This study aimed to verify whether transient inflammatory reactions incited by the administration of intra-articular platelet-rich plasma (PRP) affected joint components through short- and long-term in vivo evaluation of inflammatory biomarkers and extracellular matrix degradation products in synovial fluid. The effects of PRP were analyzed in a short phase protocol (SPP) and in a prolonged phase protocol (PPP), using saline-injected joints as controls. In the SPP, higher white blood cell counts and prostaglandin E2 and total protein concentrations were observed in the synovial fluid of PRP-treated joints (P < 0.05). There were no differences between the interleukin-1β, interleukin-1 receptor antagonist protein, tumor necrosis factor-α, chondroitin sulfate, or hyaluronic acid concentrations between PRP and saline injected joints. In the PPP, there were no differences in evaluated parameters between groups. PRP injection elicits a mild and self-limiting inflammatory response shortly after administration, without long-term deleterious effects on joint homeostasis.  相似文献

Dengue is considered a serious public health problem in many tropical regions of the world including Brazil. At the moment, there is no viable alternative to reduce dengue infections other than controlling the insect vector, Aedes aegypti Linnaeus. In the continuing search for new sources of chemicals targeted at vector control, natural products are a promising alternative to synthetic pesticides. In our work, we investigated the toxicity of a bioactive compound extracted from the red alga Laurencia dendroidea J. Agardh. The initial results demonstrated that crude extracts, at a concentration of 5 ppm, caused pronounced mortality of second instar A. aegypti larvae. Two molecules, identified as (−)-elatol and (+)-obtusol were subsequently isolated from crude extract and further evaluated. Assays with (−)-elatol showed moderate larvicidal activity, whereas (+)-obtusol presented higher toxic activity than (−)-elatol, with a LC50 value of 3.5 ppm. Histological analysis of the larvae exposed to (+)-obtusol revealed damage to the intestinal epithelium. Moreover, (+)-obtusol-treated larvae incubated with 2 µM CM-H2DCFDA showed the presence of reactive oxygen species, leading us to suggest that epithelial damage might be related to redox imbalance. These results demonstrate the potential of (+)-obtusol as a larvicide for use against A. aegypti and the possible mode of action of this compound.  相似文献

The aim of this study was to evaluate the effect of lyophilized bovine colostrum (LBC) used as partial source of dietary protein on the histological characteristics of the intestinal epithelium of juvenile dourado Salminus brasiliensis. Juveniles were fed diets containing 0%, 10% or 20% of LBC inclusion for either 30 or 60 days. For the histological study, the intestine was divided into three segments, S1, S2 and posterior intestine. In the S1 segment, interaction between treatment and period was observed in the number of goblet cells containing sialomucin, effect of treatment in the total number of goblet cells and effect of period in the number of goblet cells containing sulphomucins (P < 0.05). In the S2 segment, effect of period was observed in the number of goblet cells containing acid, neutral and total mucins, sialomucins and the partial volume of the absorptive mucosa (Vv) (P < 0.05). In the posterior intestine, effect of period was observed in the thickness of muscle layer and number of goblet cells containing sialomucins and sulphomucins (P < 0.05). Considering the aspects studied, the presence of 0%, 10% or 20% of LBC in the diet did not significantly influence the enteric histological characteristics of juvenile dourado during the period of the study.  相似文献

Fusarium oxysporum f. sp. phaseoli (Fop) is a devastating pathogen that can cause significant economic losses and can be introduced into fields through infested Phaseolus vulgaris (common bean) seeds. Efficient seed health testing methods can aid in preventing long‐distance dissemination of this pathogen by contaminated seeds. In order to improve detection of Fop in seed, a rapid, accurate and sensitive real‐time PCR assay (qPCR) protocol was developed for detection of Fop in common bean seeds. Seed lots of seven cultivars with infection incidence ranging from 0·25 to 20% were prepared by mixing known amounts of Fop‐infected seeds with Fop‐free seeds. Direct comparisons between SYBR Green and TaqMan qPCR methods were performed using primers based on the Fop virulence factor ftf1. The primers developed in this study produced a 63 bp product for highly virulent strains of Fop but did not produce an amplicon for nonpathogenic or weakly pathogenic isolates of F. oxysporum from P. vulgaris or other hosts. Under optimized conditions, both qPCR assays detected Fop infection at low levels (0·25%); however, the results suggest the TaqMan assay was more reliable at quantification than the SYBR Green assay. Linear regression models were fitted to the relationships between results of qPCR assays and infection incidence, but the models differed among cultivars. Fungal biomass per seed differed among cultivars and was related to seed size. The results indicate that the TaqMan assay developed in this study is a useful tool for the detection and quantification of Fop in bean seeds.  相似文献

