Paracoccidioidomycosis is a chronic infection that primarily affects the lungs. Here we investigated cellular and humoral immune responses after intrathoracic Paracoccidioides brasiliensis infection in BALB/c mice. P. brasiliensis-colony-forming units (CFUs), fungal DNA and granulomas in lungs increased progressively, peaking at day 90 postinfection (p.i.). IFN-γ production was highest on day 15 p.i., declining thereafter. The kinetics of the NO production was similar to that described for IFN-γ. In contrast, IL-10 increased from day 45 p.i. reaching a peak at day 90. Levels of serum IgG1 were higher than IgG2a between days 30 and 90 p.i. 30% of mice died by day 90 p.i. These data indicate that infection with P. brasiliensis by the intrathoracic route shows high IFN-γ and NO production at day 15 p.i., unable to control multiplication of fungi, which appears to be associated with a progressive increase in IL-10 and in the number and complexity of granulomas. 相似文献

ABSTRACT The aim of this study was to obtain information about genetic diversity and make some inferences about the relationship of 27 strains of Xylella fastidiosa from different hosts and distinct geographical areas. Single-nucleotide polymorphism (SNP) molecular markers were identified in DNA sequences from 16 distinct regions of the genome of 24 strains of X. fastidiosa from coffee and citrus plants. Among the Brazilian strains, coffee-dependent strains have a greater number of SNPs (10 to 24 SNPs) than the citrus-based strains (2 to 12 SNPs); all the strains were compared with the sequenced strain 9a5c. The identified SNP markers were able to distinguish, for the first time, strains from citrus plants and coffee and showed that strains from coffee present higher genetic diversity than the others. These markers also have proven to be efficient for discriminating strains from the same host obtained from different geographic regions. X. fastidiosa, the causal agent of citrus variegated chlorosis, possesses genetic diversity, and the SNP markers were highly efficient for discriminating genetically close organisms. 相似文献

A novel trypsin inhibitor (PFTI) was isolated from Plathymenia foliolosa (Benth.) seeds by gel filtration chromatography on a Sephadex G-100, DEAE-Sepharose, and trypsin-Sepharose columns. By SDSPAGE, PFTI yielded a single band with a M(r) of 19 kDa. PFTI inhibited bovine trypsin and bovine chymotrypsin with equilibrium dissociation constants (K(i)) of 4 x 10(-8) and 1.4 x 10(-6) M, respectively. PFTI retained more than 50% of activity at up to 50 degrees C for 30 min, but there were 80 and 100% losses of activity at 60 and 70 degrees C, respectively. DTT affected the activity or stability of PFTI. The N-terminal amino acid sequence of PFTI showed a high degree of homology with various members of the Kunitz family of inhibitors. Anagasta kuehniella is found worldwide; this insect attacks stored grains and products of rice, oat, rye, corn, and wheat. The velvet bean caterpillar (Anticarsia gemmatalis) is considered the main defoliator pest of soybean in Brazil. Diatraea saccharalis, the sugar cane borer, is the major pest of sugar cane crops, and its caterpillar-feeding behavior, inside the stems, hampers control. PFTI showed significant inhibitory activity against trypsin-like proteases present in the larval midguts on A. kuehniella and D. saccharalis and could suppress the growth of larvae. 相似文献

The removal of 65Zn from tidal water by underlaying sediment cores collected in a mangrove forest and a tidal creek that drains this forest in Sepetiba Bay (SE Brazil) was investigated. After 30-h experiments in laboratory microcosms, the 65Zn half-removal times from tidal creek and mangrove forest sediments were 8.7?±?1.8 and 9.2?±?0.9 h respectively. Depth penetration of 65Zn was mainly restricted to the upper 3 cm in mangrove forest cores, while detectable 65Zn activities were found in all layers (0–7 cm depth) of tidal creek cores. An unexpected 65Zn release back to the overlaying water was observed for one of the tidal creek experiments in the 12–18 h interval (corresponding to a return of 17% of the initial 65Zn activity in overlaying water), suggesting a reversibility of the 65Zn removal process (e.g., by adsorption) in tidal creek sediments. The results indicate that mangrove-vegetated sediments allowed a lower vertical mobility of Zn than observed in creek sediments and mangrove sediments appear to be less susceptible to a reversion in the process of zinc removal from overlaying water, suggesting a greater capacity to retain this metal near the water–sediment interface. This first radiotracer approach on the mangrove sediments removal of Zn from tidal waters supports earlier experimental studies employing stable Zn, contributing for a better understanding of the metal uptake kinetics by such sediments and suggesting that these sediments act as active sinks for trace metals. 相似文献

