ABSTRACT Near-isogenic lines (NILs) with resistance for scald in seventh generation backcross with 'Ingrid' as recurrent parent (RP) were tested with seven differential isolates of Rhynchosporium secalis in Norway and Canada. NILs of 'Turk', 'Brier', 'CI 8162', 'La Mesita', 'Hispont', 'Atlas 46', 'Modoc', 'Hudson', 'Abyssinian', 'Steudelli', and 'CI 2222' also were evaluated for field reactions. The genetic characterization of the NILs (degree of isogeneity with Ingrid) and with each other was carried out. The molecular marker pattern shows that the backcrossing program has resulted in from 86.3 to 100% RP genome in the NILs, depending on the marker system. On an average, 96% RP genome was found in the NILs. There were certain consistent (pairwise) differences between the NILs and RP on chromosomes 3H and 7H. Both chromosomes are known to contain loci conferring resistance to R. secalis, indicating successful introgression from the donors into the NILs. Approximately two-thirds of the observed RP-NIL polymorphisms were linked to the assumed resistance in the NIL. Based on the marker and phenotypic analyses of the NILs, suggestions for a more appropriate and updated terminology of genes for resistance to R. secalis in barley are made. The proposed changes in nomenclature also indicate the differentials that are available as NILs and those lacking.  相似文献

An 8‐year‐old, intact female, mixed‐breed dog presented to the Oklahoma State University Boren Veterinary Medical Teaching Hospital for evaluation of progressive lameness and joint effusion of multiple joints. Physical examination revealed joint effusion of the elbow, hock, and stifle joints bilaterally, enlarged left axillary and right popliteal lymph nodes, a subcutaneous mass over the left elbow, and a subcutaneous mass involving the left second and third mammary glands. Cytologic examination of the mammary mass, enlarged lymph nodes, and joint fluid from most affected joints revealed a monomorphic population of loosely cohesive neoplastic epithelial cells. The patient was humanely euthanized, and subsequent necropsy with histopathologic examination revealed a complex mammary carcinoma with metastases to enlarged lymph nodes, subcutaneous tissue over the left elbow, and the synovium of multiple joints. Immunohistochemical stains were performed and showed diffusely positive pan cytokeratin, CK8/18, and CK19 staining in the neoplastic luminal epithelial cells of the mammary carcinoma, synovium, and lymph nodes, and showed diffusely positive vimentin staining of the myoepithelial cells. Myoepithelial calponin positivity was diffuse in the mammary mass and lymph nodes but minimal in the synovium. Only the mammary mass showed p63 positivity. Metastatic mammary neoplasia is relatively common in dogs; however, metastasis to the synovium has only been reported once previously in the literature. This is the first case utilizing immunohistochemistry for confirmation and characterization of metastases.  相似文献

Zebrafish (also known as zebra danio) Danio rerio were injected intramuscularly with Edwardsiella ictaluri at doses of 6 x 10(3), 6 x 10(4), or 6 x 10(5) colony-forming units per gram (CFU/g) or sterile phosphate-buffered saline (sham) or were not injected. Mortality occurred from 2 to 5 d postinjection (dpi) at rates of 0, 76.6, and 81.3% for the low, medium, and high doses, respectively, and E. ictaluri was isolated from dead fish. Survivors were sampled at 10 dpi and E. ictaluri was not isolated. Sham-injected and noninjected controls did not suffer mortality. Histopathology trials were performed in which zebrafish were injected with 1 x 10(4) CFU/g or sham-injected and sampled at 12, 24, 48, 72, and 96 h postinjection for histological interpretation. Collectively, these zebrafish demonstrated increasing severity of splenic, hepatic, cardiac, and renal interstitial necrosis over time. To evaluate the progression of chronic infection, zebrafish were injected with 1 x 10(2) CFU/g and held for 1 month postinjection. Beginning at 12 dpi and continuing for an additional 2 weeks, zebrafish demonstrated abnormal spiraling and circling swimming behaviors. Histopathology demonstrated necrotizing encephalitis. In immersion trials, zebrafish were exposed to low, medium, and high doses (averaging 1.16 x 10(5), 1.16 x 10(6), and 1.16 x 10(7) CFU/mL of tank water) of E. ictaluri for 2 h. Mortality occurred from 5 to 9 d postexposure at rates of 0, 3.3, and 13.3% for the low, medium, and high doses, respectively; E. ictaluri was isolated from dead fish. Channel catfish Ictalurus punctatus exposed to the medium doses suffered 100% mortality, and E. ictaluri was isolated from these fish. This study demonstrates the potential use of zebrafish as a model for E. ictaluri pathogenesis.  相似文献

