High sugar content of sorghum stalk is an important factor in the sorghum silage production. To identify the genomic regions controlling sugar content and to develop molecular markers linked to sugar content in sweet sorghum, we used an Early Folger, and a normal inbred line, N32B, for genetic linkage mapping and quantitative trait locus (QTL) analysis. We constructed a genetic linkage map spanning 983.5 cM based on a total of 327 markers comprising 31 restriction fragment length polymorphism (RFLP) markers, 254 amplified fragment length polymorphism (AFLP) markers, and 42 simple sequence repeat (SSR) markers. In the 20 linkage groups detected, 98.2% of markers aligned to the 10 linkage groups of sorghum.Variations in sugar content at different growth stages and among internodes suggested that the sugar content of middle internodes is stable and suitable for measuring at early dough stage. The broad sense heritability (hB2) of sugar content was 0.64 and 0.62 estimated from the data of F3 families and each parent in 2003 and 2004. We identified one and two QTLs accounting for 22.2 to 25.0% of phenotypic variance using simple interval mapping method in 2003 and 2004, respectively.These two QTLs showed a negative additive effect, and over-dominance effect. A QTL on LG-D was detected in both two years. Above results will be help us to understand the genetic mechanism of sugar content in sorghum and the QTL detected in this study might be useful in the improvement of sugar content by marker-assisted selection.  相似文献

A high-pressure phase of TiO(2), which had been observed by shock-wave experiments and remained unresolved, has been studied by in situ x-ray diffraction. The single phase was formed at 20 gigapascals and 770 degrees C with the use of sintered-diamond multianvils; it has the same structure as baddeleyite, the stable phase of ZrO(2) at ambient conditions. The coordination number of Ti increases from six to seven across the rutile to baddeleyite transition, and the volume is reduced by approximately 9 percent.  相似文献

Reproductive performance and factors that decrease the pregnancy rate in heavy draft horses bred at the foal heat were investigated. The study was based on a total of 422 mares varying from 4 to 20 years in age, which had foaled during the period 1991 to 1994. Over the four years, the percentage of the mares bred at the foal heat was 62.6% and the pregnancy rate was 60.2%. The pregnancy rate following breeding at the foal heat in 1991 (67.7%) was significantly higher than that in 1992 (44.1%). Significant correlation was observed between the pregnancy rate and the mean placental retention time and also between the pregnancy rate and the mean temperature during the winter season. When compared on a monthly basis, the pregnancy rate following breeding at the foal heat in February was 53.3% and in May, 48.2%. The mean number of covers per mare at the foal heat was 2.3; this figure decreased to 1.8 for those bred in May. The mean number of days from parturition to the first breeding date was 11.3 days and the number of days from parturition to the last breeding day at the foal heat was 14.0 days. The number of days from parturition to the last breeding day at the foal heat decreased to 12.5 days in May. The latter may have been influenced by factors involving the stallion. When compared on a monthly basis, the mean placental retention time was longest in January (219 min.) following which it decreased gradually up to mid-summer. The mean placental retention time of mares which were not bred at the foal heat was significantly longer (197 min.) than in mares bred at the foaling heat (143 min.) and also in mares that conceived following breeding at the foal heat (135 min.). The pregnancy rate was less in mares bred shortly after parturition. The findings of this study would suggest that certain factors decrease the pregnancy rate in mares bred at the foal heat. The stress of low temperature in winter, prolonged placental retention time, breeding too early after parturition and factors pertaining to individual stallions may be involved. Expulsion of the placenta as soon as possible after foaling and appropriate breeding timing are significant factors that can influence the pregnancy rate following breeding at the foal heat.  相似文献

ABSTRACT:   Feeding activity was examined at high temperatures by using a demand feeder for thermally selected and normal rainbow trout Oncorhynchus mykiss strains. When the water temperature was raised in experiment 1 from 17.5 to 25.7°C at 0.3°C/day in 21 days, the average daily food consumption rate in the thermally selected strain rose to 7.1%, which was significantly higher than that of the normal strain (4.1%, P  < 0.05). The corresponding rate was also significantly higher in the thermally selected (0.8%) than in the normal strain (0.2%, P  < 0.05) of fish in experiment 2 where water temperature was raised to 24.1°C in 0.5°C/day increments. When water temperature was raised rapidly in experiment 3 from 16.7 to 21.7°C in one day and gradually to 24.4°C in 28 days at 0.1°C/day, the average daily food consumption rates were 1.0 and 0.1% for the thermally selected and normal strains, respectively, with significant difference ( P  < 0.01). These results suggest that the thermally selected strain has acquired thermal tolerance as a result of artificial selection.  相似文献

Tula virus (TULV) and Puumala virus (PUUV) are hantaviruses carried by the bank vole (Myodes glareolus) and European common vole (Microtus arvalis), respectively. PUUV is a causative agent of hemorrhagic fever with renal syndrome (HFRS), while TULV is thought to be apathogenic to humans. The N-terminal regions of the N proteins from TULV and PUUV were expressed and applied as enzyme-linked immunosorbent assay (ELISA) antigens. Colonized Japanese grass voles (Microtus montebelli) and BALB/c mice were used for experimental inoculation of the vole-borne hantaviruses TULV and PUUV. Voles and mice showed significant antibody production toward both viruses, but these antisera showed little cross-reactivity between TULV and PUUV in the immunofluorescence antibody assay and ELISA. In contrast, sera from patients with HFRS caused by PUUV exhibited high cross-reactivity against the TULV antigen, and sera from a natural rodent reservoir showed moderate cross-reactivity against the heterologous antigen, indicating that the antigenic cross-reactivity between TULV and PUUV differs in sera from rodents and humans.  相似文献

The objective of the present research was to assess the functional properties of the starches isolated from Australian prime hard wheat (PH) and hard red winter wheat (HRW) in the United States. These wheats are favorably used in Japan for the production of Chinese yellow-alkaline noodles (YAN). Starches isolated from PH showed higher amylose content than those of noodle wheat but lower than bread wheat. Analysis of swelling power showed that PH starch had lower values than noodle wheat starches but higher values than bread wheat starches. By contrast, HRW starch showed relatively higher amylose content and lower swelling power. There were large differences in the starch pasting properties between PH and HRW. High-performance size-exclusion chromatography studies on the components leached from the starch (80°C, 20 min) showed that PH starch had a somewhat characteristic leaching profile. Its peak ratio of high molecular weight region to low molecular weight region was greater than that of HRW starch. PH starch also characterized by having lower endothermal enthalpy (ΔH) in amylose-lipid transition. The eating quality assessment for YAN revealed that PH had more elastic texture than HRW. Additional reconstitution studies showed that these quality differences could be attributable to its starch properties.  相似文献
