Spatial genetic structure and dispersal of the cacao pathogen Moniliophthora perniciosa in the Brazilian Amazon 下载免费PDF全文

Moniliophthora perniciosa is the causal agent of witches’ broom in Theobroma cacao (cacao). Three biotypes of M. perniciosa are recognized, differing in host specificity, with two causing symptoms on cacao or Solanaceae species (C‐ and S‐biotypes), and the third found growing endophytically on lianas (L‐biotype). The objectives of this study were to clarify the genetic relationship between the three biotypes, and to identify those regions in the Brazilian Amazon with the greatest genetic diversity for the C‐biotype. Phylogenetic reconstruction based on the rRNA ITS regions showed that the C‐ and S‐biotypes formed a well‐supported clade separated from the L‐biotype. Analysis of 131 isolates genotyped at 11 microsatellite loci found that S‐ and especially L‐biotypes showed a higher genetic diversity. A significant spatial genetic structure was detected for the C‐biotype populations in Amazonia for up to 137 km, suggesting ‘isolation by distance’ mode of dispersal. However, in regions containing extensive cacao plantings, C‐biotype populations were essentially ‘clonal’, as evidenced by high frequency of repeated multilocus genotypes. Among the Amazonian C‐biotype populations, Acre and West Amazon displayed the largest genotypic diversity and might be part of the centre of diversity of the fungus. The pathogen dispersal may have followed the direction of river flow downstream from Acre, Rondônia and West Amazon eastward to the rest of the Amazon valley, where cacao is not endemic. The Bahia population exhibited the lowest genotypic diversity, but high allele richness, suggesting multiple invasions, with origin assigned to Rondônia and West Amazon, possibly through isolates from the Lower Amazon population. 相似文献

The objectives of this work were to evaluate the genetic variability of Meloidogyne enterolobii by molecular markers, and develop species‐specific molecular markers for application in detection. Sixteen M. enterolobii isolates from different geographical regions (Brazil and other countries) and hosts were used in this study. The identification and purification of the populations were carried out based on isoenzyme phenotype. The DNA amplification of the intergenic region (IGS) of the rDNA and of the region between the cytochrome oxidase subunit II (COII) and 16S rRNA genes (mtDNA) produced specific fragments of the expected size for this nematode, i.e. 780 and 705 bp, respectively. Intraspecific variability among the isolates was evaluated with three different neutral molecular markers: AFLP, ISSR and RAPD. The results showed a low level of diversity among the isolates tested, indicating that M. enterolobii is a genetically homogeneous root‐knot nematode species. The RAPD method allowed the identification of a species‐specific RAPD fragment for M. enterolobii. This fragment was cloned and sequenced, and from the sequence obtained, a set of primers was designed and tested. The amplification of a 520‐bp‐long fragment occurred only for the 16 isolates of M. enterolobii and not for the 10 other Meloidogyne species tested. In addition, positive detection was achieved in a single individual female, egg‐mass and second stage juvenile of this nematode. This SCAR species‐specific marker for M. enterolobii represents a new molecular tool to be used in the detection of this nematode from field samples and as a routine diagnostic test for quarantine devices . 相似文献

Marine life has proved to be an invaluable source of new compounds with significant bioactivities, such as xanthones. This review summarizes the advances made in the study of marine-derived xanthones from 2010 to 2021, from isolation towards synthesis, highlighting their biological activities. Most of these compounds were isolated from marine-derived fungi, found in marine sediments, and associated with other aquatic organisms (sponge and jellyfish). Once isolated, xanthones have been assessed for different bioactivities, such as antibacterial, antifungal, and cytotoxic properties. In the latter case, promising results have been demonstrated. Considering the significant bioactivities showed by xanthones, efforts have been made to synthesize these compounds, like yicathins B and C and the secalonic acid D, through total synthesis. 相似文献

Aloe barbadensis Miller, known as Aloe Vera, requires limited irrigation depending on the capacity of the soil to retain humidity, since it is a CAM species and thus naturally adapted to conditions of dryness and high temperatures. Therefore, we postulated that plants of Aloe Vera plants under conditions of water deficit should improve their water use efficiency (WUE) by performing osmotic adjustment (OA) with a temporal correlation between WUE and OA. The objective of the investigation was to determine the effect of water restriction on the WUE and OA of A. barbadensis under different water treatments. 18-month old Aloe Vera plants were cultivated in pots with a soil substrate that was a mixture of equal parts of sand and organic matter with 18% of FC and 9% of permanent wilting point. To determine the effects of the soil humidity on plant WUE and OA, four treatments were arranged in a complete random design with four repetitions; these were 100%, 75%, 50% and 25% of FC, which correspond to an evatranspiration of 11.4, 9.6, 4.0 and 1.7 L per plant, respectively. The water treatments were maintained by frequent irrigation. The following variables were determined: dry matter, leaf water potential, relative water content (RWC), amount of gel produced, sap flow, proline content, soluble and total sugars and oligo and polyfructans. Aloe Vera increased WUE with increasing water deficit; the sap flow rate decreased with water restrictions, and the plants performed osmotic adjustment by increasing the synthesis of proline, soluble and total sugars as well as the amounts of oligo and polyfructans, mainly polymers of β-(2 → 6) kestotriose, changing from the inulin type to the neofructan type. The plants most and less irrigated (100% and 25% of FC) were the groups with lowest WUE. Harmful algal blooms pose a challenge regarding food safety due to their erratic nature and forming circumstances which are yet to be disclosed. The best strategy to protect human consumers is through legislation and monitoring strategies. Global warming and anthropological intervention aided the migration and establishment of emerging toxin producers into Europe’s temperate waters, creating a new threat to human public health. The lack of information, standards, and reference materials delay effective solutions, being a matter of urgent resolution. In this work, the recent findings of the presence of emerging azaspiracids, spirolildes, pinnatoxins, gymnodimines, palitoxins, ciguatoxins, brevetoxins, and tetrodotoxins on European Coasts are addressed. In the processing of fruits such as blueberry (Vaccinium sp), that has high levels of phenolic acid, the food industry produces tons of organic waste that causes harm to the environment. Encapsulation is a technique used to take advantage of these wastes. Several methods are used to encapsulate substances, among them ionotropic gelation proves to be a simple, precise, efficient and economical method for obtaining particles with encapsulated bioactives. In this manner, the aim of this study was to test sodium alginate as wall material to encapsulate blueberry residue by ionotropic gelation. The microbeads were characterized by scanning electron microscopy (SEM), x-ray diffraction (XRD), total phenolic compounds, antioxidant capacity and in vitro dissolution. The results showed that the microbeads had surface invagination; retention of 67.01% of the phenolic compounds after encapsulation and 68.2%, phenolic release 120 min after in vitro dissolution. The results suggest that the tested matrix was suitable for encapsulation. The produced microbeads are promising for applications in food products, once the phenolic compounds present in the blueberry residues were maintained after encapsulation. 相似文献

