Most follicles undergo atresia during the developmental process. Follicular atresia is predominantly regulated by apoptosis of granulosa cells, but the mechanism underlying apoptosis via the mitochondria‐dependent apoptotic pathway is unclear. We aimed to investigate whether the mitochondria‐associated genes peroxisome proliferator‐activated receptor‐gamma, coactivator1‐alpha (PPARGC1A), nuclear respiratory factor‐1 (NRF‐1), B‐cell CLL/lymphoma 2 (BCL‐2) and BCL2‐associated X protein (BAX) played a role in follicular atresia through this pathway. The four mitochondria‐associated proteins (PGC‐1α, which are encoded by the PPARGC1A gene, NRF‐1, BCL‐2 and BAX) mainly expressed in granulosa cells. The mRNA and protein levels of PPARGC1A/PGC‐1α and NRF‐1 in granulosa cells increased with the follicular development. These results showed that these genes may play a role in the regulation of the follicular development. In addition, compared with healthy follicles, the granulosa cell in atretic follicles had a reduced expression of NRF‐1, increased BAX expression and increased ratio of BAX to BCL‐2 expression. These results suggested that changes of the mitochondria‐associated gene expression patterns in granulosa cells may lead to follicular atresia during goat follicle development.  相似文献

The root-knot nematodes (Meloidogyne spp.) are important nematode pests and cause serious diseases in pepper in the world. No molecular markers linked to the nematodes resistance N gene have been reported. In this paper, ‘Carolina Wonder’ (Capsicum annuum L.), a sweet pepper line resistant to root-knot nematode with N gene, ‘20080-5-29’ (C. annuum L.), an inbred line susceptible to root-knot nematode with good horticultural characteristics, and their F2 progeny with 320 individuals were used as materials. Evaluation of resistance and susceptibility of parental lines, F1 and F2 progeny inoculated with root-knot nematodes (Meloidogyne incognita) were carried out. ‘Bulked segregant analysis’ method was used to search for polymorphic markers from 512 pairs of AFLP primers. Based on the assessment of resistance and susceptibility and polymorphism of the AFLP marker in F2 population, the genetic linkage distance between the AFLP marker and the N gene was estimated. One AFLP marker E39/M41-339 was obtained and transferred to a SCAR marker amplifying a 315 bp DNA fragment linked to the N resistant allele and a 331 bp fragment linked to the N+ susceptible allele. The distance between the molecular marker and the nematodes resistance N gene is 6.3 cM. This research delivered a valuable tool for the marker assisted selection of nematodes resistance in pepper.  相似文献

A radiolabelling method is generally used to determine the foliar uptake of xenobiotics. This technique cannot provide any information on the localization of chemicals inside leaf tissues. The influence of an alcohol ethoxylate surfactant on the uptake of three fluorescent dyes, difluorofluorescein (hydrophilic), rhodamine B (moderately lipophilic) and a naphthalimide dye (lipophilic), into the leaves of three contrasting species, bean (Vicia faba), wheat (Triticum aestivum) and cabbage (Brassica oleracea), at 16 h after treatment was measured using a conventional wash‐off method and also visualized in vivo by confocal laser scanning microscopy (CLSM). Whereas the lipophilic dyes showed greater intrinsic uptake than the hydrophilic one, the enhancing effect of the surfactant on uptake was more pronounced for the latter. CLSM revealed that the presence of the surfactant increased the transport of difluorofluorescein into the epidermal cells of bean and wheat leaves, but not cabbage leaves. Rhodamine B showed greatest transcuticular diffusion in all three species, but most of the dye moved into the vacuole of the epidermal cells. The naphthalimide dye was strongly retained by the wax–cuticle layer of all species, even in the presence of the surfactant. CLSM has proven to be an attractive tool for studying xenobiotic diffusion. The results obtained using fluorescent dyes are believed to be applicable to the foliar uptake of herbicides.  相似文献

