Toxoplasma gondii, the agent of Toxoplasmosis, is an obligate intracellular protozoan able to infect a wide range of vertebrate cells, including nonprofessional and professional phagocytes. Therefore, drugs must have intracellular activities in order to control this parasite. The most common therapy for Toxoplasmosis is the combination of sulfadiazine and pyrimethamine. This treatment is associated with adverse reactions, thus, the development of new drugs is necessary. In previous studies, naphthoquinone derivatives showed anti-cancer activity functioning as agents capable of acting on groups of DNA, preventing cancer cells duplication. These derivatives also display anti-parasitic activity against Plasmodium falciparum and Leishmania amazonensis. The derivative pterocarpanquinone tested in this work resulted from the molecular hybridization between pterocarpans and naphtoquinone that presents anti-tumoral and anti-parasitic activities of lapachol. The aim of this work was to determine if this derivative is able to change T. gondii growth within LLC-MK2 cells. The drug did not arrest host cell growth, but was able to decrease the infection index of T. gondii with an IC(50) of 2.5 μM. Scanning and transmission electron microscopy analysis showed morphological changes of parasites including membrane damage. The parasite that survived tended to encyst as seen by Dolichos biflorus lectin staining and Bag-1 expression. These results suggest that pterocarpanquinones are drugs potentially important for the killing and encystment of T. gondii. 相似文献

Zebu breeds play an important role in cattle production systems in Brazil. To assess the genetic variability from animals in the Herd Books of Nelore, Gir and Guzerat breeds, generation intervals, inbreeding, effective population size and parameters of gene origin (effective number of founders, ancestors and founder genomes) were calculated using pedigree records from 1938 to 1998. Breed subdivision was quantified by Wright's F -statistics. Calculations were separately carried out for consecutive 4-year intervals in the period 1979–98. Generation interval was around 8 years for the three breeds. Total inbreeding increased in all the breeds reaching values of 2.13%, 2.28% and 1.75%. Effective population size decreased from 85 to 68 in Nelore, from 70 to 45 in Gir and remained nearly constant around 104 in Guzerat. The quantities assessing the number of contributing ancestors decreased with time in all the breeds, and in the last analysed period the most important ancestor accounted for 14%, 3.1% and 4.1% in Nelore, Gir and Guzerat, respectively. Results indicate that the studied breeds are suffering from a loss of genetic variability which can result in negative effects on breeding and conservation purposes. 相似文献

The purpose of this study was to determine the clinical and laboratory profiles of canine leishmaniasis in two distinct areas. Dogs from urban and rural areas were examined. The population studied in the metropolitan area included 54 dogs. Of these, 20 (37%) animals did not present with any signs suggestive of visceral leishmaniasis (VL). Among these, only eight were confirmed negative by ELISA (rK39 and CE) and 12 dogs, clinically negative for leishmaniasis, were seropositive by ELISA (rK39 and CE). Thinness, conjunctivitis and onychogryphosis were the most frequent clinical signs in the urban areas, followed by crusty lesions, alopecia, ulcerated lesions, hyperkeratosis and exfoliation. In the metropolitan area human VL cases occurred mainly in 1991, 1992, 1999 and 2000. In the rural areas the ELISA rK39 test detected a seroprevalence of 11.3% and ELISA CE (Leishmania crude extract) of 20.6%. Thirty-nine dogs were examined 6 months after the first visit. Serological exams using rK39 antigen showed seroconversion of only one dog, whereas Leishmania CE showed seroconversion of 13 (33.4%) dogs. In this rural environment 83.3% of the positive dogs were asymptomatic. Lutzomyia intermedia and Lu. longipalpis were the most predominant sandfly vector species. Amastigotes were identified in spleen and liver fragments of symptomatic necropsied animals. PCR amplification of DNA isolated from promastigote culture indicated that the species was Leishmania chagasi. This finding suggests that delayed diagnosis and euthanasia of potentially infectious animals may occur with an increased transmission risk to sandflies and subsequently to humans. 相似文献

The aim of this work was to identify the predominant yeast species present at different anatomical sites in healthy dogs and to determine their in vitro antimicrobial susceptibility using a broth microdilution assay. Samples were collected from the preputial, vaginal, oral and perianal mucosae and the isolates cultured were identified according to their morphological characteristics and biochemical profile. Malassezia pachydermatis was the most commonly isolated yeast, followed by Candida parapsilosis, Candida tropicalis, Candida albicans, Saccharomyces cerevisiae and Rhodotorula spp.Minimum inhibitory concentrations of the azole derivatives ketoconazole, itraconazole and fluconazole against Candida spp. were 0.03–16 μg/mL, 0.06 to >16 μg/mL and 0.5–64 μg/mL, respectively and Candida isolates were sensitive to caspofungin and amphotericin B. Although all isolates of M. pachydermatis were sensitive to itraconazole, fluconazole, ketoconazole and amphotericin B, they were found to be resistant to caspofungin. The study has highlighted that Candida spp., M. pachydermatis, S. cerevisiae and Rhodotorula spp. are part of the normal canine surface microbiota and some of these organisms exhibit in vitro resistance to commonly used antimicrobials. 相似文献

