Drought-tolerant maize genotypes (Huffman, Z08-004, Tuxpan, PH 9, NRC 5348, Chunco, Saint Croix, and Arizona) were compared in the field and laboratory to toxin-resistant GT-MAS:gk and Yellow Creole. SDS-PAGE, scanning electron microscopy of kernel cuticle, amount of kernel wax, Aspergillus flavus kernel colonization, Aspergillus ear rot, insect damage, aflatoxin production, and their relationships were examined. SDS-PAGE showed the presence of a 14 kDa trypsin inhibitor in the kernels of all genotypes except Chunco, which contains a protein of a larger molecular weight. The 14 kDa trypsin inhibitor protein content in these genotypes was higher than in GT-MAS:gk and Yellow Creole. Scanning electron microscopy revealed that Arizona, Huffman, and Chunco genotypes had abundant wax deposits on kernel surfaces and the amount of pericarp wax was equal to or above that from GT-MAS:gk and Yellow Creole. Differences in Aspergillus ear rot ratings, fungal colonization, and insect damage by corn earworm were observed in all drought-tolerant maize genotypes as well as in the controls. Kernel screening assays showed that aflatoxin B(1) levels in inoculated drought-tolerant genotypes differed significantly from those in GT-MAS:gk and Yellow Creole (LSD = 576). Aflatoxin B(1) levels in the inoculated genotypes differed significantly from those of GT-MAS:gk or Yellow Creole (LSD = 1389) when grown under drought stress conditions. Pearson correlation coefficients were significant between ear rot ratings and insect damage (r = 0.75; P = 0.01) and between Aspergillus ear rot and aflatoxin levels (r = 0.54; P = 0.05). On the basis of the parameters studied, there are indications that these genotypes were potential sources of A. flavus resistance.  相似文献

It has been reported that soybean peptide fractions isolated from Korean fermented soybean paste exert angiotensin I converting enzyme (ACE) inhibitory activity in vitro. In this study, further purification and identification of the most active fraction inhibiting ACE activity were performed, and its antihypertensive activity in vivo was confirmed. Subsequently, a novel ACE inhibitory peptide was isolated by preparative HPLC. The amino acid sequence of the isolated peptide was identified as His-His-Leu (HHL) by Edman degradation. The IC(50) value of the HHL for ACE activity was 2.2 microg/mL in vitro. Moreover, the synthetic tripeptide HHL (spHHL) resulted in a significant decrease of ACE activity in the aorta and led to lowered systolic blood pressure (SBP) in spontaneously hypertensive (SH) rats compared to control. Triple injections of spHHL, 5 mg/kg of body weight/injection resulted in a significant decrease of SBP by 61 mmHg (p < 0.01) after the third injection. These results demonstrated that the ACE inhibitory peptide HHL derived from Korean fermented soybean paste exerted antihypertensive activity in vivo.  相似文献

The conformation of oat globulin dispersions (10% in D2O) under the influence of pH, chaotropic salts, protein structure perturbants, and heating conditions was studied by Fourier transform infrared (FTIR) spectroscopy. The FTIR spectrum of oat globulin showed major bands from 1670 to 1634 cm(-1), corresponding to the four major types of secondary structures, that is, beta-turns, beta-sheets, alpha-helices, and random coils. At extreme acidic and alkaline pH conditions, there were changes in intensity in the bands attributed to beta-sheet structures (1626, 1634, and 1682 cm(-1)), and shifts of the bands to higher or lower wavenumbers, indicating changes in conformation. In the presence of some chaotropic salts, the 1626 and 1634 cm(-1) bands were shifted upward, with a marked decrease in the intensity of the 1634 cm(-1) peak. The addition of several protein structure perturbants led to a slight shift in the alpha-helix/random coil bands and a marked reduction in the beta-sheet peaks, suggesting protein unfolding. Heating under aggregating conditions led to slight shifts in all of the major bands and progressive changes in the intensity of the alpha-helix, beta-sheet, and beta-turn peaks, suggesting protein denaturation. This was accompanied by marked increases in intensity of the two intermolecular beta-sheet bands (1682 and 1624-1626 cm(-1)) associated with the formation of aggregated strands. The IR spectra of soluble and insoluble aggregates showed a redistribution of native and extensively denatured proteins in the two fractions.  相似文献

