Chloroplast microsatellites (SSRs) are conserved within wheat species, yet are sufficiently polymorphic between and within species to be useful for evolutionary studies. This study describes the relationships among a very large set of accessions of Triticum urartu Thum. ex Gandil., T. dicoccoides (Körn. ex Asch. et Graebn.) Schweinf., T. dicoccon Schrank, T. durum Desf., T. spelta L., and T. aestivum L. s. str. based on their cpSSR genotypes. By characterising the chloroplast diversity in each wheat species in the evolutionary series, the impact on diversity of major evolutionary events such as domestication and polyploidyisation was assessed. We detected bottlenecks associated with domestication, polyploidisation and selection, yet these constrictions were partially offset by mutations in the chloroplast SSR loci that generated new alleles. The discrete cpSSR alleles and haplotypes observed in T. urartu and Aegilops tauschii, combined with other species specific polymorphisms, provide very strong evidence that concur with current opinion that neither species was the maternal and thus cytoplasmic donor for polyploid wheats. Synthetic hexaploid wheats possessed the same chloroplast haplotypes as their tetraploid progenitors demonstrating how the novel synthetic wheat lines have captured chloroplast diversity from the maternal parents, the chloroplast is maternally inherited and novel alleles are not created by genomic rearrangements triggered by the polyploidisation event. 相似文献

- Identification of the geographic extent of population boundaries, the distribution of genetic lineages, and the amount of genetic exchange among breeding groups is needed for effective conservation of vulnerable marine migratory species. This is particularly true of the flatback turtle (Natator depressus), which only breeds in Australia but has extensive migrations that can include international waters.
- This study investigated the phylogeography and genetic structure among 17 flatback turtle rookeries across their range by sequencing an 810 bp portion of the mitochondrial DNA in 889 samples and genotyping 10 microsatellite loci in 598 samples.
- There was low phylogenetic divergence among haplotypes and evidence of recent population expansion, likely in the late Pleistocene. A predominant haplotype was found across all rookeries, but other haplotype groups were regionally specific.
- In general, there was agreement in patterns of genetic differentiation in the mitochondrial DNA and microsatellite data, and in some pairwise comparisons a higher mutation rate of microsatellites provided stronger evidence of differentiation.
- These results suggest natal philopatry operates in the choice of breeding locations for males as well as females.
- Evidence of genetic connectivity among neighbouring rookeries led to the identification of seven genetic stocks. Geographic boundaries of rookeries used by genetic stocks varied widely (160–1,300 km), highlighting a need for field studies to better understand movement patterns.
- Hierarchical analysis of molecular variance identified significant genetic differentiation based upon genetic stock, nesting phenology (summer vs. winter nesters), and a west–east discontinuity across Torres Strait. A pattern of isolation by distance was identified, which was most strongly observed in the microsatellite data.
- In combination with tagging and telemetry studies, these results will allow better quantification of stock‐specific threats along migratory routes and in foraging habitats. Implications of climate change will be stock specific and may depend upon the extent of genetic connectivity between neighbouring stocks.

This review focuses on relevant scientific information regarding the current knowledge of the yellow head complex viruses, yellow head virus and gill‐associated virus. The yellow head complex viruses have been problematic within the aquaculture industry for over 10 years and still retain their research topicality. Presently, there are numerous research papers from different journals covering the identification, disease expression and spread, pathogenesis, detection, morphology, genomic sequence and protein profiles of the yellow head complex viruses. Indeed, there has been no extensive review to compare these studies, and as a corollary, to assess flaws in contemporary research and knowledge. Additionally, the yellow head complex viruses rank within the top four prawn viruses with respect to disease impact and economic loss. This review collectively reports on all the findings and current methods of research and aims to identify weak areas of research where conclusions have been unjustifiably drawn and furthermore to elucidate areas that have a gap of knowledge. 相似文献

Development scholars and practitioners are promoting food security, food sovereignty, and the localization of food systems to prepare for the projected negative impacts of climate change. The implementation of biodiverse homegardens is often seen as a way not only to localize food production but also as a strategy in alignment with a food sovereignty agenda. While much scholarship has characterized and critiqued food security and sovereignty conceptualizations, relatively little research has examined people’s lived experiences in order to test how such theoretical visions play out on the ground in farming communities. Based on a case study of four coffee cooperatives in northern Nicaragua, we examine a non-governmental organization (NGO)-supported project promoting food security and sovereignty through development of homegardens. We ask: To what extent are homegardens an effective strategy to reach food sovereignty? And, why may farmers be resistant to changing their food production and consumption strategies to embrace biodiverse homegardens when they improve food security? We discuss characteristics of agroecological homegardens, the distinctions between food security, food sovereignty and dominant discourses of development, the history of food sovereignty in Nicaragua, and farmer perspectives on homegarden implementation. Despite historic critiques, NGOs are poised to facilitate the transformation of food and agricultural development by employing counter development strategies, a necessary step if homegardens are to be successful in the long term. To conclude, we propose some strategies NGOs and communities might pursue to move forward with homegardens as a food sovereignty strategy. This research suggests that a food sovereignty approach still rooted in mainstream development models faces significant obstacles to moving beyond food security and into a farmer-led food sovereignty agenda. 相似文献

