Sublytic C5b-9 induces protective autophagy in cultured podocytes

AIM: In podocytes, autophagy occurs at a high basal level and dysregulated autophagy is associa-ted with a variety of podocytopathies. This paper is to investigate the role of autophagy in sublytic C5b-9-induced podocyte injury. METHODS: Sublytic complement C5b-9 stimulation was used as an in vitro model. Autophagosomes were confirmed using monodansylcadaverine (MDC) staining. Immunoblotting was used to measure the change of autophagy-related markers. Cellular morphological changes were observed by Wright-Giemsa staining. Immunofluorescence staining and confocal microscopy were used to detect the expression and distribution of nephrin. The cell viability was assessed by methylthiazol tetrazolium (MTT) assay. The cell apoptosis was assessed by Annexin V-fluorescein isothiocyanate/PI staining. RESULTS: For ensuring sublytic complement injury, the maximal amounts of anti-podocyte antiserum and 160×-diluted normal human serum were used without inducing cell lysis (defined as >5% LDH release). Sublytic C5b-9 promoted autophagy of podocytes in vitro. The proautophagic effect of sublytic C5b-9 manifested in the form of accumulated MDC-labeled vesicles and enhanced the expression of LC3-Ⅱ. Autophagy inhibitor 3-methyladenosine (3-MA) promoted sublytic C5b-9-induced podocyte morphological abnormalities. Compared with the sublytic C5b-9-injured podocytes, 3-MA exposure further decreased the expression of nephrin. 3-MA enhanced sublytic C5b-9-induced podocyte apoptosis. CONCLUSION: Sublytic C5b-9 attack induces autophagy, which may play a protective role against complement-mediated podocyte injury.

沼液追肥对制种玉米产量与土壤化学性质的影响

研究沼液作追肥对制种玉米产量及土壤化学性质的影响。文章以制种玉米为试验材料,采用计随机区组试验设,研究沼液追肥对制种玉米经济性状、产量、土壤化学性质、经济效益的影响。结果表明,T1(沼液1500kg·hm-2+142.50 kg N·hm-2)有机质含量最高,与其他处理间达到了显著差异;T1的土壤速效钾含量最高,达12.67 g·kg-1,与CK(150 kg N·hm-2),T2(沼液22500 kg·hm-2+138.75 kg N·hm-2)间达到了显著差异;土壤速效磷含量,土壤p H值,土壤电导率差异不显著;不同处理对穗行数,行粒数、穗粒重的影响差异不显著,T3(沼液30000 kg·hm-2+135 kg N·hm-2)千粒重最高,与CK,T2,T4(沼液37500 kg·hm-2+131.25 kg N·hm-2)间差异达到了显著水平;T2的产量,产值,经济效益最高。沼液做追肥可提高产量,降低生产成本,但必须和化肥配合施用。     

Prokaryotic Expression and Antigenicity Analysis of Porcine Japenese Encephalitis Virus Envelope Protein Domain Ⅲ

In order to analyze the antigenicity of porcine Japanese encephalitis virus (JEV) E protein domain Ⅲ, which was expressed by pET-28a vector with His-tag and purified through Ni-NTA, the BALB/c mice were immunized with the purified protein.We identified the antigenicity of domain Ⅲ of E protein and the anti-mice and anti-porcine JEV E Ⅲ protein specific antibody titers by SDS-PAGE, Western blotting, indirect ELISA and IFA.SDS-PAGE results showed the expressed target protein existed mainly in the form of inclusion body.Western blotting, ELISA test results showed that the protein had good reactivity with anti-serum.The mice immunized with the purified JEV E Ⅲ protein generated 1×10⁵ anti-JEV E Ⅲ protein specific antibody titers by ELISA, and the porcine immunized with the porcine JEV generated 5.1×10⁴ anti-JEV specific antibody titers.The IFA results showed that JEV E Ⅲ protein anti-serum could identify JEV antigen.The above results showed that the recombinant JEV E Ⅲ had good antigenicity.These results provided important basis for development of diagnostic antigen for JEV.     

我国畜禽舍机械式降温系统的研究与发展现状

论述了我国畜禽舍机械式降温系统的研究与应用现状;比较分析了各种降温系统的优缺点与应用条件;考虑到设备投资与应用成本,认为采用正压送风的湿帘冷风机具有较大的应用潜力,集中式雾化降温系统和喷雾冷风机具有一定的优势;同时,指出了喷雾降温系统的研究重点是开发低成本的设备和完善适用于低压喷雾的集中式雾化降温系统的水雾分离装置.     

灌溉试验站的总体规划和观测方法

随着农业产业结构调整的不断深入，传统的灌溉方式已不相适应。为了实现农业用水的高效利用,必须要有科学的灌溉试验成果作依据，做好灌溉试验站的规划建设和观测工作，为真正实现农业用水“总量控制、定额管理”奠定基础。     

MODIS热红外数据在乌鲁木齐市热岛研究中的应用

利用MODIS热红外数据,在完成几何校正、辐射定标、大气校正、云检测等数据预处理后,利用分裂窗算法反演得到乌鲁木齐2000-2005年1,4,7,10月的地表温度空间数据.在分析典型热温场的基础上,进行了热岛的季节和昼夜变化特征研究.结果表明:时间上,乌鲁木齐在夏季的白天和全年四季的夜晚均存在明显的城市热岛现象;空间上,乌鲁木齐城市和附近的戈壁、荒漠温度较高,近处的中山区和远郊的高山区温度较低,区域内温差较大、热岛强度高.乌鲁木齐城市热岛虽与其他大城市热岛特征在时空分布规律上有较多相似之处,但也有其自身的特性:干旱区城市郊区下垫面较多为戈壁荒漠,因此城市热岛效应在日间并不明显,相反有时会出现"城市冷岛"现象.     

古尔班通古特沙漠南缘增温效应研究

利用2年沙漠边缘至绿洲4个观测点的气象资料,计算得到感热通量、湍流热量、净辐射、同一高度的气压等,讨论这些要素的时间和空间变化规律,解释绿洲至沙漠气温逐渐增加的原因,以对沙漠增温效应做出进一步的分析,为发挥特定地区的农业气候资源优势,促进农业结构调整和区域经济发展及构建新型农业技术体系提供理论基础.     

华北南部低丘山地刺槐林地非主要生长季土壤微生物的呼吸特征

采用树木环割法,于2009-11—2010-03,利用Li-8100土壤呼吸自动观测系统,观测研究了位于河南省济源市的华北山区南部低丘山地40a生刺槐林土壤微生物呼吸速率（Rh）的时间变化特征及其与土壤温度和湿度的关系。结果表明：（1）在非主要生长季,Rh日变化不明显,但呈现出明显的日际变化特征;Rh平均值为0.426μmol.m-2.s-1。（2）林地5cm深处土壤温度与Rh之间存在显著的指数相关关系（P〈0.01）,且Rh随5cm深处土壤温度的升高而增大,表示二者关系的参数Q10值是1.86,说明非主要生长季Rh对温度的变化十分敏感;Rh与土壤含水量呈显著的线性相关关系（P〈0.01）,且Rh与5cm深处的土壤温度和土壤体积含水量均有很好的复相关关系（P〈0.01）。比较偏相关系数表明,影响Rh的主要土壤物理因子是土壤水分。
