The innate immune system constitutes an efficient defense mechanism against invading microbial pathogens. Recent studies have revealed the intracellular signaling cascades involved in the TLR-initiated immune response to Brucella spp. infection. However, there is a piece of the puzzle missing that is the role of non-TLR receptors in innate immunity. The involvement of TLR receptors in brucellosis has been investigated by different research groups. It was demonstrated that TLR2 clearly does not play any role in controlling Brucella abortus infection in vivo, whereas TLR9 has been shown to be required for clearance of this bacterium in infected mice. The participation of adaptor molecules, such as MyD88 and TRIF has also been discussed. Recently, we and others have reported the critical role of MyD88- and not TRIF-mediated signaling in dendritic cell maturation and in vivo resistance during B. abortus infection. However, the relationship between specific Brucella molecules and non-TLR receptors and signal transduction pathways needs to be better understood. It is now clear that the interaction between TLRs and recently identified cytosolic innate immune sensors is crucial for mounting effective immune responses. Trypanosoma evansi is the most widely spread of the pathogenic African trypanosomes of animals. The disease (surra) was first diagnosed in the Canary Islands in a dromedary camel in 1997; thus, a control plan was implemented achieving the eventual eradication of T. evansi from most of the infected areas in the Archipelago. However, a little area remains still infected despite the use of the same control measures. To evaluate possible reservoirs in the area a representative sample of domestic ruminants was examined by serological, parasitological and molecular tests. Of a total of 1228 ruminants assessed, 61 (5%) were serologically positive (7 cattle, 21 goats, 33 sheep), but T. evansi could be demonstrated in none of them. According to FreeCalc assessment, cattle and goat populations would be free from disease; however, the results from sheep are not adequate to conclude that the population would be free from disease. As a conclusion, surveillance must be exercised on ruminant farms in the surroundings of the infected area in order to evaluate the possible extension of the disease and their potential role as reservoirs of T. evansi. 相似文献

The aim of this study was to establish the relationship between the stiffness of scantlings, approaching structural size materials, and small specimens of 6-year-old Eucalyptus urophylla?×?grandis wood using a resonance technique. The correlation between the elastic modulus (E) of the scantlings in longitudinal vibration and small specimens in flexural vibration was r?=?75 when comparing the scantling values with the averaged values of the specimens per scantling. However, when the E of each single specimen was compared with its respective scantling, the coefficient of correlation decreased to r?=?0.64 in the longitudinal tests and r?=?0.61 in the flexural tests. A roughly linear correlation (r?=?0.59) between specific modulus and loss tangent was obtained for the small specimens of Eucalyptus. In short, the resonance technique rapidly provided a large, accurate data set of mechanical wood properties as required for high-throughput phenotyping in recent genetic studies. 相似文献

The changes in the soil nitrate concentration were studied during 2 years in a “montado” ecosystem, in the South of Portugal.
Total rainfall, air and soil temperature and soil water content under and outside Quercus rotundifolia canopy were also evaluated. A cluster analysis was carried out using climatic and microclimatic parameters in order to maximize
the intraclass similarity and minimize the interclass similarity. It was used the k-Means Clustering Method. Several cluster
models were developed using k values ranging between 2 and 5. Thereafter, in each cluster, the data were divided according
to their origin (soil under canopy and open areas, and from surface and deep layers). Multiple regression models were tested
for each cluster, to assess the relationship between soil nitrate concentration and a set of climatic and microclimatic parameters
and the results were compared with models assessed without clustering. The models achieved with data grouped in result of
clustering analysis showed better performance than the models achieved without clustering, mostly for data from open areas
soils. When temperature is low and/or water presents excess or scarcity levels, the data from soils in undercanopy areas,
give rise to models with worst performance than models from open soil areas data. The results obtained for undercanopy area
suggest that nitrification process in soil under Quercus rotundifolia trees influence is more complex than for open areas, and subject to other relevant factors beyond water and temperature. 相似文献

Agroforestry systems (AFS) derived from the cactus forest “chichipera” and cultivated with “milpa”, the traditional multi-crop
system of maize-beans-squash were studied in the Tehuacán Valley, Mexico. Plant management types practiced by people, role
of agricultural and forest resources in households’ subsistence, and consequences of changes in AFS were investigated through
ethnobotanical, ecological, and economic approaches. People maintain in this AFS 122 plant species that provide useful products
for 14 different purposes. Nearly 90% of useful plants are native to the region, representing 54% of all plant species recorded
in the chichipera forest. Management strategies were identified occurring on 63 species at landscape, parcel, and individual
plant levels. About 67% of managed plant species are left standing during clearance of vegetation; 35% are transplanted to
the parcels’ surrounding area, 32% receive particular protection, and 10 species from other areas are cultivated inside the
fields. During years of higher rainfall (~745 mm) AFS supply nearly 75% of the maize grain and 50% of the maize straw used
as fodder by households. AFS with intermediate vegetation cover have higher economic value than monocultures and forest. However,
plots with low forest cover and monocultures are progressively more common as consequence of socio-cultural processes. Local
traditional ecological knowledge and management techniques are crucial for managing resilience of these AFS. 相似文献