The first reports of glyphosate‐resistant (GR) sourgrass (Digitaria insularis) came from Paraguay (2005) and from South and Southeast Brazil, in Paraná (2008), and São Paulo state (2009). The hypothesis of this research is that the evolution of these resistant populations might have occurred independently. To test this hypothesis, a dose–response experiment was conducted in order to confirm resistance and inter simple sequence repeat markers subsequently were evaluated in five sourgrass populations from Paraguay (PY‐R), Paraná (PR‐S and PR‐R), and São Paulo (SP‐S and SP‐R). Resistant populations were confirmed in PY‐R, PR‐R and SP‐R. The resistant populations presented lower polymorphism than susceptible populations, possibly related to the selection pressure exerted by the herbicide. Both similarity matrix and dendrogram provided evidences that the origin of resistance in the populations from Paraguay and Paraná could be the same, which also is explained by their geographical proximity. In contrast, the evolution of GR populations in São Paulo was suggested to occur independently from other locations because resistance evolved in genetically divergent populations. This study’s data provide evidences that GR sourgrass populations from Paraguay and Paraná share a similar genetic background; therefore, it is possible that resistance from Paraguay spread to Paraná through gene flow. The population from São Paulo probably was selected independently because the high values of its genetic structure and low level of gene flow. Independent selections and seed dispersion of GR sourgrass have contributed to the rapid spread of resistance across South America. 相似文献

In the last decades, selection for improved prolificacy has resulted in higher litter sizes and has thereby increased the proportion of low birthweight (LW) piglets. It is well documented that LW piglets have lower growth performance, muscle accretion and poor carcass quality. However, little is known about the relations of birthweight with subsequent reproductive performance in gilts. This study investigated the effects of birthweight on reproductive tract and ovarian follicle development in 150‐day‐old gilts. Twenty eight female pigs of different birthweight ranges (high‐HW: 1.8–2.2 kg; low‐LW: 0.8–1.2 kg) from higher parity commercial sows were reared until 150 days of age, and their body weights were recorded at weaning, end of nursery and end of the grower‐finisher phase. The animals were killed and their reproductive tracts collected for biometrical and histomorphometrical analysis. LW gilts showed significantly lower body weights and growth rates during all phases of production compared to their HW counterparts (p < .01). Most biometrical measurements of the reproductive tract were similar between the experimental groups, except vaginal length and the gonadossomatic index (relative ovarian weight), which were affected by birthweight class (p < .05). LW females also showed fewer medium size (3–5 mm; p < .01) ovarian follicles, pre‐antral follicles (p < .07) and more atretic follicles per ovarian cortex area (p < .05). Therefore, besides the effects on post‐natal growth performance, birthweight affects vaginal length and the follicular dynamics process, which may impair the reproductive performance of replacement gilts. 相似文献

Visceral leishmaniasis (VL) is a zoonotic disease characterized by infection of mononuclear phagocytes by Leishmania chagasi. The primary vector is Lutzomyia longipalpis and the dog is the main domestic reservoir. The control and current treatment of dogs using synthetic drugs have not shown effectiveness in reducing the incidence of disease in man. In attempt to find new compounds with leishmanicidal action, plant secondary metabolites have been studied in search of treatments of VL. This study aimed to evaluate the leishmanicidal activity of Musa paradisiaca (banana tree) and Spondias mombin (cajazeira) chemical constituents on promastigotes and amastigotes of L. chagasi. Phytochemical analysis by column chromatography was performed on ethanol extracts of two plants and fractions were isolated. Thin layer chromatography was used to compare the fractions and for isolation the substances to be used in vitro tests. The in vitro tests on promastigotes of L. chagasi used the MTT colorimetric method and the method of ELISA in situ was used against amastigotes besides the cytotoxicity in RAW 264.7 cells. Of the eight fractions tested, Sm1 and Sm2 from S. mombin had no action against promastigotes, but had good activity against amastigotes. The fractions Mp1 e Mp4 of M. paradisiaca were very cytotoxic to RAW 264.7 cells. The best result was obtained with the fraction Sm3 from S. mombin with IC(50) of 11.26 μg/ml against promastigotes and amastigotes of 0.27 μg/ml. The fraction Sm3 characterized as tannic acid showed the best results against both forms of Leishmania being a good candidate for evaluation in in vivo tests. 相似文献

Ehrlichiosis and babesiosis are tick-borne diseases, caused mainly by Ehrlichia canis and Babesia canis, respectively, with a worldwide occurrence in dogs, whose main vector is the brown-dog tick, Rhipicephalus sanguineus. The present work aimed to detect the presence of E. canis and Babesia sp. in 91 dog blood samples in Colombia, by molecular and serological techniques. We also performed sequence alignment to indicate the identity of the parasite species infecting these animals. The present work shows the first molecular detection of E. canis and B. vogeli in dogs from Colombia. Immunoglobulin-G (IgG) antibodies to E. canis and Babesia vogeli were found in 75 (82.4%) and 47 (51.6%) sampled dogs, respectively. Thirty-seven (40.6%) and 5 (5.5%) dogs were positive in PCR for E. canis and Babesia sp., respectively. After sequencing, amplicons showed 99% of identity with isolates of E. canis and B. vogeli. The phylogenetic trees based on 16S rRNA-Anaplasmataceae sequences and 18S rRNA-piroplasmid sequences supported the identity of the found E. canis and B. vogeli DNAs, respectively. The present work shows the first molecular detection of E. canis and B. vogeli in dogs in Colombia. 相似文献
