The S-genotypes of 16 apricot (Prunus armeniaca L.) cultivars native to China were determined by the S-allele PCR approach and the results were confirmed by cross-pollination tests among these cultivars. Primer combination EM-PC2consFD + EM-PC3consR,
based on the conserved regions C2 and C3 of RosaceousS-RNase genes, was the most useful primer combination for identifying Chinese apricot S-alleles. Twelve S-RNase alleles were identified using this primer combination, and they were defined as follows: S9 was 657 bp, S10 was 266 bp, S11 was 464 bp, S12 was 360 bp, S13 was 401 bp, S14 was 492 bp, S15 was 469 bp, S16 was 481 bp, S17 was 487 bp, S18 was 1337 bp, S19 was 546 bp and S20 was 1934 bp. S11–S20 were new S-RNase genes deposited in GenBank under accession numbers DQ868316, DQ870628-DQ870634, EF133689 and EF160078, respectively.
Our findings contribute to a more efficient breeding program of Chinese apricot and further studies on the S-RNase genes. 相似文献
The presence of high levels of sinigrin in the seeds represents a serious constraint for the commercial utilisation of Ethiopian
mustard (Brassica carinata A. Braun) meal. The objective of this research was the introgression of genes for low glucosinolate content from B. juncea into B. carinata. BC1F1 seed from crosses between double zero B. juncea line Heera and B. carinata line N2-142 was produced. Simultaneous selection for B. carinata phenotype and low glucosinolate content was conducted from BC1F2 to BC1F4 plant generations. Forty-three BC1F4 derived lines were selected and subject to a detailed phenotypic and molecular evaluation to identify lines with low glucosinolate
content and genetic proximity to B. carinata. Sixteen phenotypic traits and 80 SSR markers were used. Eight BC1F4 derived lines were very close to N2-142 both at the phenotypic and molecular level. Three of them, with average glucosinolate
contents from 52 to 61 micromoles g−1, compared to 35 micromoles g−1 for Heera and 86 micromoles g−1 for N2-142, were selected and evaluated in two additional environments, resulting in average glucosinolate contents from
43 to 56 micromoles g−1, compared to 29 micromoles g−1 for Heera and 84 micromoles g−1 for N2-142. The best line (BCH-1773), with a glucosinolate profile made up of sinigrin (>95%) and a chromosome number of
2n = 34, was further evaluated in two environments (field and pots in open-air conditions). Average glucosinolate contents over
the four environments included in this research were 42, 31 and 74 micromoles g−1 for BCH-1773, Heera and N2-142, respectively. These are the lowest stable levels of glucosinolates reported so far in B. carinata. 相似文献
Antiproliferative effect of Amaranthus mantegazzianus proteins and peptides released after simulated gastrointestinal digestion (DH% 37.8?±?3.8) was investigated on human colon cancer cell line HT-29. Inhibition of proliferation of HT-29 cells was exhibited after a 24 h treatment with different concentrations of amaranth protein isolate (API) and the peptides released after digestion (DGS), presenting IC50 values of 1.35?±?0.12 and 0.30?±?0.07 mg soluble protein/mL, respectively. Lactate dehydrogenase assay indicated that both samples caused the loss of membrane integrity and cell lysis over HT-29 cells, and DAPI fluorescence microscopies evidenced typical apoptotic features. Moreover, Annexin V-FITC flow cytometry showed a significant increase of early apoptotic and late apoptotic/necrotic HT-29 cells compared to untreated ones, and caspase-3 assay confirmed the apoptosis induction with a 43.0?±?10.3 and 65.8?±?12.7% increase of caspase-3 activity produced by a 2 mg/mL treatment of API and DGS, respectively. In conclusion, amaranth peptides successfully released after simulated gastrointestinal digestion would exert a potential antiproliferative activity over HT-29 tumor cells. This effect was linked to the induction of cell necrosis and apoptosis, supporting the idea of using amaranth proteins as a potential food alternative ingredient for functional foods.
