Transfer of a normal Chinese hamster X chromosome (carried in a mouse A9 donor cell line) to a nickel-transformed Chinese hamster cell line with an Xq chromosome deletion resulted in senescense of these previously immortal cells. At early passages of the A9/CX donor cells, the hamster X chromosome was highly active, inducing senescence in 100% of the colonies obtained after its transfer into the nickel-transformed cells. However, senescence was reduced to 50% when Chinese hamster X chromosomes were transferred from later passage A9 cells. Full senescing activity of the intact hamster X chromosome was restored by treatment of the donor mouse cells with 5-azacytidine, which induced demethylation of DNA. These results suggest that a senescence gene or genes, which may be located on the Chinese hamster X chromosome, can be regulated by DNA methylation, and that escape from senescence and possibly loss of tumor suppressor gene activity can occur by epigenetic mechanisms.  相似文献

Chitinous material was extracted from mycelia of Aspergillus niger and Mucor rouxii grown in yeast peptone dextrose broth for 15 and 21 days, respectively. The extracted material was characterized for purity, degree of acetylation, and crystallinity and tested for antibacterial and eliciting properties. The maximum glucosamine level determined in the mycelium of A. niger was 11.10% dw and in the mycelium of M. rouxii was 20.13% dw. On the basis of the stepwise extraction of freeze-dried mycelia, it appeared that M. rouxii mycelia contained both chitin and chitosan, whereas A. niger contained only chitin. The yields of crude chitin from A. niger and M. rouxii were 24.01 and 13.25%, respectively, and the yield of chitosan from M. rouxii was 12.49%. Significant amounts (7.42-39.81%) of glucan were associated with chitinous compounds from both species and could not be eliminated by the extraction method used. The degrees of acetylation were determined to be 76.53 and 50.07% for chitin from A. niger and M. rouxii, respectively, and 19.5% for M. rouxii chitosan. The crystallinity of fungal chitin and chitosan was estimated to be less intense than in corresponding materials from shrimp shells. The extracted chitin and chitosan in a concentration of 0.1% reduced Salmonella Typhimurium DT104 2576 counts by 0.5-1.5 logs during a 4 day incubation in tryptic soy broth at 25 degrees C. Furthermore, all tested chitinous materials from fungal sources significantly reduced lesions caused by Botrytis cinerea and Penicillium expansum in harvested apples.  相似文献

Abstract. Less Intensive Farming and Environment (LIFE) management is a form of integrated farming which aims to meet farming's economic and environmental requirements. We used a farm-scale LIFE demonstration to measure nitrogen (N) leaching losses over a 6 year period (1995–2001) using ceramic suction cups and a meteorological model to give estimates of drainage volumes. Losses from the system averaged 49 kg N ha−1, with an average drainage nitrate concentration of 15.5 mg N L−1. Rainfall and its distribution strongly influenced the loss, and drainage N concentration only fell below the nominal target of 11.3 mg N L−1 (the EU limit for potable water) in the two wettest seasons. Crop type did not have a significant effect on either postharvest mineral N (PHMN) in soil or the leaching loss in the subsequent winter. However PHMN and overwinter N leaching declined with increasing crop yield. Overwinter crop N uptake increased with early sowing: leaching loss was only 5 kg N ha−1 under grass sown in early September. Measurements of PHMN, crop sowing date and drainage data were used to construct simple equations to predict average drainage N concentration under various scenarios. The large N loss from our site is partially attributable to soil type (shallow over limestone), indeed on similar soil the loss from a conventional farm nearby was greater. The LIFE practices of postharvest harrowing and late cereal sowing will minimize the need for agrochemical use but they stimulate mineralization and reduce plant N uptake in autumn, leaving more N at risk to leaching. Some assessment of all environmental impacts is needed if the benefits of integrated practices such as those used in LIFE are to be quantified.  相似文献

The speciation of several tributyltin (TBT) compounds used in marine antifouling paints was studied by tin Mössbauer spectroscopy in both spiked anoxic and oxic estuarine sediments. These compounds included TBT acetate, chloride, fluoride, and oxide. To assist in the determination of the products, possible speciation products were also examined: TBT carbonate, sulfate, and sulfide. The study indicated that TBTF remains in its polymeric form after interaction with both types of sediments. TBT sulfate is unaltered in the spiked sediments. The data reported in this paper support the hypothesis that the speciation of the other TBT compounds examined in spiked sediments depends upon the nature of the sediment.  相似文献

