Technologies to edit the zygote genome have revolutionized biomedical research not only for the creation of animal models for the study of human disease but also for the generation of functional human cells and tissues through interspecies blastocyst complementation technology. The pig is the ideal species for these purposes due to its great similarity in anatomy and physiology to humans. Emerging biotechnologies require the use of oocytes and/or embryos of good quality, which might be obtained using in vitro production (IVP) techniques. However, the current porcine embryo IVP systems are still suboptimal and result in low monospermic fertilization and blastocyst formation rates and poor embryo quality. During recent years, intensive investigations have been performed to evaluate the influence of specific compounds on gametes and embryos and to avoid the use of undefined supplements (serum and serum derivate) in the incubation media. However, little consideration has been given to the use of the mineral oil (MO) to overlay incubation droplets, which, albeit being a routine component of the IVP systems, is a totally undefined and thus problematic product for the safety of gametes and embryos. In this review, we provide an overview on the advantages and disadvantages of using MO to cover the incubation media. We also review one important concern in IVP laboratories: the use of oils containing undetected contamination. Finally, we discuss the effects of different types of oils on the in vitro embryo production outcomes and the transfer of compounds from oil into the culture media.  相似文献

An average value (n=22) of Youngs modulus in the axial direction of woodwind reed grade Arundo donax has been determined from 10-5 s to 104 s, at room temperature in both the ambient-dry and saturated-wet states, using ultrasonic to creep measurements. A broad relaxation in the modulus occurs between 10-3 s and 10-1 s, in keeping with results obtained by others in previous experiments at 0.034 s in flexural resonance bar measurements. The modulus-versus-time results are fitted to a Maxwell–Weichert phenomenological model. This model allows the room temperature viscoelastic properties to be predicted as a function of frequency. The model predicts the measured damping quite well in the relaxation region.Measurements at the ultrasonic frequency of 60 kHz were made over the temperature range –35 to +35°C to seek other relaxations. The so-called -peak found at low frequencies (0.05 to 110 Hz) by other researchers appears to have been observed at the ultrasonic frequency.  相似文献

Search methods that people use to find owners of lost pets   总被引:1,自引:0,他引:1  
OBJECTIVE: To characterize the process by which people who find lost pets search for the owners. DESIGN: Cross-sectional study. Sample Population-188 individuals who found a lost pet in Dayton, Ohio, between March 1 and June 30, 2006. Procedures-Potential participants were identified as a result of contact with a local animal agency or placement of an advertisement in the local newspaper. A telephone survey was conducted to identify methods participants used to find the pets' owners. RESULTS: 156 of 188 (83%) individuals completed the survey. Fifty-nine of the 156 (38%) pets were reunited with their owners; median time to reunification was 2 days (range, 0.5 to 45 days). Only 1 (3%) cat owner was found, compared with 58 (46%) dog owners. Pet owners were found as a result of information provided by an animal agency (25%), placement of a newspaper advertisement (24%), walking the neighborhood (19%), signs in the neighborhood (15%), information on a pet tag (10%), and other methods (7%). Most finders (87%) considered it extremely important to find the owner, yet only 13 (8%) initially surrendered the found pet to an animal agency. The primary reason people did not surrender found pets was fear of euthanasia (57%). Only 97 (62%) individuals were aware they could run a found-pet advertisement in the newspaper at no charge, and only 1 person who was unaware of the no-charge policy placed an advertisement. CONCLUSIONS AND CLINICAL RELEVANCE: Veterinarians and shelters can help educate people who find lost pets about methods to search for the pets' owners.  相似文献

Ultrasound is one of the most promising forms of non‐invasive contraception and has been studied in several animal models. The objective of the current investigation was to determine the most practical and effective application protocol for dog sterilization. A total of 100 dogs were divided into five equal groups. Group A received 5‐min applications three times performed at 48‐hr intervals and covering the entire testicular area at frequency of 1 MHz; Group B received 5‐min applications three times performed at 48‐hr intervals over the dorso‐cranial area of the testis at frequency of 3 MHz; Group C received three sequential 5‐min applications (at 5‐min intervals between applications) covering the entire testicular area at frequency of 1 MHz; Group D received 15‐min applications two times performed at 48‐hr intervals and covering the entire testicular area at frequency of 1 MHz. The experimental groups' ultrasound had an intensity of 1.5W/cm2. The Control Group had the same procedure as Group A, but with the transducer switched‐off. Dogs were surgically castrated 40 days following the treatment for histological examination. Azoospermia, testicular volume reduction and apparently irreversible testicular damage were achieved by Group A. No effects were noticed in the other groups. Testosterone levels remained within physiological range with all application protocols. A regimen of three applications of ultrasound at 1 MHz, and 1.5 W/cm2, lasting 5 min with an interval of 48 h was effective as permanent sterilization in the dog without hormonal impact.  相似文献

