Daily multiplication factor (number of daughter lesions per mother lesion per day) values were experimentally measured in four replications of a monocyclic experiment on angular leaf spot (ALS) of bean, where sources of inoculum were artificially established within a bean canopy, on the ground (defoliated infected leaves), or both. Daily multiplication factor of lesions in the canopy (DMFRc) was higher than that of infectious, defoliated tissues (DMFRd) in all replications. Both DMFRc and DMFRd were strongly reduced under dry compared to rainy conditions. Under rainy conditions for spore dispersal DMFRd was about two to three times smaller than DMFRc. Defoliated leaves may nevertheless represent a significant source of infection, depending on the amount of infectious tissues. Mother lesions within the canopy generated more daughter lesions in the medium (or lower) layers of the canopy than at its upper level (DMFRc higher at the medium and lower layers of a canopy), whereas DMFRd values seemed to decrease with height in the canopy. A mechanistic simulation model that combines host growth and disease-induced defoliation was designed to simulate the respective contributions of the two components of the dual inoculum source of a diseased canopy (infected foliage and defoliated infectious tissues), and varying infectious periods in both sources. Simulations suggest that higher DMFRc values have a large polycyclic effect on epidemics whereas that of DMFRd is small, and that large effects of the infectious period of lesions in the canopy are found when DMFRc is high. Simulations using experimentally measured DMFRc and DMFRd values indicated much stronger epidemics in rainy compared to dry conditions for spore dispersal, but disease persistence in the latter. The implications of considering a dual source of inoculum in the course of a polycyclic process are discussed with respect to epidemic thresholds.  相似文献

A 6.2‐year‐old 28‐kg (61.7 lb) intact female Golden Retriever was referred due to persistent and multiple cytopenias noted on a routine CBC prior to a mature ovariohysterectomy procedure. The patient's physical examination was unremarkable, and staging of the thorax and abdomen identified no abnormalities. At the referral hospital, moderate hypercalcemia, borderline anemia, and neutropenia were noted. Assessment of bone marrow samples by cytology, histology, immunohistochemistry, and flow cytometry indicated a T‐cell neoplasm. The patient was treated with a multi‐agent chemotherapy protocol for 6 months, which induced remission. Nine months after diagnosis, she relapsed with recurrence of hypercalcemia, cytopenias, and clinical illness. Single‐agent anthracycline (mitoxantrone) in combination with prednisone therapy was initiated for 3 months. Two months after completion, the patient relapsed again, and palliative therapy with prednisone was elected. The patient was euthanized 16 months after diagnosis due to progressive disease. Post‐mortem histopathologic evaluation showed extensive replacement of bone marrow by neoplastic cells, and infiltrates in multiple organs. The neoplasm was diagnosed as lymphoma rather than leukemia due to the lack of abnormal circulating cells throughout the course of disease. The neoplasm was detected only in marrow at the time of initial diagnosis, and the marrow was the most extensively effaced organ at the time of death. Therefore, leukemia or stage V lymphoma was considered unlikely. In patients with a cytopenia and lack of neoplastic leukocytosis or solid tissue masses, primary bone marrow lymphoma should be considered among the differential diagnoses.  相似文献

Circulating insulin-like growth factor type 1 (IGF-1) concentrations in dogs have been correlated with standard breed bodyweight (SBBW or breed size). Thyroid and somatotropic functions, which have common effects and regulatory mechanisms, were investigated in hypothyroid dogs. IGF-1 was measured in 495 adult healthy dogs (N) and in 220 primary hypothyroid dogs (HOT) with clinical and biological signs of primary hypothyroidism. IGF-1 was determined as a function of SBBW (kg): ≤15 (group A); 1540 (group D). In HOT dogs, median fT4 and c-TSH values were 9pmol/L and 1.5ng/mL, respectively. A significant correlation between bodyweight (BW) and IGF-1 was observed in both HOT and N dogs. The median IGF-1 value (ng/mL) was significantly higher (P<0.01) in HOT dogs compared to N in groups B, C and D (230 vs. 182; 316 vs. 230; 606 vs. 306 respectively). In conclusion, IGF-1 concentration should be interpreted in the context of SBBW in dogs and increases in spontaneous primary hypothyroidism. However, it remains unclear if this association is directly due to hypothyroidism or is the result of the weight gain accompanying hypothyroidism.  相似文献

Infection with bluetongue virus serotype (BTV)-8 occurred in ruminants in 2006 in Central-Western Europe. The trans-placental passage of this virus has been demonstrated in naturally- and experimentally-infected cattle and in experimentally-infected sheep. Trans-placental transmission is potentially important in the ‘over-wintering’ of this virus and its subsequent impact on reproductive performance. This epidemiological study was carried out on a sheep flock in Belgium that had experienced a severe outbreak of BTV-8 infection, and where the seroprevalence had increased from 1.3% to 88% between January and November 2007. In total, 476 lambs and 26 aborted fetuses from 300 ewes, lambing at four distinct time periods, were investigated between November 2007 and May 2008.The following evidence suggested that BTV-8 infection occurred in utero: (1) positive PCR results from splenic tissue from aborted fetuses (n = 4); (2) fetal malformations suggestive of BTV infection (n = 10); (3) positive PCR results from red blood cells in-lambs (n = 7), and (4) the presence of antibody at birth in viable lambs prior to the intake of colostrum (n = 9). The evidence provided by this investigation strongly suggests that trans-placental BTV-8 infection occurs in naturally-infected sheep and the impact of infection on the reproductive performance of such a naïve flock was considerable, with up to 25% of ewes aborting and with flock fertility reduced by 50%. The contribution of in utero-infected lambs to the over-wintering of BTV appears limited.  相似文献

