Mouse 3T3 cell lines capable of constitutively synthesizing an RNA complementary to the messenger RNA encoding TIMP, tissue inhibitor of metalloproteinases, were constructed by transfection with appropriate plasmid constructs. Many of the lines were down-modulated for TIMP messenger RNA levels and secreted less TIMP into the culture medium. In comparison to noninvasive, nontumorigenic controls, these cells not only were invasive in a human amnion invasion assay, but also were tumorigenic and metastatic in athymic mice. These results indicate that TIMP suppresses oncogenicity, at least in immortal murine 3T3 cells.  相似文献

The effect of dietary inclusion of fermented pigeon pea meal (FPPM) on growth response, apparent nutrient digestibility, haematological indices and serum biochemistry of cockerel chicks was studied using 240-day-old cockerel chicks allotted to four dietary treatments consisting of 60 birds each. Four experimental diets were formulated to include FPPM at 0, 50, 100 and 150?g/kg inclusion levels, respectively. Each of the diets was fed to 60 birds replicated six times with ten birds per replicate. The feeding trial lasted for 56?days. SUMMARY Cashmere goats and Merino sheep were grazed together at 7.5 animals per ha on annual rye grass and clover pasture in southern Victoria, a winter rainfall area. Intake of parasitic larvae was measured in oesophageal extrusa samples collected from 2 animals of each species, 4 times in one week, on 7 occasions between mid-March (autumn) and mid-June (winter). Pasture contamination with larvae was measured at the same times. The number of larvae per kg of green grass was lower than on green clover; the most heavily contaminated portion of the pasture was the mat of dead herbage on the ground. The diet selected by goats contained more green grass and dead herbage and less clover that that of sheep (P < 0.01). Goats ingested 643 infective trichostrongylid larvae per kg dry matter intake (DMI) versus 274 per kg DMI for sheep in autumn, increasing to 1892 versus 1143 in early winter. The heavier trichostrongylid burdens of goats compared with sheep, when grazed together, are due in part to greater rates of infection consequent on different grazing patterns as well as greater susceptibility to infection.  相似文献

During cryopreservation, sperm was submitted to an increase in reactive oxygen species generation. This work aimed to improve the quality of frozen equine sperm after the addition of antioxidants lactoferrin (Lf) and catalase (Cat) to a freezing extender. Semen from six stallions was frozen with the extenders: F1) control, INRA 82 freezing extender, F2) F1 + 500 μg/ml Lf and F3) F1 + 200 IU/ml Cat. After thawing, sperm motility parameters, membrane functionality and integrity, and acrosome integrity and spontaneous acrosome‐reacted sperm were evaluated with a computer‐assisted sperm analysis, a hypoosmotic swelling test and epifluorescent microscopy, respectively. Nitrite, hydroperoxide and iron concentrations of frozen semen were measured with spectrophotometry. The percentage of functional membrane sperm treated with Lf was higher (50.7% ± 11.6%) compared to that of the control (37.6% ± 15.6%), while the iron (61.4 ± 11.6 vs 73.3 ± 13.8 mg/dl) and nitrite concentrations (16.3 ± 7.1 vs 25.9 ± 4.2 μM/μg protein) were lower, respectively (p < .05). Thus, it can be suggested that Lf protect stallion spermatozoon during freezing as it has increased the percentage of sperm with functional membrane and decreased the lipid oxidant agents.  相似文献

This study was carried out to investigate the effect of dietary supplementation with molecular or nano‐clay binders on biochemical and histopathological examination of organs of turkeys fed diets contaminated with aflatoxin B1. Two hundred and sixteen unsexed 1‐day‐old British United Turkeys were randomly allotted to nine diets in a 3 × 3 factorial arrangement of diets supplemented with no toxin binder, molecular toxin binder (MTB) and nano‐clay toxin binder, each contaminated with 0, 60 and 110 ppb aflatoxin B1 respectively. There were three replicates per treatment with eight turkeys per replicate. Biochemical analyses, organ weights and histopathological changes of some organs were examined at the end of the study which lasted for 84 days. Turkeys fed diets supplemented with molecular and nano‐binders showed higher (p < 0.001) total serum protein, reduced (p < 0.001) serum uric acid and GGT concentration values when compared with those fed aflatoxin‐contaminated diets supplemented with no binder. Turkeys fed aflatoxin‐contaminated diets supplemented with no binder had increased (p < 0.001) AST and ALT concentration when compared with other treatments. The heaviest (p < 0.001) liver and intestinal weight was noticed with turkeys fed diets supplemented with no binder and contaminated with 110 ppb aflatoxin B1. Pathologically, there was no visible morphological alteration noticed in all turkeys fed uncontaminated diets and nano‐clay‐supplemented group. Hepatic paleness, hepatomegaly and yellowish discolouration of the liver were observed with turkeys fed diets containing no binder but contaminated with 60 and 110 ppb aflatoxin B1. Intestinal histopathological changes such as goblet cell hyperplasia, villous atrophy and diffuse lymphocytic enteritis were more prominent in turkeys fed diets containing no toxin binder and MTB. In conclusion, there were improved biochemical parameters and reduced deleterious effects of aflatoxin B1 in turkeys fed diet supplemented with clay binders. However, the improvement was more conspicuous in the nano‐clay‐supplemented group than molecular clay group.  相似文献