In this study, we captured 60 wild New World monkeys (Cebus spp.; Alouatta caraya) at the Paraná river basin, Paraná State, Brazil, and modified agglutination test (MAT) was performed to evaluate anti-Toxoplasma gondii antibodies. Prevalence was 30.2% (13/43) in Cebus spp. (capuchin monkeys) and 17.6% (3/17) for A. caraya (black and golden howler monkeys). MAT showed antibody titers of 16 (15/16) and 64 (1/16). Herein, we have observed an odds ratio (OR)=4.67 (1.060.05). The present work is the first report on serum occurrence of anti-T. gondii antibodies in wild capuchin monkeys and in wild black and golden howler monkeys. 相似文献

Serum samples from 77 animals belonging to 38 species and 19 families in Rio de Janeiro Zoo, Brazil were tested for antibodies against serovars of Leptospira interrogans by microscopic agglutination test. Antibodies were detected in serum samples of 37.7% of all animals belonging to 10 families. Seropositivity was more common in the Carnivora Canidae (7/9), Procyonidae (5/9) and the Edentata Myrmecophagidae (5/9). Icterohaemorrhagiae was the most common serogroup. Marine biofouling is a natural process that represents major economic, environmental, and health concerns. Some booster biocides have been used in biofouling control, however, they were found to accumulate in environmental compartments, showing negative effects on marine organisms. Therefore, it is urgent to develop new eco-friendly alternatives. Phenyl ketones, such as benzophenones and acetophenones, have been described as modulators of several biological activities, including antifouling activity (AF). In this work, acetophenones were combined with other chemical substrates through a 1,2,3-triazole ring, a strategy commonly used in Medicinal Chemistry. In our approach, a library of 14 new acetophenone–triazole hybrids was obtained through the copper(I)-catalyzed alkyne-azide cycloaddition “click” reaction. All of the synthesized compounds were evaluated against the settlement of a representative macrofouling species, Mytilus galloprovincialis, as well as on biofilm-forming marine microorganisms, including bacteria and fungi. The growth of the microalgae Navicula sp. was also evaluated after exposure to the most promising compounds. While compounds 6a, 7a, and 9a caused significant inhibition of the settlement of mussel larvae, compounds 3b, 4b, and 7b were able to inhibit Roseobacter litoralis bacterial biofilm growth. Interestingly, acetophenone 7a displayed activity against both mussel larvae and the microalgae Navicula sp., suggesting a complementary action of this compound against macro- and microfouling species. The most potent compounds (6a, 7a, and 9a) also showed to be less toxic to the non-target species Artemia salina than the biocide Econea®. Regarding both AF potency and ecotoxicity activity evaluation, acetophenones 7a and 9a were put forward in this work as promising eco-friendly AF agents. 相似文献

Gene-flow methodology was used to calculate the cumulative discounted expressions (CDE) for annual/lactation, replacement heifer, cull cow, birth, yearling, and slaughter traits in alternative cattle production systems. Generic equations were presented and parameters representing dairy-beef production systems in Ireland and Brazil were inputted. Cumulative discounted expressions using input parameters from a hypothetical purebred dairy production system with poor cow longevity were also calculated. Cumulative discounted expressions were calculated assuming either an initial purebred or crossbred mating within each production system. Absolute and relative differences in CDE existed among trait categories across the three alternative production systems investigated. For example, the CDE of beef-related traits accruing from an initial purebred mating ranged from 0.42 to 0.75 CDE of an annual/lactation trait across the three contrasting systems investigated which differed in various input parameters. Such variation may alter the relative emphasis of traits on overall profitability thereby contributing to genotype by environment interactions. The results of this study highlight the necessity to consider auxiliary traits in sire selection over and above those representing the principal intended use of the sire. This was particularly so for integrated dairy-beef cattle production systems. 相似文献

The catadromous life cycle of the European eel encompasses a spawning migration to the Sargasso Sea. Prior to their migration, eels develop a silver coloration being referred to as “silver eels.” Due to the dramatic European eel recruitment decline, it is crucial to quantify silver eel escapement to evaluate the success of measures taken under Eel Management Plans (Regulation EC (1100/2007)). Thus, the percentage of silver eels escaping from the River Mondego (Portugal) was estimated during two consecutive spawning seasons (2014–2016) and their migratory behaviour was studied. Thirty-six male silver eels were tagged with acoustic transmitters and their downstream migration was tracked using automatic receivers deployed at four locations along the river course, from the first impassable obstacle up to the river mouth (67 km). Among the tagged eels, 15 (42%) escaped to the sea. In both years, individuals displayed faster migration speed in the downstream reaches of the river. BRT (“Boosted Regression Trees”) models were used to explain downstream migration and escapement of silver eels. The analyses revealed that the downstream migration is triggered by several environmental variables with water conductivity as the most influential predictor. Escapement of eels from the estuary occurred a few hours after their arrival and was primarily explained by the offshore wind intensity, which promotes a current in the coast that may favour their transport to the open sea. These findings contribute to a better understanding of silver eel migration in southern latitudes and highlight and reinforce the need to monitor spawner escapement. 相似文献
