The mammalian sperm membrane undergoes cholesterol efflux during maturation and fertilization. Although ATP‐binding cassette (ABC) transporters are known to transport cholesterol through cell membranes in other organs, their presence in canine testis, epididymis and sperm has not been proven to date. Hence, the aim of the present study was to localize the ABC transporters ABCA1 and ABCG1 in canine testicular and epididymidal tissue as well as in spermatozoa membranes. To this end, semen samples from 12 dogs as well as testicles and epididymides of four young and healthy dogs were prepared for immunohistochemistry, respectively. Capacitation and acrosome reaction (AR) were induced in aliquots of the semen samples before immunostaining to assess changes in the expression of ABCA1 and ABCG1. Evaluation by confocal microscopy revealed the presence of both ABCA1 and ABCG1 in canine testicles and of ABCA1 in the epididymides. In spermatozoa, only ABCA1 immunoreactivity was detected, mainly in the region of the acrosome and midpiece. After induction of capacitation, ABCA1 signal persisted in the acrosome but disappeared after AR, indicating a loss of ABCA1 with the loss of the acrosome. We conclude that ABCA1 and ABCG1 are expressed in canine testis, whereas only ABCA1 is expressed in epididymis and spermatozoa membrane, both transporters probably contributing to the regulation of membrane cholesterol content.  相似文献

A summary analysis for a new drug was required after five early phase veterinary clinical trials had been carried out. Such a meta-analysis of clinical trials involving repeated measurements would appear to be straightforward. However, this is not necessarily the case since different problems can arise. The choice of model is important since it may fit the data closely but have unwanted properties. As well, the trials to be regrouped may not have originally been designed to be combined. Hence, this can lead to various additional problems such as deciding on a time origin, analyzingan unbalanced design, and discovering confounded or collinear variables. Most of these problems were encountered in the five veterinary studies. These will be discussed and solutions suggested.  相似文献

African wild dogs are endangered, and in South Africa as elsewhere, they inhabit a fraction of their former range. In this study, we assessed the potential for economic benefits derived from ecotourism to offset the costs of three wild dog conservation options using a contingent valuation study of the willingness of visitors to four protected areas to pay to see wild dogs at the den - within a viable population in a large protected area (Kruger National Park), through reintroduction into nature reserves, and through the conservation of wild dogs occurring on ranchland in situ. Results indicated that tourism revenue from wild dogs in large protected areas is more than sufficient to offset the costs and could potentially be used to subsidise wild dog reintroductions or the conservation of wild dogs in situ on ranchland. On ranchland and for reintroductions, tourism revenue was generally predicted to offset most of the costs of conserving wild dogs where predation costs are low, and to exceed the costs where willingness to pay is high, and/or where the costs of predation by wild dogs are zero. Conservation efforts should facilitate the derivation of eco-tourism-related benefits from wild dogs on ranchland and in private reserves to create incentives for wild dog conservation. Ecotourism should be part of a multifaceted approach to wild dog conservation which also includes education and awareness campaigns, and efforts to encourage landowners to cooperate to form conservancies.  相似文献

ObjectiveTo investigate changes in colloid osmotic pressure (COP), total protein (TP) and osmolality (OSM) during anesthesia in horses given intravenous lactated Ringer’s solution (LRS) or LRS and hetastarch (HES).Study designProspective, clinical trial.AnimalsFourteen horses presented for surgery. Mean age 8.3 ± 1.9 years; mean weight 452 ± 25 kg.MethodsHorses were premedicated with xylazine intravenously (IV); anesthesia was induced with ketamine and diazepam IV, and maintained with sevoflurane. Butorphanol was administered IV with pre-medications or immediately after induction. Xylazine was administered IV for recovery if necessary. LRS was administered IV to all horses with a target rate of 5–10 mL kg?1 hour?1. Half of the horses also received 6% HES, 2.5 mL kg?1 over 1 hour in addition to LRS. Horses that received LRS only were considered the LRS group. Horses that received both LRS and HES were considered the LRS/HES group. Blood was drawn pre- and post-anesthesia, immediately following induction, and every 30 minutes throughout anesthesia. COP, TP and OSM were measured.ResultsCOP and TP significantly decreased at similar rates for both treatment groups from pre-anesthetic values. Pre-anesthetic COP was significantly greater in the LRS group when compared to the LRS/HES group pre-, post- and throughout anesthesia. In the LRS group post-anesthetic OSM was significantly different than the pre-anesthesia value and that for the LRS/HES group.Conclusions and clinical relevanceAdministration of IV HES (2.5 mL kg?1, over 1 hour) in combination with LRS does not attenuate the decrease in COP typically seen during anesthesia with crystalloid administration alone. Based on these results, administration of HES at this rate and total volume would not be expected to prevent fluid shifts into the interstitium through its effects on COP.  相似文献

Objective To evaluate the effects of a constant rate infusion (CRI) of romifidine on the requirement of isoflurane, cardiovascular performance and recovery in anaesthetized horses undergoing arthroscopic surgery. Study design Randomized blinded prospective clinical trial. Animals Thirty horses scheduled for routine arthroscopy. Methods After premedication (acepromazine 0.02 mg kg?1, romifidine 80 μg kg?1, methadone 0.1 mg kg?1) and induction (midazolam 0.06 mg kg?1 ketamine 2.2 mg kg?1), anaesthesia was maintained with isoflurane in oxygen. Horses were assigned randomly to receive a CRI of saline (group S) or 40 μg kg?1 hour?1 romifidine (group R). The influences of time and treatment on anaesthetic and cardiovascular parameters were evaluated using an analysis of variance. Body weight (t‐test), duration of anaesthesia (t‐test) and recovery score (Wilcoxon Rank Sum Test) were compared between groups. Significance was set at p < 0.05. Results All but one horse were positioned in the dorsal recumbent position and ventilated from the start of anaesthesia. End tidal isoflurane concentrations were similar in both groups at similar time points and over the whole anaesthetic period. Cardiac output was significantly lower in horses of the R group, but there were no significant differences between groups in cardiac index, body weight or age. All other cardiovascular parameters were similar in both groups. Quality of recovery did not differ significantly between groups, but more horses in group R stood without ataxia at the first attempt. One horse from group S had a problematic recovery. Conclusions and clinical relevance No inhalation anaesthetic sparing effect or side effects were observed by using a 40 μg kg?1 hour?1 romifidine CRI in isoflurane anaesthetized horses under clinical conditions. Cardiovascular performance remained acceptable. Further studies are needed to identify the effective dose of romifidine that will induce an inhalation anaesthetic sparing effect in anaesthetized horses.  相似文献