Seeds of 49 accessions of corn (Zea mays ssp. mays), 9 accessions of teosinte (Zea species that are thought to be ancestors and probable progenitors to corn), and 3 accessions of Job's tears (Coix lacryma), obtained from a germplasm repository, were ground and extracted with hexane. Whole kernel oil yields and levels of four phytonutrients (free phytosterols, fatty acyl phytosterol esters, ferulate phytosterol esters, and gamma-tocopherol) in the oils were measured. Among the seeds tested, oil yields ranged from 2.19 to 4.83 wt %, the levels of ferulate phytosterol esters in the oil ranged from 0.047 to 0.839 wt %, the levels of free phytosterols in the oil ranged from 0.54 to 1.28 wt %, the levels of phytosterol fatty acyl esters in the oil ranged from 0.76 to 3.09 wt %, the levels of total phytosterols in the oil ranged from 1.40 to 4.38 wt %, and the levels of gamma-tocopherol in the oil ranged from 0.023 to 0.127 wt %. In general, higher levels of all three phytosterol classes were observed in seed oils from accessions of Zea mays ssp. mays than in seed oils from accessions of the other taxonomic groups. The highest levels of gamma-tocopherol were observed in teosinte accessions.  相似文献

Polygodial and (2E)-hexenal were found to possess antibacterial activity against Salmonella choleraesuis with the minimum bactericidal concentrations (MBC) of 50 microg/mL (0.17 mM) and 100 microg/mL (0.98 mM), respectively. The time kill curve study showed that these two alpha,beta-unsaturated aldehydes were bactericidal against this food-borne bacterium at any stage of growth. However, they showed different effects on the growth of S. choleraesuis. The combination of polygodial and anethole exhibited strong synergism on their bacteriostatic action but only marginal synergism on their bactericidal action.  相似文献

