Quantitation of o- and p-sulfamoylbenzoic acid residues in saccharin and its sodium salt is achieved by a method comprising methanolic extraction and high-performance ion exchange chromatography. A commercially available anion exchange column was employed with an aqueous buffered (pH 9.2) mobile phase. As little as 80 ppm of the ortho-isomer and 25 ppm of the para-isomer can be accurately determined. The levels of detectability (2 times noise) are estimated as 8 ppm (0.16 mug on column) and 2.5 ppm (0.05 mug on column), respectively. Recoveries from saccharin ranged from 92.7 to 96.5% (ortho) and from 92.2 to 103.3% (para). Recoveries from the sodium salt ranged from 93.1 to 104.4% (ortho) and from 93.5 to 97.8% (para). Of 9 other potential saccharin impurities tested separately, only one was found to interfere slightly in the chromatographic part of the procedure.  相似文献

Rosa multiflora Thunb. (Rosaceae), an invasive plant that currently infests millions of hectares in the eastern half of the USA, was planted initially in the 1940s as a “living fence”, cover for game animals and for erosion control. The larvae of Megastigmus aculeatus var. nigroflavus Hoffmeyer (Hymenoptera: Torymidae) feed on the developing R. multiflora seeds and have the potential to reduce the seed output of R. multiflora. Rosehips were collected from 49 sites across eastern and southern Iowa, USA, to determine the presence and distribution of M. aculeatus, the larvae of which were found in 266 of the 979 (27%) rosehips that were dissected and at 31 of the 49 sites (63%) that were sampled. The levels of M. aculeatus infestation over 2–3 years were determined at three selected sites and it was documented that more seeds were aborted than were viable or contained wasp larvae. A negative relationship was observed between the numbers of both the aborted and the viable seeds and the probability of a rosehip being attacked by M. aculeatus, indicating that M. aculeatus females are not preferentially selecting rosehips with higher numbers of viable or aborted seeds for oviposition. There is a significant reduction in both the viable seeds and the aborted seeds in hips that have been attacked by M. aculeatus. Detailed knowledge of R. multiflora demography is necessary to determine the level of seed predation that is required to reduce the recruitment of new individuals into the population.  相似文献

Assemblages of plants were studied at 14 sites in northern Patagonia corresponding to localities at which we (Monjeau et al. 1997) earlier studied the relationship between small mammal assemblages and landscape classifications. This allowed us to test predictions that both plants and small mammals correspond to the more inclusive hierarchical landscape divisions but that plants track better than small mammals the less inclusive divisions. Species presence or absence of plants at each locality was used in a series of multivariate analyses and compared by correlation analysis with those generated from small mammal species data. Assemblages of both plants and small mammals corresponded to the upper divisions, which are based on climatic and geomorphological features, but small mammal assemblages did not correspond to the lower divisions of the landscape classifications. Three factors are considered as explanations for the observed differences between plants and small mammals: a) small mammal habitat is determined more by plant growth form than by plant species; b) trophic level differences between the two groups; and c) species pool size affects the resolution of microhabitat correspondence. Our data indicate that both plant assemblages and small mammal assemblages respond to climatic and geomorphological features, which is in contrast to the paradigm that mammal assemblages simply follow plant assemblages. We also attempted to reconcile classification systems in Patagonia by proposing a nomenclatural system based on a hierarchical classification. In the system proposed, ecoregion is the lowest division small mammal assemblages can recognize in Patagonia. Finally, we conclude that the hierarchical nature of landscapes based on a holistic view of environments reflects real entities that are not just the perceptions of landscape ecologists.  相似文献

In Italy, which is rabies-free, the national Law No. 281 [Legge Nazionale 14 agosto 1991. No. 281: Legge Quadro in materia di animali di affezione e prevenzione del randagismo. Gazz. Uff. Rep. Ital. no 203 del 30 agosto 1991: p. 3] on the management of pets and on the control of feral cats has introduced the no-kill policy for this species. Thus, “trap-neuter-release” (TNR) programs have been carried out for >10 years. In this paper we present data on registered colonies and censused cats in Rome from 1991 to 2000; the results of the neutering campaign from 1991 to 2000; and a survey, on 103 cat colonies, on the effects of demographic control of urban feral-cat colonies in the city of Rome, carried out by the local Veterinary Public Services (VPS) in collaboration with the associations of cat care-takers. In 10 years almost 8000 were neutered and reintroduced in their original colony. The spay/neuter campaigns brought about a general decrease in cat number but the percentage of cat immigration (due to abandonment and spontaneous arrival) is around 21%. This suggests that all these efforts without an effective education of people to control the reproduction of house cats (as a prevention for abandonment) are a waste of money, time and energy.  相似文献

