Objectives To examine strain variation amongst Australian isolates of Mycobacterium paratuberculosis .
Design Pulsed field gel electrophoresis was optimised for differentiation of M paratuberculosis strains, and this typing technique was then applied to a collection of Australian isolates.
Procedure DNAs from 35 Australian isolates of M para-tuberculosis and a UK reference strain were digested with one or other of three restriction endonucleases. The banding patterns obtained after pulsed field gel electrophoresis of the DNA fragments were compared.
Results The Australian isolates were divided into two groups on the basis of their DNA banding pattern. Both were different from the UK reference strain. Seven isolates from cattle in Victoria and the Northern Territory had the same pattern as five isolates from alpacas in Victoria and Western Australia. Another 20 isolates from cattle in Victoria, Western Australia and the Northern Territory had the same pattern as isolates from two sheep and a goat in New South Wales.
Conclusion Pulsed field gel electrophoresis was a useful tool for strain typing of M paratuberculosis , and could be used to study the transmission of strains in Australia.  相似文献

Aphidicidal activity of hot and cold water extracts of some indigenous plants, Azadirachta indica A. Juss (neem), Calotropis procera (Aiton) W.T. Aiton (akanda), Polygonum hydropiper L. (biskatali) and Ipomoea sepiaria J. Koenig ex Roxb. (bankalmi), were tested against the bean aphid, Aphis craccivora Koch. Hot water extract of P. hydropiper and A. indica was found to be the most effective (87.6–94.5 and 80.47–89.6% mortality respectively, P < 0.01) among all the extracts. Other hot and cold water extracts also appeared to be useful (59.5–87.5% mortality) as pesticides for A. craccivora. The highest yield (3.25 kg per plant) was obtained using hot water extract of P. hydropiper followed by hot water extract of A. indica (3.15 kg per plant). The lowest yield (0.32 kg per plant) was recorded from the control block. All the phytoproduct treatments had significantly (P < 0.01) better yield than the control block.  相似文献

Fusarium head blight (FHB), a devastating disease that affects wheat, is caused by a complex of Fusarium species. The overall impact of Fusarium spp. in wheat production arises through the combination of FHB and mycotoxin infection of the grain harvested from infected wheat spikes. Spike infection occurs during opening of flowers and is favoured by high humidity or wet weather accompanied with warm temperatures. Available possibilities for controlling FHB include the use of cultural practices, fungicides and biological approaches. Three cultural practices are expected to be of prime importance in controlling FHB and the production of mycotoxins: soil preparation method (deep tillage), the choice of the preceding crop in the rotation and the selection of appropriate cultivar.  相似文献

A transition from metal-like double-layer capacitive charging to redox-like charging was observed in electrochemical ensemble Coulomb staircase experiments on solutions of gold nanoparticles of varied core size. The monodisperse gold nanoparticles are stabilized by short-chain alkanethiolate monolayers and have 8 to 38 kilodaltons core mass (1.1 to 1.9 nanometers in diameter). Larger cores display Coulomb staircase responses consistent with double-layer charging of metal-electrolyte interfaces, whereas smaller core nanoparticles exhibit redox chemical character, including a large central gap. The change in behavior is consistent with new near-infrared spectroscopic data showing an emerging gap between the highest occupied and lowest unoccupied orbitals of 0.4 to 0.9 electron volt.  相似文献

Abstract

Aerobic rice is a potential water-wise rice production system, but high weed infestation has threatened its sustainability, which demands an efficient and cost-effective weed management technique. Eight commercial herbicide products were applied singly or as tank-mix or in sequence to evaluate their efficacy, rice selectivity and cost-effectiveness in aerobic rice. The study was conducted under field conditions in Malaysia during 2010/2011 following a randomized complete block design. Most of the herbicide treatments provided excellent weed control, and produced much higher net benefit than weedy or weed-free check. None of the herbicides caused significant phytotoxicity to rice plants. Among the herbicide treatments, sequential application of Cyhalofop-butyl+Bensulfuron at early growth stage followed by Bentazon/MCPA at mid growth stage provided the highest weed control efficiency, productivity and net benefit. Application of Bispyribac-sodium at early growth stage followed by Bentazon/MCPA at mid growth stage performed very close to the above-mentioned treatments. Sequential application of Pretilachlor/safener just after seeding followed by Propanil/Thiobencarb at early growth stage also provided satisfactory results in terms of efficacy and economic return. Since manual weeding was not economic, herbicide rotation using the above chemicals may be recommended for effective weed management in aerobic rice.  相似文献