ABSTRACT

Close relationships usually exist among biomass accumulation, nutrient uptake, and seed yield during the growing season. Field experiments with pea (Pisum sativum L.) and lentil (Lens cultinaris L.) were conducted in 1998 and 1999 at Melfort, Saskatchewan, Canada, to determine relationships of biomass accumulation and nutrient uptake with days after emergence (DAE) or growing degree days (GDD). For both biomass accumulation and nutrient uptake, maximum rates and amounts increased with time at early growth stages and reached a maximum value at late growth stages. The R2 values for cubic polynomial regressions were highly significant, indicating their suitability to estimate the progression of biomass accumulation and nutrient uptake as a function of days after emergence (DAE). Both pulse crops followed a similar pattern in biomass accumulation and nutrient uptake, which increased in the early growth stages and reached a maximum late in the growth cycle. Pulse crops usually reached their maximum biomass accumulation rate and amount at early to late bud formation (42–56 DAE or 390–577 GDD) and at medium pod formation to early seed filling (75–82 DAE or 848–858 GDD) growth stages, respectively. Maximum biomass accumulation rate was 175–215 kg ha? 1d?1 for pea and 109–140 kg ha? 1d? 1 for lentil. Maximum nutrient uptake rate and amount usually occurred at branching to early bud formation (28–49 DAE or 206–498 GDD) and at the flowering to seed filling (66–85 DAE or 672–986 GDD) growth stages, respectively. Maximum uptake rate of nitrogen (N), phosphorus (P), potassium (K), and sulfur (S), respectively, was 4.6–4.9, 0.4–0.5, 5.0–5.3 and 0.3 kg ha? 1d? 1 for pea, and 2.4–3.8, 0.2–0.3, 2.0–3.4 and 0.2 kg ha? 1d? 1 for lentil. In general, maximum nutrient uptake rate and amount occurred earlier than maximum biomass accumulation rate and amount, respectively; and the maximum accumulation rates of both biomass and nutrients occurred earlier than maximum amounts. The findings suggest that adequate supply of nutrients from soil and fertilizers at early growth stages, and translocation of biomass and nutrients to seed at later growth stages are of great importance for high seed yield of pulse crops.  相似文献

Theileria equi (T. equi) is an obligate intra- and extra-erythrocytic parasite that causes equine theileriosis (ET) in equids. Equine theileriosis is considered a notifiable disease of global significance, a major constraint to the international movement of horses, and endemic in many countries. This disease may be difficult to diagnose, as it can produce variable and nonspecific clinical signs. A cross-sectional study was designed for the molecular characterisation of T. equi and to investigate the associated risk factors of ET accompanied by its consequences on haematological and sero-biochemical parameters. A convenience sampling of 500 blood samples were collected from ET suspect horses from January to December 2017. PCR was performed on all blood samples targeting the 18S rRNA gene of T. equi followed by sequencing; 9% animals tested positive with confirmed sequences. The isolates of this study showed high homology with Cuban, Russian and Brazilian isolates of T. equi (accession numbers KY111762.2 , MG551915.1 and KY952237.1 , respectively). Based on multivariate analysis, the principal risk factors consisted of absence of dogs on the premises and presence of tick infestation. The haemato-biochemical parameters showed a decrease in granulocytes and erythrocytes, and an increase in lymphocytes, monocytes, mean corpuscular volume, mean corpuscular haemoglobin, mean platelet volume, glucose, phosphorus and aspartate aminotransferase in positive horses. This is the first study which identified ET in Punjab (Pakistan) using molecular techniques and risk factors together with the haemato-biochemical variations in horses.  相似文献

While some composting had been practiced on a limited basis in Nigeria 50 years ago, the practice still is largely neglected. Farmers find it more convenient to use fertilizers because of the ease of carrying, storing and applying. However, since 1986, there has been a major change in the composition of refuse in Nigeria. With greater numbers of people in the low socioeconomic groups scavenging on the refuse dumps collecting paper, metal, glass and plastics, the remaining refuse is mostly organic and ideal for making compost. Studies undertaken at the University of Ibadan reveal that once mobilized, communities can be effective in launching composting projects. Appropriate technology is favored in processing the wastes rather than extensive and, often unaffordable, mechanization.  相似文献