In urban areas, the pattern of trees is often a result of municipal policy, built form, neighborhood socioeconomic conditions, and the actions of local actors. Recent research has focused on the role of neighborhood socioeconomics, and begun to explore the underlying causes of uneven distributions of urban forests associated with different socioeconomic groups. To date, little work has explored property-level tree conditions in relation to disaggregated household characteristics and actions, yet the household is the scale where most decisions about residential tree planting and care are made. This study examines the role of property-level built conditions, household socioeconomics, and residents’ actions and attitudes in relation to property-level canopy cover and tree density. The study area is four neighborhoods in the City of Mississauga (ON, Canada). Regression analyses were conducted to explore significant variables related to the two tree measures for all properties together and separately by neighborhood. The results indicate that property conditions and residents actions are more important in relation to tree variations than socioeconomic factors. Additionally, several significant factors have opposite relationships with percent canopy cover and tree density. These results highlight the need to consider property-level built conditions, residents’ actions, and multiple measures of the urban forest to better understand the patterns of trees in cities.  相似文献

The development of pepper transplants is sensitive to the prevailing temperature in the field. High temperatures immediately after transplanting pepper seedlings delay their development in the field. During nine days of post-transplanting heat stress (30/25°C or 35/25°C, day/night temperature regimes), root and shoot growth were inhibited compared to the control transplants (25/18°C) with a significant decline in total protein and soluble acid invertase activity in the roots of the heat stressed plants. Acid invertase activity was less sensitive to the heat stress in the leaves than in the roots. A significant reduction in the root invertase activity occurred as early as two days after the beginning of the stress. Transplanting caused a 72% inhibition in the carbon allocation to the roots, compared with the non-transplanted seedlings. Exposure to heat after transplanting did not further reduce carbon allocation to the roots. The protein profile on SDS-PAGE from leaves shows that during the heat stress, some heat related proteins were synthesized concomitant with the reduction in the synthesis of constitutive proteins. In the roots, the synthesis of heat related proteins was barely detectable with no visible changes in the protein profile. The results suggest that roots are more sensitive to heat stress than leaves, possibly because they are less able to synthesize heat related proteins. The causal relationship between protein synthesis during heat stress and the performance of the transplants is not well understood.  相似文献

SUMMARY

Heat stress causes abscission of flowers in pepper plants and thus reduces yield. In the present study, we investigated the involvement of ethylene in heat-stress related flower abscission, by comparing the response of flowers of bell pepper, cv. Maor, known to be sensitive to flower abscission, and flowers of paprika, cv. Lehava, a resistant cultivar of Capsicum annuum. Their differing susceptibilities to high temperatures depend on light. Under high-temperature, high-light conditions, bell pepper was less susceptible than paprika, but, under high-temperature low-light, bell pepper was more susceptible. At high temperatures, flower explant abscission was much higher with bell pepper than paprika. Ethylene production (EP) by bell pepper explants reached a maximum at 34°C and decreased at higher temperatures (42 and 48 C), while in paprika EP rates were lower and reached the maximum at 42°C. Explants of bell pepper flowers were more susceptible to exogenous ethephon than paprika flowers. The sensitivity of a collection of pepper cul-tivars to heat induced flower abscission was more closely correlated with their flower sensitivity to ethephon than with the flower EP rate. We suggest that the differential susceptibility of bell pepper and paprika to heat stress is a result of both different ethylene production by their flowers and their differing sensitivities to ethylene produced under high-temperature stress. However, the sensitivity of the flower to ethylene may be more important in inducing their abscission.  相似文献

To further our understanding of invasive species?? novel distributions, knowledge of invasive species?? relationships with environmental variables at multiple spatial scales is paramount. Here, we investigate which environmental variables and which spatial scales best explain the invasive mute swan??s (Cygnus olor) distribution in southern Ontario (Canada). Specifically we model mute swan distribution changes according to ecologically-relevant spatial scales: average territory size radius, 140?m; median dispersal distance of cygnets, 3,000?m; and average activity distance of males, 8,000?m. For individual spatial scales, global models using variables measured at each particular scale result in the highest Akaike weights, AUC, and Cohen??s Kappa values. Yet composite models (models combining variables measured at different scales) elicit the best models, as determined by higher Akaike weights and high AUC and Cohen??s Kappa values. Overall, percent water, waterbody perimeter density, temperature, precipitation, and road density are positively correlated with mute swan distribution, while percent forest and elevation are negatively correlated at all scales of analysis. Only percent water and annual precipitation are more influential in determining mute swan distribution at the 3,000 and 8,000?m zone scales than the territory scale. While most species distribution models are performed at a single scale, the results of our study suggest that composite models reflecting a species?? ecological needs provide models of better fit with similar, if not better, predictive accuracy. When analyzing species distributions, we also recommend that ecologists consider the scale of the underlying landscape processes and the effect that this may have on their modelling outcomes.  相似文献
