Grass-clover mixtures are essential in many low-N-input cropping systems, but the importance of various root fractions for the below-ground N dynamics are not well understood. This may be due to the difficulties of studying root longevity and turnover in situ in mixtures. The present field study, investigated (1) the development in root biomass over two growing seasons and (2) the turnover of dual 15N- and 14C-labelled ryegrass and white clover root material. Litter bags containing various dual-labelled plant materials were incubated in cylinders inserted in the topsoil of a young ryegrass-clover ley. Disappearance of 14C and 15N from the litter bag material were studied for 1 year following incubation. Four times during two growing seasons, roots were divided into two classes: large roots, retained on a 1-cm sieve, and small roots, passing a 1-cm sieve but retained on a 100-µm sieve. Large root biomass increased during the two growing seasons, and small root biomass increased during the growing seasons but decreased during autumn and winter. White clover roots lost 14C and 15N almost twice as fast as ryegrass roots. The disappearance pattern of 14C and 15N from dual-labelled ryegrass and white clover roots and the C and N contents of the recovered root material indicate that large roots are determining soil C pool build-up, whereas small roots determine soil N pool build-up.  相似文献

In Denmark, it was agreed to lower the Salmonella prevalence in pork to 1.2% before the end of 2006. The current control did not seem to be sufficient to attain this goal. Therefore, four alternatives to the existing Danish control strategy for Salmonella in pork were compared in a cost-benefit analysis: (1) hot-water decontamination of all pigs at slaughter, (2) sanitary slaughter of pigs from herds with high levels of Salmonella, (3) use of home-mixed feed in herds with slaughter pigs and (4) use of acidified feed for slaughter pigs. The data originated from official statistics, published papers as well as expert opinion. The partial cost-benefit analysis was restricted to slaughterhouses affiliated with the Danish Meat Association and Danish human cases ascribable to pork from these slaughterhouses. Only hot-water decontamination was socio-economically profitable. Hot-water decontamination had a net present value over 15 years of 3.5 million euros. For sanitary slaughter the net present value was - 43.6 million euros, for home-mixed feed it was - 262.3 million euros and for acidified feed it was - 79.9 million euros. For all alternatives the costs were born solely by the pig sector, whereas primarily the consumers and public authorities received the benefits. Betulinic acid is a pentacyclic triterpene found in many plant species, among others, in the bark of white birch Betula alba. Betulinic acid was reported to display a wide range of biological effects, including antiviral, antiparasitic, antibacterial, anticancer and anti-inflammatory activities. The effects of betulinic acid (50, 5, 0.5 mg/kg) administered orally five times at 24 hours intervals to non-immunized and red blood cells (SRBC)-immunized mice were determined. The present study examined the total number of lymphocytes in the thymus, spleen and mesenteric lymph nodes, and the percentage of subsets of T cells (CD4+CD8+, CD4CD8, CD4+, CD8+) in thymus,T (CD3+, CD4+, CD8+) and B (CD19+) lymphocytes in the spleen and mesenteric lymph nodes, as well as white blood cell (WBC) and differential leukocyte counts in non-immunized mice, and humoral immune response in SRBC-immunized mice. SRBC was injected 24 hours after administration of the last dose of betulinic acid. It was found that betulinic acid administered orally five times at the dose of 0.5 mg/kg increased the total number of thymocytes, splenocytes, lymphocytes of mesenteric lymph node cells, and the weight ratio of the spleen and mesenteric lymph nodes in non-immunized mice. Betulinic acid also changed the percentage of T cell subsets in the thymus and T and B lymphocytes in peripheral lymphatic organs. The effects of betulinic acid on T and B cell subpopulations depended on the dose applied. The strongest stimulating effect of betulinic acid was observed when the drug was administered at the dose of 0.5 mg/kg. Five exposures to betulinic acid (0.5 mg/kg) decreased the percentage of immature CD4+CD8+ thymic cells with corresponding increases in the percentage and absolute count of mature, single-positive CD4+ thymocytes and decreased the percentage and total count of CD3+ splenocytes and mesenteric lymph node cells with corresponding decreases in the percentage and absolute count of CD4+ and CD8+ cells. Multiple administration of betulinic acid at the investigated doses augmented the percentage and absolute count of CD19+ cells in the peripheral lymphatic organs. Moreover, betulinic acid at the dose of 5 mg/kg administered prior to SRBC immunization increased the number of plaque forming cells (PFC) but decreased the production of anti-SRBC antibodies on day 4 after priming. Thus, betulinic acid is a potential biological response modifier and may strengthen the immune response of its host.  相似文献