Mexican landrace avocados are naturally distributed mainly in high areas of central Mexico, where they have been produced and consumed since pre-Hispanic times. However, trees of these species are being replaced by improved varieties with greater global demand, and many species have been lost due to the destruction of their natural habitats. Many people in Mexico like to consume the pulp and peel of these fruits and have done so since pre-Hispanic times. This is because the peel of Mexican landrace avocados, unlike the peel of Hass avocados, is very thin and flavorful. The peel color may be bluish-purple or dark reddish due to the presence of anthocyanins, which are compounds with antioxidant activity. The objective of this study was to assess the oil and anthocyanin contents and to evaluate the antioxidant activity in fruits of 11 accessions collected from producing-regions of Mexico. The oil content was 16.2 to 32.3 g 100 g?1 in pulp, and the main unsaturated fatty acids were oleic, linoleic and palmitoleic acids, depending on the accession. The anthocyanin contents in peels ranged from 0.64 to 47 mg g?1 fresh weight. The highest antioxidant activity was found in the peel (53.3–307.3 mmol g?1 fresh weight). The results confirm that the pulp and peel of dark-peel Mexican landrace avocados could be important nutraceuticals for humans.
Using different genetic lines of pepper, this study analyses the effect of boron (B) on pollen quality and its capacity to pollinate and fertilize. Three different B concentrations (100, 150, and 200 mg L?1) were applied to six genetic lines, evaluating the in vitro pollen quality and pollen formation. In a second experiment, the highest and the lowest B concentrations were applied to two low-pollen genetic lines, evaluating the growth of the pollen tubes in flowers pollinated with treated pollen. The higher B concentrations significantly improved pollen quality and formation in vitro, while improved pollination and fecundity was observed after monitoring the pollen tubes. For all variables, the specific genetic line also showed some effect. This technique represents a tool for improving pollen in manual hybrid seed programs. 相似文献
Persistent organic pollutants (POPs) are globally dispersed substances considered to have adverse effects on health and ecosystems. Organochlorine pesticides (OCPs) are POPs. OCPs can be collected from the air by passive samplers using polyurethane foam (PUF). POPs in PUF are generally extracted by Soxhlet, considered as reference extraction method. We offer a different methodology approach to extract 16 OCPs from the PUF. This technology extracts, filters, collects the sample and evaporates the solvent, without sample transfer. It is a modification of the ultrasound-assisted extraction micro-scale cell (M-UAE-MSC) previously patented by our group. A full factorial experimental design (23 centred) was used to optimise the extraction conditions. The most desirable conditions were 50 °C, 40% ultrasound potency and 20-min extraction time, two extractions. OCP recoveries ranged from 67 to 110%. OCP recoveries were equivalent by M-UAE-MSC and Soxhlet techniques. The method was not suitable for five OCPs, and further refinement is needed. Method linearity (r2) was ≥?0.98. Limits of detection were observed between 1.2 and 4.6 ng PUF?1, while limits of quantification were found between 3.9 and 15.2 ng PUF?1. The method was applied to determine OCPs collected by PUF passive samplers in Mexico. Endosulfan I was the OCP most frequently observed. The M-UAE-MSC optimised extraction conditions, decreased the extraction time from several hours to less than 1 h and reduced three times the solvent consumption with respect to Soxhlet. This afforded a decrease in the waste generation and a reduction in the OCP-extraction cost up to 80%. The results of this study reveal an efficient and consistent procedure, as well as a simpler, faster, cost-saving and more environmentally friendly method to determine OCPs collected by PUF in passive samplers compared to Soxhlet extraction. 相似文献
