The molecular mechanism to control the oxidative burst exerted by Mn accumulation in rice (Oryza sativa L.) plants, grown in hydroponics containing 0.5, 2 and 8 mg l–1 Mn and irradiated with a total biological effective UV‐B irradiation of 20.825 kJ m?2, was investigated in the chloroplasts at the 15th and 21st days after germination. In both experimental periods, Mn accumulation kinetics in the leaves and in the chloroplast lamellae displayed overall increases. Coupled to higher membrane selectivity, superoxide production and acyl lipids peroxidation in the thylakoids decreased, prompting upper rates of the Hill and Mehler reactions. Connected to UV‐B irradiation, higher accumulated Mn in thylakoids was found to be chelated in a 36.5 kDa protein, with Mn/protein ratio of about 1 and high content of Gln, Asp, Glu, Leu and Gly, being its EPR spectrum characteristic of high‐spin Mn(II), in a S = 5/2 ground state. As this protein exhibited enzymatic catalysis of superoxide dismutation, it was concluded that, under UV‐B irradiation, the high internal tolerance of Oryza sativa L. to Mn during the vegetative growth also triggers the synthesis of a manganese protein that mimics superoxide dismutase functioning, therefore furnishing an additional intimate protection against oxidative stress.  相似文献

Potato cultivars Norland, Kennebec, and Russet Burbank were inoculated with a Minnesota isolate of potato virus S (PVS) in the field. The incidence of secondary infection, measured by enzyme-linked immunosorbent assay (ELISA), indicated that significantly more PVS infection occurred in Russet Burbank than Norland or Kennebec, and that Norland had significantly more infection than Kennebec (P≤ 0.01). Mature plant resistance was not observed. Rapid reinfection of PVS-free seedlots resulted from inoculum point-sources when routine cultivation practices were followed. Reinfection rates were significantly greater for seedlots grown at Grand Forks, ND, when compared with seedlots grown at Becker, MN, during both years of the study (P≤ 0.01). After two years in the field, reinfection rates for Norland (71.8%) and Russet Burbank (73.0%) did not differ significantly, but were significantly greater than the reinfection rate for Kennebec (29.5%) (P≤ 0.01). In greenhouse-grown plants, PVS moved out of rub-inoculated leaves within 24 hr, however, 13 and 20 days were required before PVS was detected with ELISA in foliage above and below the inoculated leaf, respectively. The frequency of PVS detection was significantly greater in foliage above the inoculated leaf compared to foliage below the inoculated leaf (P≤ 0.05). Translocation of PVS from inoculated leaves to daughter tubers occurred within 13 days for Russet Burbank and Norland and within 20 days for Kennebec.  相似文献

Until recently, it has been impossible to approach learning with the techniques of cell biology. During the past several years, elementary forms of learning have been analyzed in higher invertebrates. Their nervous systems allow the experimental study of behavioral, neurophysiological, morphological, biochemical, and genetic components of the functional (plastic) changes underlying learning. In this review, we focus primarily on short-term sensitization of the gill and siphon reflex in the marine mollusk, Aplysia californica. Analyses of this form of learning provide direct evidence that protein phosphorylation dependent on cyclic adenosine monophosphate can modulate synaptic action. These studies also suggest how the molecular mechanisms for this short-term form of synaptic plasticity can be extended to explain both long-term memory and classical conditioning.  相似文献

The blue mussel Mytilus edulis is a commercially important species whose fishery and culture generally relies on natural spat collection. Hatchery-production could provide an alternative source of seed, enabling reliable expansion of the industry. Mussel spawning and larval rearing trials were carried out to optimise elements of hatchery production. Culturing fertilised eggs at low density (20–200 eggs cm−2) rather than high density (400–720 eggs cm−2) significantly improved the quality of first veliger larvae and differences in this improvement were evident between the eggs from different females (maternal effects). Veliger larval growth at 17 or 21°C was significantly faster than growth at 14°C. Feeding veliger larvae an identical total ration either daily or at 2–3 day intervals did not significantly affect their growth. Different microalgal diets (1: Isochrysis sp. (clone T-ISO), 2: Chaetoceros calcitrans forma pumilus, 3: C. muelleri, 4: mixed Isochrysis sp. (clone T-ISO) and C. calcitrans f. pumilus, and 5: mixed Isochrysis sp. (clone T-ISO) and C. muelleri) were tested on veliger larval growth and mixed diets outperformed single-species diets.  相似文献

This study examined the effects of feeding frequency (daily versus every other day [EOD]) on nutrient digestibility/availability of channel catfish, Ictalurus punctatus, reared at optimal (30 C) and suboptimal (24 C) temperatures. A 28% protein practical diet was used as the test diet, and chromic oxide was used at 0.75% of the diet as a maker. Twenty channel catfish of a mean weight of 141 g/fish were stocked into 12 flow‐through aquaria (110 L). Fish were fed daily or EOD to apparent satiation for 7 d before fecal samples were collected by the dissection method. Fish fed EOD to apparent satiation consumed more diet on days fed than those fed daily, and fish reared at 30 C consumed more diet than those reared at 24 C. Neither feeding frequency nor water temperature significantly affected apparent digestibility coefficients for energy and protein and apparent availability coefficients for total amino acids. Results show no advantage by feeding EOD over feeding daily on nutrient digestibility and diet utilization efficiency.  相似文献