The effects of 2 different continuous rate infusions (CRIs) of medetomidine over an 8-hour period on sedation score, selected cardiopulmonary parameters, and serum levels of medetomidine were evaluated in 6 healthy, conscious dogs using a crossover study design. The treatment groups were: CONTROL = saline bolus followed by saline CRI; MED1 = 2 μg/kg body weight (BW) medetomidine loading dose followed by 1 μg/kg BW per hour CRI; and MED2 = 4 μg/kg BW medetomidine loading dose followed by 2 μg/kg BW per hour CRI. Sedation score (SS), heart rate (HR), respiratory rate (RR), temperature (TEMP), systolic arterial pressure (SAP), mean arterial pressure (MAP), and diastolic arterial pressure (DAP), arterial and mixed venous blood gas analyses, lactate, and plasma levels of medetomidine were evaluated at baseline, at various intervals during the infusion, and 2 h after terminating the infusion. Statistical analysis involved a repeated measures linear model. Both infusion rates of medetomidine-induced dose-dependent increases in SS and dose-dependent decreases in HR, SAP, MAP, and DAP were measured. Respiratory rate (RR), TEMP, central venous pH, central venous oxygen tension, and oxygen extraction ratio also decreased significantly in the MED2 group at certain time points. Arterial oxygen and carbon dioxide tensions were not significantly affected by either infusion rate. In healthy dogs, both infusion rates of medetomidine-induced clinically relevant sedative effects, accompanied by typical alpha2 agonist-induced hemodynamic effects, which plateaued during the infusion and subsequently returned to baseline. While additional studies in unhealthy animals are required, the results presented here suggest that medetomidine infusions at the doses studied may be useful in canine patients requiring sedation for extended periods. 相似文献

Commercially available attenuated strains of Mycoplasma gallisepticum (MG) are commonly used within the layer industry to control MG-induced mycoplasmosis. Among these are two live MG vaccines derived from the moderately pathogenic MG "chick F" strain. In the present study, the commercially available F strain derivatives were compared for their ability to elicit seroconversion, persist in vivo, and protect against virulent MG-induced airsacculitis. In addition, a noncommercial laboratory-derived high-passage F strain isolate was included in the study. Commercial (Hy-Line W-36) layers were placed in biological isolation units at 9 wk of age (woa). At 10 woa, birds within each biological isolation unit were treated via eye-drop application with one of the three F strain-derived vaccines at one of four levels (1x, 10(-1)x, 10(-2)x, or 10(-3)x). For the commercially available F strain derivatives, 1x equaled the manufacturer's recommended dose. The 1x dose of the noncommercial laboratory-maintained F strain derivative equaled 20 microl of a 48 hr culture. For wk 1-6 postvaccination (p.v.), sera were collected weekly from each bird, and seroconversion was assessed via serum plate agglutination (SPA). Virulent MG (strain R(low)) challenge occurred via intratracheal inoculation at 7 wk p.v. Necropsies were subsequently performed to assess challenge-associated airsacculitus. For each F strain derivative applied at 1x and 10(-1)x, 100% seroconversion, as measured by SPA, was demonstrated by 6 wk p.v., and rates at the 10(-2)x dosage were 10% and 90% for the commercial vaccines and 60% for the laboratory-derived strain in this period. Following challenge, airsacculitis was observed in 66.67% of the nontreated controls but not in any 1x- or 10(-1)x-treated bird independent of applied F strain derivative. 相似文献

Tuberculosis (TB) is a significant animal health problem in many parts of the world, and reservoirs of infection in wild animals complicate disease control efforts in farmed livestock, particularly cattle. Badgers (Meles meles) are a significant wildlife reservoir of Mycobacterium bovis infection for cattle in the United Kingdom (UK) and Republic of Ireland (ROI). Vaccination of badgers using an M. bovis strain bacille Calmette-Guérin (BCG) vaccine could potentially be an option in the national TB eradication strategy. Wildlife vaccination has been used successfully for other diseases in wildlife species, and may have a role to play in reducing M. bovis transmission at the wildlife-livestock interface. Research to date has provided evidence that BCG is protective in badgers, and a parenteral badger BCG vaccine has been licensed in the UK. Further research is required to develop effective strategies for vaccine deployment and to determine the effect of badger vaccination on cattle TB incidence. 相似文献