To evaluate the effects of pre- and post-conception undernutrition (UN) on fetal and placental development at mid-gestation, 28 Katahdin × Pelibuey multiparous ewes were blocked by weight and assigned to the following four dietary treatments (n = 7 each): ewes fed 100% (control) or 60% of their nutritional requirements 30 days before mating (UNPre), 50 days after mating (UNPost) or during both periods (UNB). Four twin-bearing ewes were selected per treatment at day 50 post-conception and then slaughtered at day 75 of gestation to analyze their fetuses. Control fetuses were heavier (P < 0.05) than UNPost and UNB fetuses in 14.6 and 9.4%, respectively. Organ weights as percentage of the fetal weight (except for liver) and morphometric measurements (except for abdominal girth) were similar between control and UN fetuses (UNPre, UNPost, and UNB). Placental mass was heavier (P < 0.05) in control ewes than UNB ewes, but not relative to ewes of other treatments. The number of placentomes per ewe and placental efficiency were unaffected by UN treatments. Compared to control, only UNB ewes exhibited variations (P < 0.05) in the proportion of placentomes, specifically for type A (+13.8%) and B (?12.6%). Placentomes of type A and B had lower weight, length, and width of placentas in UNPost and UNB ewes than placentas of control ewes (P < 0.05). Overall results indicate that fetal and placental development of ewes carrying twins is mainly altered when nutritional restriction occurs simultaneously before conception and during the first third of pregnancy. 相似文献
Transversus abdominis plane (TAP) is a fascial plane containing the thoracolumbar nerve branches that innervate the abdominal wall. Limited information is available on the anatomical organization of these nerve branches in the dog, which is of great importance for the success of the TAP block anaesthetic technique. The aim of this study was to describe the origin and conformation of thoracolumbar nerves running through the TAP in 20 hemi‐abdominal walls of 10 adult mongrel dog cadavers with an average body weight of 12.6 kg (range: 9.6‐15.6). The abdominal walls were dissected from superficial to deep direction, the skin and both obliquus externus abdominis and obliquus internus abdominis muscles were dissected and reflected dorsally to expose the transversus abdominis muscle and the thoracolumbar nerve branches located in this plane. The anatomical features of ventral nerve branches were described. The thoracic nerve branches: T7–T12 and costoabdominalis; and the lumbar nerve branches: iliohypogastricus cranialis, iliohypogastricus caudalis, ilioinguinalis and cutaneus femoris lateralis were identified in all the cadavers. Anatomical variations related to the presence or absence within the TAP of the T7, T8 and T9 nerve branches were found. These variations should be taken into account when planning the TAP block technique in dogs. 相似文献
The study was designed to determine the relation between β‐hydroxybutyrate (BHB) concentrations in milk from dairy cows after calving and the length of the previous lactation, the dry period and the 305‐day normalized production, and to assess the influence of BHB concentrations on culling and test‐day milk productions and somatic cell counts (SCC) throughout the lactation that followed the BHB measurement. The data used in the study were obtained from 59 187 cows in the Galicia region (Spain). BHB determination was performed using Fourier‐transformed infrared spectrometry from the milk samples collected from each cow on the first post‐partum test day. For statistical analysis, the following methods were applied: (i) ordinal regression to assess the effect of the length of the previous lactation, the dry period and the 305‐day normalized milk production on milk BHB, (ii) a Cox model to estimate the influence of the BHB concentration on risk of culling (overall and for a variety of reasons) and (iii) linear regression to assess the link between BHB and the milk yield and SCC obtained from each of the tests day performed throughout lactation. The probability of having higher BHB concentrations increased when the length of the previous lactation (p = 0.006), the dry period (p = 0.003) and the 305‐day normalized milk yield (p = 0.005) increased. However, the slight increase observed (especially for the case of the dry period and the 305‐day milk yield) would not justify that measures be implemented to reduce these traits. Higher concentrations of BHB led to an increased risk of culling due to ‘death’ (p ≤ 0.001) and ‘urgent slaughter’ (p ≤ 0.002) (both causes of involuntary culling). It also led to a reduction in milk production (p < 0.001) and an increase in SCC (p < 0.001) in the post‐partum; from that moment onward (including peak lactation), there were no differences in those two parameters depending on the BHB levels. 相似文献
