmers. The genetic diversity of 155 accessions conserved at the Genebank of Balcarce (INTA) was tested using four
microsatellites. Three commercial potato varieties of Tuberosum group and one accession of Curtilobum group were used as outgroups.
The presence of bands was scored for each microsatellite and for each accession and the data were analysed by principal coordinate
analysis. The polymorphism information content was obtained for each molecular marker from banding patterns. Analysis of molecular
variance was carried out with a variable number of accessions for each landrace, from different departments and sites within
departments. More than one genotype was detected in the majority of the potato landraces. Some accessions within each landrace
did not differentiate. AMOVA revealed that most of the genetic variation occurred among sites within departments and among
local varieties. These findings are discussed considering the agricultural practices carried out in the Andean farming system. 相似文献

Although cyclodextrins (CDs) have been successfully used as antibrowning agents in different fruit juices, no research has studied the effect of these compounds on enzymatic browning in peach juice. In this paper, the color of fresh peach juice was evaluated in the presence of two types of natural (alpha-CD and beta-CD) and a modified (maltosyl-beta-CD) CD, and the effectiveness of these compounds as browning inhibitors was determined using the color space CIELAB system. Moreover, to clarify the mechanism by which CDs inhibit peach juice enzymatic browning, the process was kinetically modeled in the absence and presence of CDs using a colorimetric method; the apparent complexation constants between the mixtures of diphenols present in peach juice and some types of CD were calculated. The results show that the highest affinity constant was presented by alpha-CD (Kc = 18.31 mM-1) followed by maltosyl-beta-CD (Kc = 11.17 mM-1), whereas beta-CD was incapable of inhibiting peach juice enzymatic browning. The effects of fruiting body maturity on chemical composition and antimicrobial activity of the wild mushrooms, Lactarius deliciosus and Lactarius piperatus, were evaluated. Immature and mature samples were analyzed for proximate constituents (moisture, fat, crude protein, ash, carbohydrates), nutritional value, and individual composition in fatty acids and sugars. Protein content, MUFA (monounsaturated fatty acids), and PUFA (polyunsaturated fatty acids) percentages increased with the fruiting body maturity stage, while carbohydrate and SFA (saturated fatty acids) content decreased. The maturity stage did not significantly affect the individual sugar profile. The antimicrobial activity of the mushrooms was screened against Gram positive and Gram negative bacteria and fungi, and correlated to the amounts of phenols, flavonoids, ascorbic acid, beta-carotene, and lycopene present in the immature and mature fruiting bodies. Mature fruiting bodies with mature spores presented lower antimicrobial activity, which was in agreement with the bioactive compound content found in those samples. 相似文献