Toxoplasma gondii infections are prevalent in many avian species and can cause mortality in some bird hosts. Although T. gondii has been isolated from various species of birds, the role of many different species of wild birds in the epidemiology of T. gondii remains unknown. Neospora caninum, a closely related parasite to T. gondii, has been recently confirmed to infect domestic chickens and wild birds such as house sparrows (Passer domesticus). The present study reports the presence of T. gondii and N. caninum DNA by PCR in brain tissues of 14 species of wild birds from Spain. From a total of 200 samples analyzed, 12 samples (6%) were positive for T. gondii [5 Eurasian jays (Garrulus glandarius), 5 magpies (Pica pica), 1 black kite (Milvus migrans) and 1 Griffon vulture (Gyps fulvus)], while 3 samples (1.5%) were positive for N. caninum [2 magpies and 1 common buzzard (Buteo buteo)]. This is the first report of detection of T. gondii in magpies, griffon vulture and black kite and of N. caninum in common buzzard and magpies, extending the list of natural intermediate hosts for T. gondii and N. caninum infections to these species. 相似文献

Antigen-specific antibody responses, T cell subsets and cytokine profiles were studied in 7 heifers naturally infested by Hypoderma sp. in Northwestern Spain. Immunoglobulin G (IgG) levels increased significantly at the end of the endogenous life cycle of the parasite (Mr). Similarly, IgG1 subclass increased considerably when first instars (L1) started their migration towards the back (Nv-Ja), whereas IgG2 increased earlier, coinciding with the arrival of L1 to the resting sites (Jn-Jl). Both subclasses decreased significantly when L3 began to leave the host. IgM levels and CD4 and CD8 profiles hardly oscillated throughout the life cycle of the parasite into the host. The CD4/CD8 ratio showed helper T cell predominance. Serum interferon-γ (IFN-γ) concentrations decreased from October to the end of the study. Interleukin 4 (IL-4) concentrations decreased in January and increased in February and May. There were a significant positive relationship between IL-4 and IgG2 subclass and a negative correlation between IFN-γ, IgG and IgG1 and also between IgM and CD2 and CD8 counts. These results suggest that in the early phases of natural primoinfestations by Hypoderma there is a slight predominance of a Th1 response, characterized by high IgG2 and IFN-γ levels, which is followed by a Th2 response with a clear predominance of IgG1 and IL-4. 相似文献

Two separate controlled and blinded studies were conducted to confirm the dose and non-interference of spinosad and milbemycin oxime (MO) administered orally in combination or alone to dogs for the treatment and control of experimentally induced flea infestations (Ctenocephalides felis) and adult hookworm infections (Ancylostoma caninum). For each study, dogs were allocated randomly based on pre-treatment adult flea and hookworm egg counts to one of four treatment groups of 10 animals each. In each study, spinosad and MO in combination, using the lower half (30-45 mg/kg spinosad; 0.5-0.75 mg/kg MO) of the US commercial dose band (30-60 mg/kg spinosad; 0.5-1.0mg/kg MO) of each active ingredient, or individually alone using the full dose range, were given orally to dogs on Day 0 using a tablet formulation. A placebo control was treated similarly. In one study, on Days -1, 5, 12, 19, 28 and 35 each dog was infested with approximately 100 unfed adult C. felis obtained from the investigator's established flea colony. All dogs were infested via the same method. Forty-eight hour post-infestation flea comb counts were conducted on Days 1, 7, 14, 21, 30 and 37 and were used to determine the knockdown and residual flea activity. In the second study, on Day -27 each of 48 dogs were experimentally inoculated with 100 third-stage infective larvae of the hookworm, A. caninum. Dogs were treated on Day 0 and necropsied on Day 7 or Day 8. All nematodes in the intestinal tract were collected on Day 7 or Day 8, identified and counted by species and stage. Post-treatment, the geometric mean live flea counts were significantly different (p-value<0.0001) between the spinosad/MO combination and the spinosad only treatment groups as compared to the vehicle control group. The flea counts in the MO only group and the control group were not statistically different. The spinosad and MO combination group and the spinosad only treatment group demonstrated significantly different knockdown (100%) and post-treatment residual flea efficacy at Day 30 was 100% for both groups as compared to the vehicle control. The presence of MO in combination with spinosad did not interfere with the flea efficacy of spinosad as compared to the spinosad only group. MO alone did not demonstrate any flea efficacy. Post-treatment, the geometric mean A. caninum worm counts were significantly different (p-value<0.0001) between the spinosad and MO combination group as compared to the vehicle control group. The worm counts in the MO only group and the combination group were not statistically different. The spinosad and MO combination group (99.8% reduction) and the MO only treatment group (99.5% reduction) both demonstrated significantly different hookworm efficacy as compared to the vehicle control group. The presence of spinosad in combination with MO did not interfere with the hookworm efficacy of MO as compared to the MO only group. Spinosad alone did not demonstrate any hookworm efficacy. In summary, flavored spinosad and MO combination tablets administered orally to dogs at the lower end (30-45 mg/kg spinosad; 0.5-0.75 mg/kg MO) of the US commercial tablet unit dose range (30-60 mg/kg spinosad; 0.5-1.0mg/kg MO) were both safe and highly efficacious delivering 100% knockdown and 30 days of residual adult flea control on experimentally infested dogs as well as >99% adult hookworm efficacy evaluated under laboratory conditions.