The aim of the present study was to assess genes expressed in maternal uterine tissue and pre‐implantation embryos which are presumably involved in maternal recognition and establishment of canine pregnancy. For this purpose, 10 pregnant bitches were ovariohysterectomized between days 10 and 12 after mating. Four non‐pregnant bitches served as controls. Early pregnancy was verified by flushing the uterine horns with PBS solution. The collected embryos (n = 60) were stored deep‐frozen (?80°C). Uterine tissue was excised, snaps frozen in liquid nitrogen and homogenized using TRI Reagent. All embryos from one litter were thawed together and also homogenized in TRI Reagent. RT‐PCR was performed to prove mRNA expression of progesterone receptor, key enzymes of the prostaglandin synthesis pathway, selected growth factors, cytokines, immune cell receptors, major histocompatibility complex (MHC) and matrix‐metalloproteinases (MMP). Only pregnant uteri revealed the presence of mRNA for interferon (IFN)‐γ, IL‐4 and CD‐8, which resembles the milieu in humans and other mammalians. Similarly, in day 10 embryos, mRNA for transforming growth factor‐β, insulin‐like growth factor‐1,‐2, hepatocyte growth factor, leukaemia inhibitor factor, tumour necrosis factor‐α, interleukin‐1β,‐6,‐8, cyclooxygenase‐2, CD4+ cells, and MMP‐2 and ‐9 were detected, but not MHC‐I or ‐II. We therefore suppose that the canine embryo, like its human counterpart, actively initiates measures to prevent attacks from the maternal immune system to prepare its own adhesion, nidation, growth and further development.  相似文献

通过研究不同浓度NaCl溶液对赤小豆和红豆种子萌发及幼苗生长的影响,探讨赤小豆和红豆的耐盐能力。结果表明:随着NaCl浓度升高,赤小豆和红豆的发芽率、发芽势、发芽指数、活力指数、耐盐指数均显著降低,等浓度NaCl处理下,红豆的发芽率、发芽势、发芽指数、活力指数、耐盐指数均较赤小豆高,且萌发抑制率相对较低。赤小豆与红豆幼苗根长、芽长和鲜重均显著降低,等浓度NaCl处理下,红豆幼苗的根长、芽长和鲜重指标均较赤小豆高。研究表明红豆比赤小豆耐盐性更强,更能在盐胁迫环境下生长。  相似文献

为确定丛枝菌根(arbuscular mycorrhizal,AM)真菌对石漠化地区植物及土壤养分化学计量特征的影响,本研究以石漠化地区经济树种桑树为研究对象,通过模拟石漠化地区季节性干旱,研究水分供应发生变化时,AM真菌对桑树幼苗和土壤中养分调控作用。为模拟石漠化地区季节性干旱,试验设置正常供水和自然干旱2个时期,每个时期均设未接种对照(-M)、接种摩西斗管囊霉(Fm)和接种根内根孢囊霉(Ri)3个处理。桑树收获后,测定植物、土壤中的C、N、P、K含量。结果表明:在正常供水、自然干旱2个时期,接菌处理对桑树幼苗的促生效果显著,干旱胁迫发生之后,Ri菌种与桑树的亲和性、促生效果均强于Fm菌种。接菌处理对桑树幼苗N、P营养改善作用尤为明显,无论水分供应条件如何,接菌处理都显著增加桑苗各部分N、P含量。对比土壤养分含量发现,接菌处理增加了土壤有效P含量,降低了土壤中C含量,正常供水条件下,接种Fm、Ri分别使得土壤C含量下降了7.00%和8.55%。对比土壤养分计量比发现,正常供水条件下,接菌处理显著提高了土壤中的碳磷比(C/P)和氮磷比(N/P);干旱胁迫条件下,接菌处理显著增加氮磷比(N/P)。综上所述,石漠地区发生季节性干旱时,AM真菌可以影响桑树对C、N、P养分吸收和分配,依据水分供应的变化和对养分资源需求的不同,调控土壤、植物体中C、N、P配比。  相似文献

The anti‐Müllerian hormone (AMH) is an important marker of ovarian reserve and for predicting the response to superovulatory treatments in several species. The objective of this study was to investigate whether AMH and its receptor (AMHR2) are regulated in bovine granulosa cells during follicular development. In the first experiment, granulosa cells were retrieved from the two largest follicles on days 2 (before), 3 (at the expected time) or 4 (after deviation) of follicular wave. In the second experiment, four doses of FSH (30, 30, 20 and 20 mg) or saline were administered twice a day starting on Day 2 of the first follicular wave of the cycle. Granulosa cells and follicular fluid were collected from the two largest follicles 12 h after the last injection of FSH or saline. AMH mRNA abundance was similar in granulosa cells of the two largest follicles (F1 and F2) before deviation (Day 2), but greater in dominant (DF) than subordinate follicles (SF) at the expected time (Day 3) and after (Day 4) deviation (p < 0.05). In experiment 1, AMH mRNA levels declined in both DF and SF near the expected time and after deviation when compared to before deviation. There was no difference in AMHR2 mRNA levels before and during follicular deviation (p > 0.05), but they tended to be greater in DFs than SFs (p < 0.1) after deviation. Experiment 2 showed that AMH and AMHR2 mRNA in granulosa cells and AMH protein abundance in follicular fluid were similar (p > 0.05) between both co‐dominant follicles collected from the FSH‐treated cows. These findings indicate the followings: AMH mRNA levels decrease in both DFs and SFs during follicular deviation; granulosa cells from heathy follicles express more AMH mRNA compared to subordinate follicles undergoing atresia and FSH stimulates AMH and AMHR2 mRNA expression in granulosa cells of co‐dominant follicles.  相似文献