Several studies have focused on the formation and losses of dissolved organic matter in forest systems, whereas a limited number have dealt with this aspect in agricultural soils. The purpose of this study was to estimate the leaching of dissolved organic carbon (DOC) and nitrogen (DON), with focus on the period after cultivating grass-clover swards. Grass-clovers were ploughed in the spring prior to sowing cereals followed by either catch crops or bare soil. The concentrations of DOC and DON decreased with soil depth and ranged at 90-cm soil depth between 7 and 21 mg C L−1 and between 1 and 3 mg N L−1, respectively, in a sandy loam soil, and between 16 and 63 mg C L−1 and between 1 and 10 mg N L−1, respectively, in a coarse sandy soil. The resulting DOC/DON ratios were in the range between 2 and 42, with higher values in the coarse sandy soil than in the sandy loam soil. The total percolation was 218 mm in the sandy loam soil and 596–645 mm in the coarse sandy soil, which resulted in an annual leaching of 22–40 kg DOC ha−1 year−1 and 3–4 kg DON ha−1 year−1 in the sandy loam soil, and 174–310 kg DOC ha−1 year−1 and 10–31 kg DON ha−1 year−1 in the coarse sandy soil. It was shown that higher amounts of DOC were lost by leaching under the catch crops than from bare soil, that losses of DON were higher from bare soil than from soils with catch crops and that DON contributed significantly to the total N loss. Thus, DON needs to be taken into account in N-balance calculations.  相似文献

The heat-shock consensus element (HSE), CTNGAANNTTCNAG, is found in multiple copies upstream of all heat-shock genes. Here, the sequence requirements for heat-shock induction are tested by Drosophila germline transformation with an hsp70-lacZ gene fused to a pair of synthetic HSEs. Certain single-base substitutions in either HSE cause a dramatic reduction (forty-fold) in expression. Surprisingly, variations in sequences immediately flanking the HSEs also reduced levels of induction. One such variant that contains two perfect 14-base pair HSEs, which are correctly spaced relative to each other and the TATA box, retained only 7% of wild type-induced expression. These and additional analyses indicate that the heat-shock regulatory element includes sequences beyond the 14-base pair HSE and may be better described as a dimer of a 10-base pair sequence, NTTCNNGAAN.  相似文献

How proteins bind carbohydrates: lessons from legume lectins   总被引:2,自引:0,他引:2  
The pioneering studies of Irvin Liener on soybean agglutinin (SBA) in the early 1950s served as the starting point of our involvement in lectin research during the past four decades. Initially we characterized SBA extensively as a glycoprotein and showed that its covalently linked glycan is an oligomannoside commonly present in animal glycoproteins. We have also introduced the use of the lectin to the study of normal and malignant cells and to the purging of bone marrow for transplantation. Our recent work focuses on the combining site of Erythrina corallodendron lectin, closely related to SBA. In this legume lectin, as in essentially all other members of the same protein family, irrespective of their sugar specificity, interactions with a constellation of three invariant residues (aspartic acid, asparagine, and an aromatic residue) are essential for ligand binding. Lectins from other families, whether of plants or animals, also combine with carbohydrates by H-bonds and hydrophobic interactions, but the amino acids involved may differ even if the specificity of the lectins is the same. Therefore, nature finds diverse solutions for the design of binding sites for structurally similar ligands, such as mono- or oligosaccharides. This diversity strongly suggests that lectins are products of convergent evolution.  相似文献

Leaching of dissolved inorganic N (DIN) and dissolved organic N (DON) is a considerable loss pathway in grassland soils. We investigated the white clover (Trifolium repens) contribution to N transport and temporal N dynamics in soil solution under a pure stand of white clover and white clover-ryegrass (Lolium perenne) mixed stand. The temporal white clover contribution to N leaching was analysed by 15N incorporation into DIN and DON in percolating soil solution collected at 25-cm depth following white clover 15N leaf labelling that was applied at different times during the growing season. The white clover contribution to N transport in the soil profile was investigated over 2 years by analysing 15N in DIN and DON in percolating soil solution collected at 25-, 45- and 80-cm depth following 15N leaf labelling of white clover. The results showed that clover was a source of both DIN and DON. White clover autumn deposition contributed the most to N leaching. The leaching of DIN from the white clover in pure stand exceeded that of the mixed stand and confirmed that leaching of DIN is a function of N loadings and N demand. The DON leaching was unaffected by the presence of a companion grass, suggesting that the DON leaching from our grassland derived from the lysis of soil microbial biomass living on recent white clover deposits. White clover contributed to the leaching of DIN and DON at all depths, and the fact that the contents of DI 15N and DO 15N did not change with depth indicated that surplus of DIN and DON, formed in the uppermost soil layer, was transported in the soil profile.  相似文献