The objective of this study was to evaluate the effects of continuous low dose infusion of lipopolysaccharide (LPS) on inflammatory responses and milk production and quality in lactating dairy cows. Eight Holstein cows were assigned to two treatments in a cross‐over experimental design. Cows were infused intravenously either with saline solution or with saline solution containing LPS from Escherichia coli O111:B4 at a dose of 0.01 μg LPS/kg body weight for approximately 6 hr each day during a seven‐day trial. The clinical symptoms and milk production performance were observed. Milk samples were analysed for conventional components, fatty acids and amino acids. And jugular vein and mammary vein plasma samples were analysed for concentrations of cytokines and acute phase proteins. LPS infusion decreased feed intake and milk yield. An increase in body temperature was observed after LPS infusion. LPS infusion also increased plasma concentrations of interleukin‐1β, serum amyloid A, LPS‐binding protein, C‐reactive protein and haptoglobin. LPS infusion decreased the contents of some fatty acids, such as C17:1, C18:0, C18:1n9 (trans) and C18:2n6 (trans), and most amino acids except for methionine, threonine, histidine, cysteine, tyrosine and proline in the milk. The results indicated that a continued low dose infusion of LPS can induce an inflammatory response, decrease milk production and reduce milk quality.  相似文献

The traits of elongation, volume expansion, and water absorption are very important in determining the quality of cooked rice grains. In this study, quantitative trait loci (QTL) analysis of these traits was performed using a recombinant inbred population derived from a cross between two indica cultivars, ‘Zhenshan 97’ and ‘Minghui 63 ,’ which are the parents of the most widely grown hybrid rice in China. Using a linkage map based on 221 molecular marker loci covering a total of 1796 cM, a total of 33 QTLs were identified for the nine traits tested. QTLs were detected on chromosomes 1– 3 , 5– 9 , and 11 , respectively. The QTLs identified included three for cooked rice grain length elongation (chromosomes 2 , 6 , and 11), six for width expansion (chromosomes 1‐ 3 , 6 , 9 , and 11) and two for water absorption (chromosomes 2 and 6). Interestingly, a single QTL located near the wx gene on chromosome 6 seemed to influence all the traits tested for the cooked rice quality.  相似文献

The present study investigated the chemical coding of neurons in the celiac-superior mesenteric ganglion complex supplying the normal (n=4) and inflamed (n=4) ileum (chemically-induced inflammation) in juvenile pigs using retrograde tracing combined with immunohistochemistry. Ileum-projecting neurons (IPN) were predominantly distributed in the left and right superior mesenteric pools of the ganglion. The majority of them were adrenergic (tyrosine hydroxylase-positive) and also contained neuropeptide Y, somatostatin or galanin. No clear-cut differences in the distribution and chemical coding of IPN were found between normal and inflamed pigs. However, in the inflamed group, the density of peptidergic, IPN-associated nerve fibres was higher than that found in the control group.  相似文献

Iron and manganese removal from water is considered a routine operation in most potable water treatment plants. However, when there is dissolved organic carbon (DOC) present in the water source, it can significantly influence the success of iron and manganese removal. This investigation focused on the use of two different coagulants, alum and ferrichloride, used in conjunction with hydrogen peroxide as an oxidant, to reduce dissolved iron and manganese in water with a high DOC loading. The two coagulants behaved in a totally different manner during the treatment process, with alum being successful in reducing the iron content in the treated water, and ferrichloride yielding a good removal of the dissolved manganese present. Since these results were only possible in cases where hydrogen peroxide concentrations were also present, it was concluded that treatment with a single reagent would not yield the desired removal of contaminants from this specific water. Both treatment options significantly reduced the DOC levels in the water to below the desired concentration after treatment.  相似文献
