Immunoglobulin K genes are constructed during lymphocyte differentiation by the joining of two DNA elements, VK and JK, to form both a VKJK coding unit and a reciprocal recombination product. The two products formed in single VK-to-JK joining events can be directly isolated through the use of a retrovirally introduced recombination substrate. The structural analysis of a number of recombinants and the derivation of secondary recombination products define some of the basic features of the mechanism of immunoglobulin gene assembly. 相似文献

Fluxes of biogenic carbonates moving out of the euphotic zone and into deeper undersaturated waters of the North Pacific were estimated with free-drifting sediment traps. Short-duration (1 to 1.5 day) sampling between 100 and 2200 meters points to a major involvement in the oceanic carbonate system by a class of organisms which had been relegated to a secondary role-aragonitic pteropods. Pteropod fluxes through the base of the euphotic zone are almost large enough to balance the alkalinity budget for the Pacific Ocean. Dissolution experiments with freshly collected materials shed considerable light on a mystery surrounding these labile organisms: although plankton collections from net tows almost always contain large numbers of pteropods, these organisms are never a major component of biogenic materials in long-duration sediment trap collections. Their low abundance in long-duration collections results from dissolution subsequent to collection. Shortduration sampling showed significant increases in the ratio of calcitic foraminifera to aragonitic pteropods in undersaturated waters, indicating the more stable mineralogic form, calcite, was preserved relative to aragonite. Approximately 90 percent of the aragonite flux is remineralized in the upper 2.2 kilometers of the water column. 相似文献

The sequence of the Drosophila melanogaster genome presented in this issue of Science is the latest milestone in nine decades of research on this organism. Genetic and physical mapping, whole-genome mutational screens, and functional alteration of the genome by gene transfer were pioneered in metazoans with the use of this small fruit fly. The calcium-activated adenosine triphosphatase activity of cultured single beating heart cells of the rat was localized and visualized by incubating the cells, after extraction in a mixture of glycerol and water, with adenosine triphosphate and calcium. The calcium phosphate, precipitated at the site of the enzyme reaction, was converted into a visible precipitate. The enzyme activity appeared in bands perpendicular to the myofibrils. 相似文献

A carbon-chromite fraction from the Allende C3V chondrite shows strikingly large isotopic enrichments of neodymium-142 (0.47 percent) and neodymium- 143 (36 percent). Both apparently formed by alpha decay of samarium-146 and samarium-147 (half-lives 1.03 x 10(8) and 1.06 x 10(11) years), but the isotopic enrichment was greatly magnified by recoil of residual nuclei into a carbon film surrounding the samarium-bearing grains. These data provide an improved estimate of the original abundance of extinct samarium-146 in the early solar system [(146)Sm/(144)Sm = (4.5 +/- 0.5) x 10(-3)], higher than predicted by some models of pprocess nucleosynthesis.
