It has been evident the improvement of in vitro embryo production (IVEP) in dairy cows. Nevertheless, it is known that differences in the number and quality of oocytes between taurine and zebu females impact the efficiency and economic viability of IVEP. As the insulin‐like growth factor (IGF) system is related to follicular and oocyte development, we aimed to quantify mRNA abundance of IGF system members and pregnancy‐associated plasma protein‐A (PAPPA) in the cumulus–oocyte complexes (COCs) of Gir, 1/2 Holstein × 1/2 Gir and Holstein cows. Four pools of 30 immature COCs from Gir, 1/2 Holstein × 1/2 Gir and Holstein cows were obtained by ovum pickup (OPU), and the oocytes and cumulus cells (CC) were mechanically separated and stored at ?80°C. Total RNA was extracted from pools of 30 oocytes and their respective CC. Expression of target genes was assessed by real‐time RT‐PCR. In oocytes, the abundance of IGFR1 mRNA was higher (< .05) in Gir cows compared with the other breeds. In contrast, in CC, mRNA encoding IGF2 (< .05), IGFR2 (< .05) and IGFBP4 (< .01) was higher in Holstein donors compared with Gir and 1/2 Holstein × 1/2 Gir cows. Additionally, the abundance of PAPPA mRNA was higher in oocytes (< .001) and CC (< .01) in Gir and 1/2 Holstein × 1/2 Gir cows compared with the Holstein donors. In conclusion, the higher abundance of PAPPA mRNA in the oocytes and CC from Gir and cross‐breed donors combined with the low expression of IGFBP4 in the CC suggests an enhancement of the bioavailability of IGF‐free when compared with Holstein COCs.  相似文献

1. Male and female broiler chickens (144 in total) were given diets supplemented with clenbuterol (CB) at 0 (control) and at 1 mg/kg between 28 and 49 d of age to study the effect of CB on growth, carcase and skeletal muscle.

2. CB improved growth in males by increasing daily weight gain and final live weight and by lowering food conversion ratio. In females it changed the carcase composition by reducing abdominal fat pad and by increasing the proportion of protein. Consequently, carcase protein gain was increased in both sexes (11% and 16%, respectively).

3. Skeletal muscle weights were enhanced by between 6% and 22%. Muscle fibre diameters were increased in extensor hallucis longus (EHL) but not in gastrocnemius (GAS) muscle. This increase was more pronounced in females. EHL total muscle fibre number remained unchanged. The proportion of fast‐twitch glycolytic fibres was increased at the expense of fast‐twitch oxidative fibres in males only. Nuclear/cytoplasm and DNA/protein ratios tended to be decreased by CB.

4. From the elevated EHL muscle RNA/DNA, unchanged protein/RNA and translation activity it is suggested that CB stimulated protein synthesis at the pretranslational level. Reduced protein degradation is deduced from decreased neutral calcium‐dependent proteolytic activity.

5. It is concluded that broiler chickens respond to long‐term CB treatment as has been shown in various mammals. However, the sex‐specific response in growth, carcase composition and skeletal muscle cellularity is more clearly apparent in broiler chickens.  相似文献

This article reviews control options for Neospora caninum infection and abortion in cattle, drawing on published literature and the authors' own research in this field. Apart from the successful use of embryo transfer to prevent congenital infection in calves born to infected cows, there are currently no accepted control methods for the prevention of abortions in cattle. The epidemiological data at hand suggest that concomitant infections with bovine pestivirus increase the risk of abortion significantly and that these infections, for which effective vaccines exist, should therefore be controlled. While vertical transmission appears to be the major route of infection in cattle, there is also a role for postnatal transmission, involving a definitive host. Presently, the control of dogs and their access to bovine tissues, particularly potentially infected placentae and other foetal tissues, appear to be the most prudent control methods. There are some indications that vaccination against N. caninum may aid in the prevention of abortions. Suggestions for control options are limited by our current lack of actual experiences with control strategies. Further practical fieldwork is needed in this area.  相似文献