Downy mildew of soybean, caused by Peronospora manshurica, is widely spread throughout Brazil. The objective of this study was to evaluate the use of potassium phosphite to control this disease. Field experiments were conducted during the growing season of 2006/2007 and 2007/2008 in the state of Parana in southern Brazil. The experimental design consisted of completely randomized blocks in a factorial arrangement (4 × 2) with four replications. Four rates of potassium phosphite (0, 375, 750 and 1500 g P2O5 + K2O ha−1) were applied at two growth stages, V6 (fifth trifoliolate leaf) and R2 (full flowering), followed by one or two applications of pyraclostrobin and epoxiconazole (66.5 + 25 g a.i. ha−1) at R3 (pod development) or R2 and R5.1 (10% of pod filling), mainly for the control of Asian soybean rust (Phakopsora pachyrhizi) and powdery mildew (Microsphaera diffusa). Field experiments were conducted to quantify the severity of downy mildew on leaves, nutrient content in leaf tissue (N, P and K), leaf area index (LAI), yield and seed weight. The maximum severity of downy mildew was observed at a growth stage of R5.3 (50% of pods were ripe), with 14% and 46% of the leaf area affected in 2006/2007 and 2007/2008, respectively. Also it was detected some effect of phosphite on Asian rust control but it was mostly in the trial of 2007/08 when the epidemic was very low (9.7-21.8% of severity). There was a linear reduction in the severity of downy mildew and a significant improvement in the LAI with an increase in the rate of phosphite applied. During the 2006/2007 growing season, a significant yield improvement was observed due to the application of the highest rate of phosphite. Two fungicide applications following phosphite application significantly improved the control of Asian soybean rust and powdery mildew, yield and seed weight when compared to a single fungicide application. 相似文献

Cellulose acetate is one of the components employed in drug controlled-release systems in the form of membranes. The aim of this study was to examine the controlled-release of doxycycline employing cellulose acetate symmetric and asymmetric membranes as matrices. The cellulose triacetate was produced from sugarcane bagasse through a homogeneous acetylation reaction, using acetic acid as the solvent, acetic anhydride as the acetylating agent and sulfuric acid as the catalyst. The viscosity average molecular weight of the cellulose acetate produced was 39,000 g mol−1. The symmetric membranes were produced using a system solvent of dichloromethane/ethanol (9:1, v/v) and the asymmetric membranes were produced from the same solvent system and 10% of water. For the formulation of both, 5% of doxycycline was used. The membranes were characterized by thermal analysis (DSC and TGA) and scanning electron microscopy SEM. The release of doxycycline through cellulose triacetate matrices was examined using spectrophotometric analysis in the ultraviolet-visible region, at 275 nm. The results revealed that asymmetric membranes release 80% of the drug in 100 min, while symmetric membranes release 14% of the drug during the same time interval. 相似文献

The morphometric study of spermatogenic cysts in sexually mature tilapias, during the evolution of spermatogenesis, showed a dramatic increase in both number of germ cells and cyst volume. However, the opposite trend was observed for germ cell size. Nevertheless, the number of Sertoli cells increased gradually up to leptotene/zygotene cysts, stabilizing thereafter. Based on the number of spermatids supported by each Sertoli cell and compared to mammals, Sertoli cell efficiency in tilapias is remarkably high. Sertoli cell proliferation was frequently observed, mainly in spermatogonial cysts, and probably is the major factor related to the testis growth and the increase in sperm production that normally occurs in adult tilapias. The combined duration of spermatocytes (5 days) and spermiogenic (5–6 days) phases of spermatogenesis in fish kept at 25 °C was 10–11 days. Mainly due to acceleration in meiosis, these two phases lasted a total of 6 days in tilapias kept at 30 °C, in the opposite way, at 20 °C spermatogenesis was arrested at pachytene spermatocytes. To our knowledge, this is the most comprehensive investigation performed up to date on testis morphometry and function in adult tilapias. 相似文献