Objectives : The objective of the study was to examine the changes in vertebral heart scale, and left atrial and ventricular dimensions before and at onset of congestive heart failure in cavalier King Charles spaniels with mitral regurgitation. Methods : Records and radiographs from 24 cavalier King Charles spaniels with mitral regurgitation were used. Vertebral heart scale (24 dogs), and left atrial dimension and left ventricular end diastolic and end systolic diameters (18 dogs) and their rate of increase were measured at intervals over years to the onset of congestive heart failure. They were plotted against time to onset of congestive heart failure. Results : Dimensions and rates of change of all parameters were highest at onset of congestive heart failure, the difference between observed and chance outcome being highly significant using a two-tailed chi-square test (P<0·001). Clinical significance : The left heart chambers increase in size rapidly only in the last year before the onset of congestive heart failure. Increasing left ventricular end systolic dimension is suggestive of myocardial failure before the onset of congestive heart failure. Rate of increase of heart dimensions may be a useful indicator of impending congestive heart failure.  相似文献

Abstract. Precision farming is the process of adjusting husbandry practices within a field according to measured spatial variability. In this review, we explore the prospects for precision farming using the principles that underly conventional soil management and agronomy. The cost-effectiveness of precision farming is determined by the cost of defining zones within fields, the stability of zones through time, the difference in treatment between zones in terms of cost, and the responsiveness of the crop in terms of yield and quality to changes in treatment. Cost-effective precision farming is most likely where prior knowledge indicates large heterogeneity and where treatment zones can be predicted, for example from soil type or field history. Soil related factors are likely to provide the main basis for precision farming because they tend to be stable through time and influence crop performance. In particular, soil mapping may usefully indicate the moisture available for crop growth, organic matter maps may be utilized for precision application of fertilizers and soil acting herbicides, and variation in soil pH can be mapped and used as a basis for variable lime application. However, comprehensive nutrient mapping is less likely to be economic with existing techniques of chemical analysis. The value of yield mapping lies in identifying zones which are sufficiently stable to be of use in determining future practices. Maps of grain quality and nutrient content would significantly augment the value of yield maps in guiding marketing decisions and future agronomy. Interactions between soil differences and seasonal weather are large, so yield maps show considerable differences from season to season. Interpretation of such maps needs to follow a careful, informed, analytical process. Extensive and thorough field experimentation by crop scientists over many years has shown that yield variation arises as a result of a large and complex range of factors. It is highly improbable that simple explanations will be appropriate for much in-field yield variation. However, the capacity to sense yield variability within fields as opposed to between fields, where there are many confounding differences, provides an opportunity for the industry to improve its understanding of soil-based effects on crop performance. This should support its decision taking, whether through precision farming or through field-by-field agronomy. The main obstacle to the adoption of precision farming is the lack of appropriate sensors. Optimal sensor configurations that will measure the specific needs identified by end-users need to be developed. The conclusions reached in this paper probably apply to farming throughout northern Europe.  相似文献

Coupled radiocarbon and thorium-230 dates from benthic coral species reveal that the ventilation rate of the North Atlantic upper deep water varied greatly during the last deglaciation. Radiocarbon ages in several corals of the same age, 15.41 +/- 0.17 thousand years, and nearly the same depth, 1800 meters, in the western North Atlantic Ocean increased by as much as 670 years during the 30- to 160-year life spans of the samples. Cadmium/calcium ratios in one coral imply that the nutrient content of these deep waters also increased. Our data show that the deep ocean changed on decadal-centennial time scales during rapid changes in the surface ocean and the atmosphere.  相似文献

Patent ductus arteriosus with pulmonary hypertension and right to left shunting of blood flow was diagnosed in a 9-month-old female cat. Because of the pulmonary hypertension, the cat did not have typical signs of patent ductus arteriosus; thus, cardiac angiography and catheterization were utilized to confirm the diagnosis. Scintigraphic evaluation of the spinous processes of the equine spine has been done by subjective evaluation of radiotracer uptake in clinically abnormal horses. To determine the range of variation in asymptomatic riding horses, 33 normal horses were examined. Two 60 degrees oblique views of the thoracic and lumbar spine were evaluated subjectively, and a semi quantitative method based on standardized regions of interest in each spinous process in T10-L3 was also applied. A ratio between each spinous process and a reference area (Rib 15 or 16) was calculated. The effects of two different color displays and a smoothing filter were assessed. The results of the subjective evaluation was compared to the calculated ratios, and the associations between age, gender, weight, height, use and increased radiotracer uptake, and ratios were analyzed. No significant association was found between age, gender, weight, height, use and increased radiotracer uptake, or ratio. One or more areas of increased radiotracer uptake between T13 and T18 were common in the asymptomatic riding horse, which may have implications for the interpretation of clinical patients. The blue, green and red color display had a higher sensitivity for detecting increased radiotracer uptake compared to the continuous grayscale. The smoothing filter used in this study had no effect on the detection of areas with increased radiotracer uptake. The association between the results of the subjective evaluation and the semi quantitative method was apparent when analyzed in T13-17. To evaluate if the semi quantitative method can replace the subjective evaluation, studies on clinically affected horses are necessary.  相似文献
