Abstract  – A 2-year study of the threatened Australian grayling, Prototroctes mareana, was undertaken to investigate its reproductive strategy in relation to river flow. Monitoring of reproductive maturity in the Barwon River in south-eastern Australia indicated that no spawning occurred in the 2 years. In both years, ovarian involution was observed in a large proportion of the sampled population; flows during this period were less than the historical average. When coupled with the historical evidence, these observations suggest that spawning coincides with increased river discharge. River regulation may inhibit spawning.  相似文献

Hydroelectric development and the disruption of migration in caribou   总被引:1,自引:0,他引:1  
We investigated the effects of hydroelectric development on the movements and space-use of caribou (Rangifer tarandus caribou) in west-central Newfoundland, Canada. We compared patterns of range use, site fidelity, and timing of migration before, during, and after project construction. Coincidental with the first year of project construction, caribou were less likely to be found within 3 km of the site; this persisted at least 2 years after construction was completed. Relative timing of migration was individual-specific; the rank order of spring arrival on, and autumn departure from, the calving and summer grounds tended to be consistent year-to-year. This is the first report of such individual-specific consistency in migration for a non-avian species. This predictability disappeared during development: the year-to-year consistency of fall and spring migration among individuals was apparent before and after construction, but not during construction. Variation in calving site fidelity was correlated to year-to-year differences in snowfall. We conclude that the development caused a disruption of migrational timing during construction and longer-term diminished use of the range surrounding the project site. Long-term studies of individually marked animals can aid in environmental assessments for migratory animals.  相似文献

The yeast Saccharomyces cerevisiae was used in a high-throughput bioassay to identify phenolic agents for control of the aflatoxigenic fungus Aspergillus flavus. Veratraldehyde, 1, cinnamic acid, 5, and the respective benzoic acid derivatives vanillin, 2, vanillic acid, 3, and vanillylacetone, 4, and cinnamic acid derivatives o-coumaric acid, 6, m-coumaric acid, 7, and p-coumaric acid, 8, showed significant antifungal activities (from highest to lowest, 2, 5 > 1 > 6, 7 > 4 > 3, 8) in the yeast system, with caffeic acid, 9, having little to no effect. Antifungal activity levels against A. flavus were similar. This similarity in antifungal activity demonstrated the usefulness of the S. cerevisiae bioassay for screening antifungal compounds. Assays using deletion mutants of yeast identified signal transduction and antioxidative stress response genes important to fungal tolerance. Targeting the antioxidative stress response system with certain compounds (e.g., 4) in combination with strobilurin fungicides had a synergistic effect against both fungi.  相似文献

Dying-arm disease in grapevines, produced by infection with the ascomycete Eutypa lata, is responsible for major production losses in vineyards. Dieback of the shoots and cordon is believed to be due to acetylenic phenol metabolites produced by the fungus. To identify specific metabolites that could potentially be used for diagnosis of infection, eight E. lata isolates were grown in vitro on hot water extracts from grape varieties with various degrees of tolerance to the foliar symptoms of E. lata dieback. HPLC analysis showed that eutypinol was consistently produced in large amounts, together with smaller amounts of methyleutypinol and eulatachromene; eutypine, the putative toxin, was produced solely on Sauvignon Blanc extract and then in only barely detectable amounts. When E. lata isolates from Cabernet Sauvignon and Merlot were grown on identical media, the amounts of metabolites produced differed significantly between isolates but the pattern of metabolites was quite similar, with eutypinol again predominating. The consistent production of eutypinol indicated that this was the most suitable metabolite for which to analyze in order to diagnose the presence of E. lata. Extraction and analysis of grapevine tissues exhibiting symptoms of dieback failed to show the presence of any metabolites. However, when infected cordon sections were placed in water and cultured for 5 days, eutypinol was readily detected in the aqueous solution; metabolites were not produced from uninfected tissue. This provides a method for detection of infected tissue and indicates that the toxic metabolites react at the point of production, disrupting the vascular structure and inhibiting transport of nutrients, rather than being translocated to tissues that exhibit symptoms.  相似文献

The current study examined the protective effects of l ‐glutamine and cytochalasin B during vitrification of immature bovine oocytes. Oocyte vitrification solution (PBS supplemented with 10% FCS, 25% EG, 25% DMSO and 0.5 m trehalose) was the vitrification control. Treatments were the addition of 7 μg/ml cytochalasin B, 80 mm glutamine or both cytochalasin and glutaminine for 30 s. After warming, oocytes were matured in vitro for 24 h, fixed and stained with Hoechst (33342) for nuclear maturation evaluation. l ‐glutamine improved the vitrified/warmed immature bovine oocytes viability (32.8%), increasing the nuclear maturation rates compared to other treatments and the no treatment vitrified control (17.4%). There was, however, no effect of cytochalasin B on in vitro maturation (14.4%).  相似文献

Aflatoxigenic aspergilli inflict major economic damage to the tree nut industry of California, with the highest negative impact to almonds. Aspergilli and fungi in general are known to emit volatiles in varying quantity and composition dependent upon their growth media. The goal of the study was to determine the volatile emission of whole and blanched almonds that had been picked out and labeled as inedible by processors. The aflatoxin content and number of colony forming units of each sample were also determined. A total of 23 compounds were consistently detected and identified. Several volatiles from the blanched almonds demonstrated significant increases when compared to the emissions of whole almonds. Several of these volatiles are considered fatty acid decomposition products and included hexanal, heptanal, octanal, nonanal, 3-octen-2-one, tetramethylpyrazine, and decanal. The almond samples investigated were characteristic of a typical postharvest environment and illustrative of potential contamination within a stockpile or transport container. Volatiles indicative of fatty acid decomposition were predominant in the samples that underwent some form of blanching. The emission amounts of hexanal, heptanal, octanal, and hexanoic acid increased 3-fold in samples contaminated with aflatoxin; however, due to variability between samples they could not be considered as indicator volatiles for aflatoxin content. The emission profile of volatiles from almond kernels contaminated with naturally occurring aspergilli and associated fungi is heretofore unreported.  相似文献