 Soybean cultivars capable of nodulating with indigenous Bradyrhizobium spp. have been developed by the International Institute of Tropical Agriculture (IITA) and national programs in Africa in order to avoid artificial inoculation by resource-poor farmers in Africa. The current selection procedure for enhanced N2 fixation is based on an assessment of nodule formation which does not directly quantify the proportions of crop N derived from the atmosphere. We have monitored N accumulation patterns and N2 fixation in nine promiscuous soybean cultivars with different maturity periods, using the 15N dilution technique. Nodule development generally peaked at the early podfill stage for all cultivars except Tgx 1519-1D and Tgx 1447-2D in which it continued to increase. The proportion of crop N derived from fixation (%NDFA) ranged between 51% and 67%, 77% and 84%, and 66% and 73% at full bloom, early podfill, and physiological maturity stages, respectively. Total N accumulation increased in all soybean genotypes with increasing plant age. Significant correlations (P<0.001) were established between nodule weight and %NDFA, even though this did not explain the relationship between nodule development and N2 fixation in cultivars such as Tgx 1519-1D. Promiscuous soybean cultivars retained between 10% and 19% of total N accumulated at the final harvest, in belowground biomass. Our results indicated that these soybean cultivars can derive substantial proportions of plant N from N2 fixation in soils where compatible indigenous bradyrhizobia populations are adequate and effective. Also, we have substantiated the claims that qualitative nodulation parameters currently used to select varieties with a high N2 fixation capacity need to be validated with other measurements of N2 fixation.  The effects of salt type and its concentration on nitrification, N mineralization and N2O emission were examined under two levels of moisture content in Yellow soil and Andosol samples as simulated to agriculture under arid/semi-arid conditions and under heavy application of fertilizer in a glass-house, respectively. The salt mixtures were composed of chlorides (NaCl and NH4Cl) or sulphates [Na2SO4 and (NH4)2SO4] and were added at various concentrations (0, 0.1, 0.2, 0.4 and 0.6 M as in the soil solution). These salts were added to non-saline Yellow soil at different moisture contents (45 or 40 and 65% of maximum water-holding capacity; WHC) and their effects on the changes in mineral N (NH4 +-N and NO3 -N) concentration as well as N2O emission were examined periodically during laboratory incubation. We also measured urease activities to know the effect of salts on N mineralization. Furthermore, Ca(NO3)2 solution was added at various concentrations (0, 0.1, 0.3, 0.5 and 0.8 M as in the soil solution) to a non-saline Andosol taken from the subsurface layer in a glass-house and incubated at different moisture contents (50% and 70% of WHC) to examine their effects on changes in mineral N. Nitrification was inhibited by high, but remained unaffected by low, salt concentrations. These phenomena were shown in both the model experiments. It was considered that the salinity level for inhibition of nitrification was an electric conductivity (1 : 5) of 1 dS m–1. This level was independent of the type of salts or soil, and was not affected by soil moisture content. The critical level of salts for urease activities was about 2 dS m–1. The emission rate of N2O was maximum at the beginning of the incubation period and stabilized at a low level after an initial peak. There was no significant difference in N2O emission among the treatments at different salt concentrations, while higher moisture level enhanced N2O emission remarkably. Vertisol soils of central India are heavy in texture, with high clay content and low organic matter. These soils are prone to degradation and the soil loss is due to poor management practices including excessive tillage. Based on a long-term study conducted for improving the quality of these soils, it was found that management practice such as low tillage (LT) + 4 t ha?1 compost + herbicide (Hb) recorded significantly higher organic carbon (OC) (6.22 g kg?1) and available N (188.5 kg ha?1) compared to conventional tillage (CT) + recommended fertilizer (RF) + off-season tillage (OT) + hand weeding (HW) (OC: 4.71 g kg?1, available nitrogen (N) (159.3 kg ha?1). Among the physical soil quality parameters, mean weight diameter (MWD) was significantly higher under LT + 4 t ha?1 straw + Hb (0.59 mm). The practice of LT + 4 t ha?1 straw + HW recorded significantly higher microbial biomass carbon (MBC) (388.8 μg g?1). The order of key indicators and their contribution towards soil quality was as follows: OC (29%) >, MBC (27%) > available zinc (Zn) (22%) > MWD (9%) > available boron (B) (8%), > dehydrogenase activity (DHA) (5%). The order of the best treatment which maintained soil quality index (SQI) values reasonably good (>1.5) was as follows: LT + 4t ha?1 compost + HW (1.65) > LT + 4 t ha?1 compost +Hb (1.60) > LT + 4t ha?1 straw + HW (1.50). Hence, these treatments could be recommended to the farmers for maintaining higher soil quality in Vertisols under soybean system. Correlation studies revealed stronger relationship between key indicators like OC (R2 = 0.627), MBC (R2 = 0.884), available Zn (R2 = 0.739) and DHA (R2 = 0.604) with Relative Soil Quality Index (RSQI). A field experiment was conducted to evaluate nine genotypes of sorghum grown for two consecutive cropping seasons in 2006 and 2007 under rain-fed conditions in a semiarid zone at the Research Farm of El Fasher Research Station, Sudan. The objective of the study was to assess genetic variability and heritability among sorghum genotypes using phenotypic morphological traits. A randomized complete block design with four replications was used for the experiment. The grain yield (kg ha?1), the number of heads/plant, followed by straw yield (kg ha?1) had the highest genotypic coefficients of variation in both seasons. High heritability (above 95%) was shown for plant height in both seasons. High genetic advance was reported in straw yield, 1000-grain weight and days to flowering in both seasons. Highly significant differences among genotypes were found for all characters. The high-yielding genotype was Adv-Edo-CWS (E-9) with grain yields of 2780.6 kg ha?1. Grain yield was significantly and positively correlated with straw yield (r = 0.91), number of heads/plant (r = 0.69), plant height (r = 0.53) and 1000-grain weight (r = 0.36). However, it was significantly and negatively correlated with days to 50% flowering (r = ?0.21). Based on their positive association with grain yield, the character’s straw weight, number of heads/plant, plant height and 1000-grain weight would be the preferable selection criteria for sorghum improvement program in the country.  相似文献

Lemon balm (Melissa officinalis L.) is used since ancient times because of its sedative, spasmolytic and antiviral effects. Its therapeutic impact is due to the content of essential oil and rosmarinic acid. A set of 68 M. officinalis genotypes was evaluated for content and composition of essential oil and the content of rosmarinic acid. For all genotypes the level of ploidy was determined. The 68 genotypes were clone plants grown and evaluated for two years at Quedlinburg. For analysis of secondary metabolites distillation, gas chromatography and high performance liquid chromatography was used. The content of essential oil varied in this study in ranges from 0.03 to 0.33% for the second cut 2010 and 0.01–0.35% for the second cut 2011. The rosmarinic acid content ranged in the year 2010 from 3.67 to 7.55% and in the year 2011 from 4.92 to 8.07%. Via statistical analyses two chemotypes of essential oil were found: chemotype citral and chemotype β-caryophyllene oxide. Ploidy was determined for all genotypes and two cytotypes were found: diploid 2n = 2x = 32 (62 of 68 genotypes) and triploid 2n = 3x = 48 (6 of 68 genotypes).  相似文献