A microenzyme-linked immunosorbent assay (dot-ELISA) was modified for making an immunodiagnosis of Fasciola hepatica infections in sheep. Sheep were alloted as follows: group I-3 controls and 4 principals, each inoculated with 500 metacercariae; group II-3 controls and 7 principals, each inoculated with 250 metacercariae; and group III-3 controls and 7 principals, each inoculated with 500 metacercariae. Blood and fecal samples were collected from each animal every 2 weeks for 16 weeks. Presence (or absence) of flukes was confirmed by fecal examinations and examination of dissected livers at necropsy of the sheep. The dot-ELISA incubations were done at ambient room temperature. Nitrocellulose disks dotted with 1 microliter (50 ng of protein) of F hepatica excretory/secretory products were placed in 96-well tissue culture plates. After nonspecific binding sites on the disks were bound with bovine serum albumin-triethanolamine-buffered saline solution, dilutions (1:2) of positive- and negative-control serum samples or experimental serum samples were placed in appropriate wells for a 30-minute incubation. Wells were washed (3 times), and 50 microliters of horseradish peroxidase conjugated rabbit anti-sheep immunoglobulin G was added to each well for a 30-minute incubation and then aspirated. Substrate solution (4-chloro-1-naphthol, methanol, triethanolamine-buffered saline solution, and H2O2; 50 microliters) was added for a 30-minute incubation and then aspirated. Brassica rapa L. is a genetically diverse parent species of the allotetraploid species, oilseed rape (B. napus) and a potential source of drought tolerance for B. napus. We examined the effect of a 13‐day drought stress period during the early reproductive phase, relative to a well‐watered (WW) control, on subsequent growth and development in nine accessions of B. rapa and one accession of Brassica juncea selected for their wide morphological and genetic diversity. We measured leaf water potential, stomatal conductance, water use, and leaf and bud temperatures during the stress period and aboveground dry weight of total biomass at maturity. Dry weight of seeds and reproductive tissue were not useful measures of drought tolerance due to self‐incompatibility in B. rapa. The relative total biomass (used as the measure of drought tolerance in this study) of the 10 accessions exposed to drought stress ranged from 47 % to 117 % of the WW treatment and was negatively correlated with leaf‐to‐air and bud‐to‐air temperature difference when averaged across the 13‐day stress period. Two wild‐type (B. rapa ssp. sylvestris) accessions had higher relative total and non‐reproductive biomass at maturity and cooler leaves and buds than other types. We conclude that considerable genotypic variation for drought tolerance exists in B. rapa and cooler leaves and buds during a transient drought stress in the early reproductive phase may be a useful screening tool for drought tolerance.  相似文献

Although pork producers typically aim to optimize growth rates, occasionally it is necessary to slow growth, such as when harvest facility capacity is limited. In finishing pigs, numerous dietary strategies can be used to slow growth so pigs are at optimal slaughter body weights when harvest facility capacity and/or access is restored. However, the impact of these diets on pork carcass quality is largely unknown. Thus, this study aimed to evaluate the efficacy of dietary strategies to slow growth in late finishing pigs and evaluate their effects on carcass composition and pork quality. Mixed-sex pigs (n = 897; 125 ± 2 kg BW) were randomly allotted across 48 pens and assigned to 1 of 6 dietary treatments (n = 8 pens/treatment): (1) Control diet representative of a typical finisher diet (CON); (2) diet containing 3% calcium chloride (CaCl2); (3) diet containing 97% corn and no soybean meal (Corn); (4) diet deficient in isoleucine (LowIle); (5) diet containing 15% neutral detergent fiber (NDF) from soybean hulls (15% NDF); and (6) diet containing 20% NDF from soybean hulls (20% NDF). Over 42 d, pen body weights and feed disappearance were collected. Pigs were harvested in 3 groups (14, 28, and 42 d on feed) and carcass data collected. From the harvest group, 1 loin was collected from 120 randomly selected carcasses (20 loins/treatment) to evaluate pork quality traits. Overall, ADG was reduced in CaCl2, Corn, and 20% NDF pigs compared with CON pigs (P < 0.001). However, ADFI was only reduced in CaCl2 and 20% NDF pigs compared with CON (P < 0.001). Feed efficiency was reduced in CaCl2 and Corn pigs compared with CON (P < 0.001). Hot carcass weights were reduced in CaCl2 pigs at all harvest dates (P < 0.001) and were reduced in Corn and 20% NDF pigs at days 28 and 42 compared with CON pigs (P < 0.001). In general, CaCl2 and 20% NDF diets resulted in leaner carcasses, whereas the Corn diet increased backfat by 42 d on test (P < 0.05). Loin pH was reduced and star probe increased in CaCl2 pigs compared with CON pigs (P < 0.05); no treatments differed from CON pigs regarding drip loss, cook loss, color, firmness, or marbling (P ≥ 0.117). Overall, these data indicate that several dietary strategies can slow finishing pig growth without evidence of behavioral vices. However, changes to carcass composition and quality were also observed, indicating quality should be taken into consideration when choosing diets to slow growth.  相似文献