Soil organic matter (SOM) and its different pools have key importance in nutrient availability, soil structure, in the flux of trace gases between land surface and the atmosphere, and thus improving soil health. This is particularly critical for tropical soils. The rates of accumulation and decomposition of carbon in SOM are influenced by several factors that are best embodied by simulation models. However, little is known about the performance of SOM simulation model in an acid tropical soil under different tillage systems including no-tillage (NT). Our objective was to simulate soil organic matter dynamics on an Acrisol under no-tillage and different plowed systems using Century model. Tillage systems consisted of no-tillage, disc plow, heavy disc harrow followed by disc plow, and heavy disc harrow. Soil C stocks simulated by Century model showed tendency to recovery only under no-tillage. Also, simulated amounts of C stocks of slow and active pools were more sensitive to management impacts than total organic C. The values estimated by Century of soil C stocks and organic carbon in the slow and passive pools fitted satisfactorily with the measured data. Thus fitted, except for the active pool, Century showed acceptable performance in the prediction of SOM dynamics in an acid tropical soil.  相似文献

In Brazil, no tillage (NT) is a soil conservation practice now widely adopted by farmers, including smallholders. The effect of NT and conventional tillage (disc ploughing followed by two light disc harrowings, CT) was investigated on the aggregation properties of a clayey Rhodic Ferralsol from southern Brazil under different crop rotations. The same soil type under secondary forest was used as reference. Macro- and microaggregate classes were separated by wet sieving using a series of eight sieves (8, 4, 2, 1, 0.5, 0.25, 0.125, 0.053 mm) at four sampling layers (0–5, 5–10, 10–20, 20–30 cm). The soil in general had high structural stability. At 0–5 cm, meanweight diameter (MWD, 11.1 mm) and total organic C in macroaggregates (TOC, 39 g kg−1 soil) were highest for the forest soil. Soil under NT had a more similar distribution of aggregate size classes and TOC to the forest soil than CT. The most pronounced difference between tillage systems was observed in the surface soil layer (0–5 cm). In this layer, NT had higher aggregate stability (ASNT: 96%; ASCT: 89%), had higher values of aggregate size distribution (MWDNT: 7.9 mm, MWDCT: 4.3 mm), and had on average 28% greater TOC in all aggregate size classes than CT. Soil under NT had greater TOC in macroaggregates (NT: 22 g kg−1; CT: 13 g kg−1). Crop rotation did not have a significant effect on soil aggregate distribution and TOC. By increasing macroaggregation NT increased organic carbon accumulation in soil.  相似文献

Microalgae have become a promising novel and sustainable feedstock for meeting the rising demand for food and feed. However, microalgae-based products are currently hindered by high production costs. One major reason for this is that commonly cultivated wildtype strains do not possess the robustness and productivity required for successful industrial production. Several strain improvement technologies have been developed towards creating more stress tolerant and productive strains. While classical methods of forward genetics have been extensively used to determine gene function of randomly generated mutants, reverse genetics has been explored to generate specific mutations and target phenotypes. Site-directed mutagenesis can be accomplished by employing different gene editing tools, which enable the generation of tailor-made genotypes. Nevertheless, strategies promoting the selection of randomly generated mutants avoid the introduction of foreign genetic material. In this paper, we review different microalgal strain improvement approaches and their applications, with a primary focus on random mutagenesis. Current challenges hampering strain improvement, selection, and commercialization will be discussed. The combination of these approaches with high-throughput technologies, such as fluorescence-activated cell sorting, as tools to select the most promising mutants, will also be discussed.  相似文献

Prototheca zopfii has been considered one of the most important causes of environmental mastitis in Brazil. These algae are refractory to conventional therapy and cause great damage to the mammary gland. The present study evaluated the in vitro algaecide effect of sodium hypochlorite and iodine based antiseptics on 27 P. zopfii strains isolated from the milk of cattle. Low concentrations of sodium hypochlorite (0.0390625-0.15625%) and iodine (0.15625-0.625%) were effective against the isolates. These antiseptics may be recommended for hygiene routines, pre and postdipping and cauterization of bovine mammary glands infected by P. zopfii.  相似文献

Brassica rapa can be infected with Turnip mosaic virus (TuMV) as a result of manual inoculation or aphid transmission, but infected plants have not been found in the field. In this study, B. rapa plants grown from seed collected from two field sites in southern England were mechanically inoculated with one of two distinct isolates (pathotypes) of TuMV under glasshouse conditions. These had either been isolated from Brassica oleracea growing wild in Wales, (GBR 83, pathotype 3) or Dorset (GBR 98, pathotype 1). Use of ELISA as an index of infection in manually inoculated B. rapa showed that although seed provenance had a small effect on the proportion of plants infected, the biggest factor was the virus isolate. Both virus isolates infected both lines of B. rapa , but invaded at different rates, although both resulted in easily discernible symptoms. The severity of symptoms was not related to amounts of virus in the infected plants. A significantly greater proportion of plants were infected with GBR 83 at 45 days post-inoculation (d.p.i.) than GBR 98. but GBR 98 caused significantly more severe and obvious symptoms as well as greater mortality at 119 d.p.i., in plants from both sites than GBR 83.  相似文献