The application of nitrogen (N) fertilizers and liming (CaCO3) to improve soil quality and crop productivity are regarded as effective and important agricultural practices. However, they may increase greenhouse gas (GHG) emissions. There is limited information on the GHG emissions of tropical soils, specifically when liming is combined with N fertilization. We therefore conducted a full factorial laboratory incubation experiment to investigate how N fertilizer (0 kg N ha−1, 12.5 kg N ha−1 and 50 kg N ha−1) and liming (target pH = 6.5) affect GHG emissions and soil N availability. We focussed on three common acidic soils (two ferralsols and one vertisol) from Lake Victoria (Kenya). After 8 weeks, the most significant increase in cumulative carbon dioxide (CO2) and nitrous oxide (N2O) fluxes compared with the unfertilized control was found for the two ferralsols in the N + lime treatment, with five to six times higher CO2 fluxes than the control. The δ13C signature of soil-emitted CO2 revealed that for the ferralsols, liming (i.e. the addition of CaCO3) was the dominant source of CO2, followed by urea (N fertilization), whereas no significant effect of liming or of N fertilization on CO2 flux was found for the vertisol. In addition, the N2O fluxes were most significantly increased by the high N + lime treatment in the two ferralsols, with four times and 13 times greater N2O flux than that of the control. No treatment effects on N2O fluxes were observed for the vertisol. Liming in combination with N fertilization significantly increased the final nitrate content by 14.5%–39% compared with N fertilization alone in all treatment combinations and soils. We conclude that consideration should be given to the GHG budgets of agricultural ferralsols since liming is associated with high liming-induced CO2 and N2O emissions. Therefore, nature-based and sustainable sources should be explored as an alternative to liming in order to manage the pH and the associated fertility of acidic tropical soils.  相似文献

Objective To assess quantitatively the spatial distribution of seroconversion of Queensland cattle to bluetongue viruses.
Design A sentinel herd study. Sample population Sixty-nine sentinel herds at 30 locations.
Procedure Spatial clustering of seroconversion to blue-tongue viruses was investigated during the period from 1990 to 1994.
Results Seroconversion to only two bluetongue virus serotypes, 1 and 21, was observed. The 14 herds, in which seroconversion to bluetongue virus serotype 1 was detected, were located only along the eastern coastal and subcoastal region of Queensland, and were significantly (P < 0.05) clustered. Locations at which seroconversion to serotype 21 was detected, were not significantly clustered. The results generally agree with field observations, except for the failure to detect seroconversion to bluetongue viruses in north-western Queensland.
Conclusion Bluetongue infection of cattle in north-western Queensland may be temporally sporadic. The dominance of serotype 1 in the Queensland cattle population may be the result of differential transmission by potential vector species. Long-term surveillance programs are important for defining disease status of animal populations.  相似文献

In periods of social crisis, policymakers become particularly vulnerable to interest groups mobilizing to compete for scarce funds. At this point, legislators are no longer able to address the specific needs of their primary constituency directly, but rather are forced to do so in pretext only. New, unfamiliar technologies provide ample ammunition for astute interest groups to take advantage of times of economic turmoil and maneuver for policy support through dramatic campaigns of “salesmanship.” By publicizing a crisis situation, dramatizing it effectively, and advertising an innovation as the solution to the crisis, legislators may be effectively persuaded to give priority to interest group pressures above and beyond those of the local constituency. Iowa's attempts to address the farm crisis through economic development strategies relying on biotechnology is examined in this paper. The results of extensive surveys of Iowa's legislators and farmers are examined and the consequences for Iowa's policy process of using biotechnology under the auspices of economic development are discussed.  相似文献

Biosecurity risks are a major threat to the profitability of the industry as well as impacting human and animal health. Livestock producers play a crucial role in biosecurity as the first to notice changes in the health or productivity of their stock and are generally responsible for implementing protective measures. However, uptake of biosecurity measures by producers is variable. We critically appraised the current literature regarding biosecurity practices in Australian livestock industries and highlight aspects that are well understood as well as those where further research or information is needed. Findings from 12 cross-sectional studies suggest that Australian producers' knowledge of biosecurity methods and importance might have a positive influence on their willingness to implement or incorporate biosecurity practices. There is moderate evidence supportive of biosecurity being well understood by livestock producers across Australia. Barriers to producers using biosecurity practices included lack of information or communication from agricultural, veterinary or government organisations. It was found that larger stock numbers were positively correlated with biosecurity implementation and that producers used veterinarians, government and industry agencies as resources for trusted information.  相似文献