A family of the cross of the northern highbush blueberry ‘Draper’?×?the southern highbush ‘Jewel’ (D?×?J) was propagated and phenotyped for plant size, yield, rate of flower, bud, and fruit development, leafing ability, winter hardiness, and fruit quality for 2 years in the field in Florida (FL), Georgia (GA), Michigan (MI), and Oregon (OR). In addition, a controlled greenhouse study was conducted on these progenies to estimate their chilling requirements. Significant levels of genotypic variability were observed for all the traits and the majority showed significant location and genotype?×?environmental interactions. Most of these traits were normally distributed across genotypes. There were many more flower buds per cane in GA than FL and OR, and many more vegetative buds broke in OR than in the southeastern states. Most of the various yield and growth traits were significantly correlated, except for flower buds per cane with plant height and width, and strength of leafing with plant width and flower buds/cane. Plants in OR developed much more slowly than those in FL and GA. There was a significant G?×?E effect on days to early green tip, floral bud break, and full bloom. Significant correlations were observed between all the developmental rates except between floral bud break and the days to full flowering and first blue fruit. Most individuals were severely damaged by winter cold in MI, although a few showed good tolerance. Significant location effects were observed for pH, titratable acidity (TA), soluble solids (SS), and SS/TA. Fruit in FL were a little softer and had higher SS/TA ratios than those in GA. There were significant interactions between genotype and environment for all the fruit quality traits except for fruit scar, firmness, anthocyanin content (ACY), and % juice. Fruit scar had a significant positive relationship with firmness and a significant negative relationship with flavor. Fruit color was significantly associated positively with soluble solids and ACY. Firmer fruit had significantly lower levels of ACYs. Fruit pH showed significant positive correlations with TA, TA/SS, and % juice, and significant negative relationship with SS and ACY. TA was significantly associated negatively with TA/SS and positively with ACY and % juice. TA/SS and ACY were positively correlated significantly with % juice. Two or three individuals were identified at each location that had high yields, were very early or late ripening and had excellent overall fruit quality. There were significant levels of genotypic, environmental (chilling hours) and G?×?E interaction among vegetative and floral buds for numbers of buds per shoot, percentage that developed and their developmental rate. The chilling requirement of highbush blueberry appears to be regulated polygenically as the D?×?J family segregated normally. On average, genotypes did more poorly at 250 and 350 chilling hours than the other chilling treatments.  相似文献

The content of the biologically active amino acid theanine in 15 commercial black, green, specialty, and herbal tea leaves was determined as the 2,4-dinitrophenyltheanine derivative (DNP-theanine) by a validated HPLC method. To define relative anticarcinogenic potencies of tea compounds and teas, nine green tea catechins, three black tea theaflavins, and theanine as well as aqueous and 80% ethanol/water extracts of the same tea leaves were evaluated for their ability to induce cell death in human cancer and normal cells using a tetrazolium microculture (MTT) assay. Compared to untreated controls, most catechins, theaflavins, theanine, and all tea extracts reduced the numbers of the following human cancer cell lines: breast (MCF-7), colon (HT-29), hepatoma (liver) (HepG2), and prostate (PC-3) as well as normal human liver cells (Chang). The growth of normal human lung (HEL299) cells was not inhibited. The destruction of cancer cells was also observed visually by reverse phase microscopy. Statistical analysis of the data showed that (a) the anticarcinogenic effects of tea compounds and of tea leaf extracts varied widely and were concentration dependent over the ranges from 50 to 400 microg/mL of tea compound and from 50 to 400 microg/g of tea solids; (b) the different cancer cells varied in their susceptibilities to destruction; (c) 80% ethanol/water extracts with higher levels of flavonoids determined by HPLC were in most cases more active than the corresponding water extracts; and (d) flavonoid levels of the teas did not directly correlate with anticarcinogenic activities. The findings extend related observations on the anticarcinogenic potential of tea ingredients and suggest that consumers may benefit more by drinking both green and black teas.  相似文献