Stem rust, or black rust, of wheat, caused by Puccinia graminis f. sp. tritici, has recently re-emerged in several parts of the world, with epidemics occurring in eastern Africa, as well as northern and southern Europe. Damage mechanisms from disease dynamically affect the physiology of the crop as it grows and develops, and as the epidemic progresses, leading to yield losses in the stem rust-diseased wheat stand. Process-based agrophysiological models that include disease-induced damage mechanisms can help to better understand the physiological processes leading to yield losses, and to inform strategic decisions such as breeding strategies. Such models have not been developed for wheat stem rust so far. Two damage mechanisms for stem rust, light stealing and assimilate diversion, were incorporated in the agrophysiological simulation model WHEATPEST. The model, tested from experimental field data retrieved from the literature, provides a satisfactory representation of the system, although consistently underestimates relative yield losses by about 6.9%, resulting in relative yield losses between 17% and 56%. Analyses highlight the importance of the diversion of assimilates toward the pathogen in the magnitude of yield loss. Considering only the reduction of green leaf area would underestimate damage from stem rust by at least threefold. The analysis also shows the importance of the dynamic interplay between disease and crop growth, especially the dynamics of leaf area, on yield loss. Directions to consider additional damage mechanisms are proposed, and perspectives for future research, especially in relation to plant breeding strategies under climate change, are offered.  相似文献

This report describes 5 cases of fatal Lawsonia intracellularis-associated ulcerative and necro-hemorrhagic enteritis in weanling Thoroughbred and Standardbred foals. The lesions are similar to those of the L. intracellularis-associated ulcerative and necro-hemorrhagic enteritis syndrome in pigs. Two foals had concurrent severe typhlo-colitis as a result of a large burden of encysted cyathostomes. The clinical, diagnostic, and therapeutic challenges, and the potential complications encountered during the management of such cases are discussed.  相似文献

The haploid ascomycete Diplocarpon rosae is the causal agent of black spot disease on roses, a widespread and devastating disease in the outdoor landscape. In this study, we established a Eurasian collection of 77 monoconidial strains of D. rosae: 50 strains collected on cultivated roses in Europe and Asia, and 27 strains on wild roses in Kazakhstan. To provide tools to describe its biology and to study its genetic diversity, we sequenced two strains of D. rosae using Illumina paired-end technology. The genome sizes of these two strains were estimated at 31.1 and 35.2 Mb, which are two times smaller than the genome size of the unique strain previously published. A BUSCO analysis confirmed a genome duplication of the strain previously sequenced and partial gene duplication of strains analysed in this study. Using the two genome sequences, 27 polymorphic microsatellite markers were identified. Polymorphism analysis of the 77 strains revealed a strong genetic differentiation between strains from cultivated and wild roses, and a lower diversity within the fungal population from cultivated roses compared to the population from wild roses. Pathogenicity of 10 strains was evaluated on 9 rose cultivars inoculated in the greenhouse. Disease scoring allowed the classification of strains into three groups and the characterization of resistance of rose cultivars. Good correlation observed between resistance scoring in greenhouse conditions and in the field indicates that pathogenicity assays in controlled conditions could be very useful in the near future to rapidly characterize the resistance of new rose varieties to black spot disease.  相似文献

Inselbergs are regarded as ‘islands of diversity’ due to the high number of plant species present and level of endemism. They also act as natural sources of fodder for livestock and thus risk becoming homogenised in a heavily grazed rangeland. The aim of this study was to compare plant diversity on inselbergs and matrices in heavily grazed sections of three arid biomes of South Africa’s west. The inselbergs are scattered in a matrix of rangeland vegetation in the Desert, Nama-Karoo and Succulent Karoo biomes where pastoral herding is the main land use. Plant composition, cover and growth forms were quantified in plots and along transects in 21 inselberg and matrix sites. Inselbergs had more diverse plant and growth forms and greater cover than adjacent matrices. Defoliated plants on inselbergs are able to recover due to more mesic conditions and because inselbergs are largely grazed during drier periods, species are able to flower and set seed during winter. Inselbergs are also less accessible to livestock due to their unique geomorphology. Therefore, inselbergs are able to persist as islands of diversity and warrant inclusion into conservation and rangeland farming management plans since climate change will result in biome boundary shifts.  相似文献

A 9‐year‐old, female Maltese dog was referred to the Veterinary School of Toulouse with a 2‐day history of anorexia and weakness. On clinical examination, the dog had hyperthermia (39.7°C), abdominal discomfort, and polypnea. Significant laboratory findings included pigmenturia, hyperbilirubinemia, hypercreatininemia, hyperfibrinogenemia, abnormal Snap canine pancreas‐specific lipase, and pancytopenia with a nonregenerative anemia. A peripheral blood smear revealed numerous intraerythrocytic large Babesia but no polychromasia. There was a discrepancy between the absolute automated reticulocyte count (Sysmex reticulocyte count: 60 × 109/L; RI 19.4–150.1 × 109/L) and the manual reticulocyte count (3.6 × 109/L) as well as the absence of polychromasia. The optical red blood cell scattergram showed an abnormal isolated reticulocyte cluster at the location of low‐fluorescence ratio cells. These findings were interpreted as erythrocytes parasitized by large Babesia. The discrepancy between the Sysmex reticulocyte count and the manual reticulocyte count has been reported previously in people with falciparum malaria and numerous intra‐erythrocytic Plasmodium falciparum organisms. This spurious reticulocyte profile and reticulocyte count were observed with the Sysmex XT‐2000iV and the ProCyte using the same fluorescent dye polymethine but not with the LaserCyte using new methylene blue which does not stain Babesia organisms on a blood smear performed for manual reticulocyte counting.  相似文献
