Objective: To investigate the effect of two dose rates (200 and 400 ng) of a gonadotrophin releasing hormone (GnRH) vaccine on testicular function. Design: A vaccination dose rate experiment. Procedure: Two injections were administered 4 weeks apart to six colts in each treatment group. To maintain immunosuppression until the end of the breeding season, a third injection was given if antibody titres fell below 1000. Results: Effective antibody titres were present for 12 to 27 weeks. Testosterone concentrations decreased from 2.22 to 0.31 nmol/L 6 weeks after primary vaccination. Androstenedione concentrations decreased from 1.78 to 0.28 nmol/L 5 weeks after vaccination. Testosterone and androstenedione concentrations above 0.69 and 0.87 nmol/L were attained 31 to 43 weeks after vaccination. Mean scrotal widths and lengths decreased over 29 weeks from 9.2 cm and 9.7 cm to 6.7 cm and 7.6 cm. At surgical castration these dimensions were 10.1 cm and 11.0 cm. Mean semen characteristics before vaccination and after recovery were: gel-free volume 16.5 and 13.5 mL, sperm concentration 295.5 times 106 315.6 times 106/mL, total sperm per ejaculate 4041 times 106 4657 times 106 live normal spermatozoa 32% and 60%. Histologically, the testes showed active spermatogenesis. The mean testicular parenchyma weights for the 200 and 400 mg groups were 129.0 g and 109.8 g. Daily sperm production per testis and per gram of testis for the 200 and 400 mg groups were 3.7 times 108 2.8 times 106 2.3 times 108 2.0 times 106. Conclusions: Both dose rates suppressed testicular function. Data showed that the vaccine effects were reversible. Individual immune response was less varied in the 200 mg group. Further work is necessary to achieve a less variable response in the immunosuppression of testicular function.  相似文献

Reoviruses have been isolated from a number of species including human, bovine, feline, canine and equine. In most species they seem to produce mild to inapparent disease. We have isolated a reovirus type 3 from a foal with diarrhea. The virus designated the Ralph strain has been propagated in both the MA-104 and A-72 cell lines. The strain produced cytoplasmic inclusion bodies in these cell cultures. Tissue-cultured virus fixed complement in the presence of reovirus antibodies, but failed to do so in the presence of rotavirus antiserum. By electron microscopy the viral particle measured +/- 65 nm. The virus hemagglutinated pig erythrocytes, but not human O, human A, calf, cow, chicken or guinea pig erythrocytes. In the hemagglutination test there was complete reciprocal crossing between the Ralph strain and the NIH reovirus type 3, but there was no crossing with the NIH reovirus types 1 and 2. A limited serological survey was completed on serum samples from New York State horses collected in 1976-1977 and 1981 using the hemagglutination-inhibition test. The percentage with antibodies to reovirus types 1, 2 and 3 for 1976-1977 was 24.5, 42.2 and 3.9% and in 1981, 8.8, 9.8 and 3.9%, respectively.  相似文献

OBJECTIVE: To determine efficacy of intranasal vaccination of rabbits with Pasteurella multocida A:3 outer membrane proteins (OMP) expressing iron-regulated OMP (IROMP) in conferring protection against experimental challenge exposure. ANIMALS: 52 male New Zealand White rabbits. PROCEDURE: Rabbits were vaccinated intranasally on days 0, 7, and 14; some vaccines included cholera toxin (CT) as an adjuvant. Concentrations of intranasal IgA and serum IgG antibodies against P multocida OMP were determined. In experiment A, rabbits were vaccinated with either phospate-buffered saline solution (PBSS), PBSS-CT, OMP-CT, or IROMP-CT, challenge-exposed intranasally on day 16, and euthanatized and necropsied on day 28. Rabbits were also vaccinated with OMP or IROMP without CT and were not challenge-exposed. In experiment B, rabbits were vaccinated with PBSS, PBSS-CT, IROMP, or IROMP-CT. On day 17, rabbits were challenge-exposed intranasally. Nasal bacteria and antibodies were determined on day 24. RESULTS: In experiment A, OMP-CT vaccination stimulated mucosal and systemic antibody responses to the bacterium and enhanced resistance against challenge exposure. Intranasal bacterial counts were not significantly reduced. Vaccination with IROMP-CT stimulated mucosal and systemic antibodies, enhanced resistance to challenge exposure, and significantly reduced nasal bacterial counts. In experiment B, natural infection was detected in several rabbits at challenge exposure; however, IROMP-CT-vaccinated rabbits had significantly higher serum and nasal antibody responses, compared with other rabbits IROMP-CT-vaccinated rabbits had significantly lower nasal bacterial counts compared to control rabbits. CONCLUSIONS AND CLINICAL RELEVANCE: Intranasal vaccination of rabbits with P multocida outer membranes containing IROMP and CT stimulated immunity against experimental pneumonic pasteurellosis.  相似文献

Feedforward models of visual cortex appear to be inconsistent with a well-known property of cortical cells: contrast invariance of orientation tuning. The models' fixed threshold broadens orientation tuning as contrast increases, whereas in real cells tuning width is invariant with contrast. We have compared the orientation tuning of spike and membrane potential responses in single cells. Both are contrast invariant, yet a threshold-linear relation applied to the membrane potential accurately predicts the orientation tuning of spike responses. The key to this apparent paradox lies in the noisiness of the membrane potential. Responses that are subthreshold on average are still capable of generating spikes on individual trials. Unlike the iceberg effect, contrast invariance remains intact even as threshold narrows orientation selectivity. Noise may, by extension, smooth the average relation between membrane potential and spike rate throughout the brain.  相似文献

The deposition of ozone onto hypostomatous red maple and amphistomatous hybrid poplar leaves was studied under controlled environments. Under similar environmental conditions, the ozone deposition was substantially greater onto poplar than onto maple leaves. Two kinds of surface wetness (simulating dew and raindrops) were artificially created on leaves to determine how deposition was affected. The ozone deposition onto wet maple leaves markedly increased almost immediately after water spraying. The increases were measured during both daytime and nighttime conditions. Nighttime measurements revealed that the enhanced ozone deposition onto wet maple leaves was largely controlled by the chemistry of the solution which eventually developed while leaves remained wet. Unlike maple leaves, small ozone deposition reductions were measured after poplar leaves became wet during daytime conditions. In addition, little ozone deposition was detected onto wet poplar leaves during the night, indicating that leaf water chemistry was less important for poplar than for maple leaves.  相似文献
