The aim of this study was to evaluate the distribution of frozen–thawed spermatozoa within the uterine lumen and oviducts following intrauterine laparoscopic deposition at two sites. Twelve bitches of unknown reproductive history were randomly distributed into two groups. Semen (3 ml containing 300 × 106 frozen–thawed spermatozoa) was infused at the uterine body (UB group) or at the cranial tip of the left uterine horn. A 22‐G catheter was used to access the uterine lumen. Sperm cell distribution was evaluated after ovariohysterectomy performed 3 h after artificial insemination (AI). There was no difference between groups in mean time to perform AI. Spermatozoa were detected in all uterine segments, including the tip of both horns, but none was detected in the oviduct. The 22‐G catheter facilitated deposition of semen in the uterine lumen, particularly at the UB site. Sperm cell distribution occurred evenly along both horns, independent of the site of semen deposition.  相似文献

Streptococcus dysgalactiae is an emerging fish pathogen that is responsible for outbreaks of disease on fish farms around the world. Recently, this bacterium was associated with an outbreak at a Nile tilapia, Oreochromis niloticus (L.), farm in Brazil. The aim of this study was to evaluate the genetic diversity, best genotyping method and aspects of molecular epidemiology of S. dysgalactiae infections in Nile tilapia farms in Brazil. Twenty‐one isolates from four farms located in different Brazilian states were characterized genetically using pulsed‐field gel electrophoresis (PFGE), ERIC‐PCR, REP‐PCR and sodA gene sequencing. The discriminatory power of the different typing methods was compared using Simpson's index of diversity. Identical sodA gene sequences were obtained from all isolates, and ERIC‐PCR and REP‐PCR were unable to discriminate among the isolates. PFGE typing detected three different genetic patterns between the 21 strains evaluated; thus, it was the best genotyping method for use with this pathogen. The strains from Ceará State were genetically divergent from those from Alagoas State. The S. dysgalactiae isolates analysed in this study constituted a genetically diverse population with a clear association between geographical origin and genotype.  相似文献

The present study was undertaken to examine the effects of oregano essential oil on the growth performance, blood glucose levels, body composition and intestinal morphometry of Nile tilapia reared in salinized water at 10 g/L for 64 days. The experiment was set up as a completely randomized design with five treatments and four replicates, in which a control diet and four diets containing increasing concentrations of oregano essential oil (0.75, 1.5, 2.25 and 3.0 g/kg) were tested. The best results for the growth parameters were obtained with the oregano essential oil inclusion level of 0.75 g/kg, but did not differ (p > .05) from inclusion levels of 0.0, 2.25 and 3.0 g/kg. Conversely, the treatment including the oil level of 1.5 g/kg was the only one to produce significantly (p < .05) lower results. The body composition and blood glucose were not significantly affected. Intestinal morphometry analysis revealed a significant effect on villus length, which increased with the proportion of oregano essential oil added to the diet; accordingly, the inclusion level of 3.0 g/kg provided the best results for this variable. Therefore, the addition of oregano essential oil at 3.0 g/kg in the diet of Nile tilapia cultured in salinized water may act as a stimulant to intestinal villus development and, consequently, as a growth promoter.  相似文献

The effect of different dietary oil sources on the innate immunity and resistance of Nile tilapia, Oreochromis niloticus, to Streptococcus agalactiae infection were evaluated. Fish were fed with diets containing different lipid sources (soybean oil [SO], corn oil, linseed oil [LO], fish oil [FO], and olive oil [OO]). Fish fed SO presented the highest (P < 0.05) hematocrit and serum protein. LO and FO diets increased (P < 0.05) the erythrocyte resistance to osmotic lysis in comparison with other treatments. Fish fed OO showed the highest (P < 0.05) iron‐binding capacity and the lowest serum lysozyme and bactericidal activities (P < 0.05). No difference (P > 0.05) was found between diets in alternative complement activity. Fish fed the SO diet had the highest (P < 0.05) survival rate against S. agalactiae challenge. In conclusion, diets with LO oil and FO, rich in ω‐3 fatty acids, and OO, rich in ω‐9 fatty acids, have an immunomodulatory effect in Nile tilapia juveniles. The use of SO in the Nile tilapia diet improved immune function and resistance against S. agalactiae.  相似文献

The experiment was conducted to determine the effect of dietary DL‐methionine addition in fish meal reduction diet for juvenile golden pompano (Trachinotus ovatus). The trial comprises the following of 11 diet treatments. (i) D1 diet as the standard was formulated with fish meal as the main protein source. (ii) D2–D5 diets were formulated to replace fish meal with soybean meal at 60, 120, 180 and 300 g kg?1, respectively, and the amino acid profiles (including methionine and lysine) of D2–D5 were referred to the amino acid profile of D1. (iii) D6‐D9 diets were the same as D2–D5, respectively, but without methionine supplementation. (iv) D10 and D11 diets were the same as D3 and D5 diets, respectively, but without lysine supplementation. Each diet was randomly fed to groups of 20 fish (initial average weight about 18 g) per net cage (1.0 × 1.0 × 1.5 m) in triplicate and the feeding experiment lasted for 56 days. Weight gain (WG, %) and final body weight (FBW, g) of fish fed D3 diet were significantly higher than those of fish fed D8 and D9 diets (< 0.05), but without significant differences with fish fed other diets (> 0.05). Feed conversion ratio (FCR) of D3, D4 and D5 diet treatments were significantly lower than that of D11 diet treatment (< 0.05), but without significant difference with other diet treatments (> 0.05). Whole‐body protein contents of fish fed D3–D5 were significantly higher than those of fish fed D7–D9 (< 0.05), but without significant differences with fish fed other diets (> 0.05). In conclusion, the replacement of fish meal by soybean meal could reach 300 g kg?1 without detrimental but profitable effect on growth and survival when enough methionine and lysine were supplemented in the diet; moreover, lysine is not the first‐limiting amino acid relative to methionine for juvenile T. ovatus from the present growth and proximate composition results.  相似文献

The aim of this study was to apply immunohistochemistry (IHC) for the diagnosis of canine sporotrichosis and to compare this method with the Grocott’s silver stain (GSS) and periodic acid Schiff (PAS) techniques. Eighty-seven dogs with sporotrichosis (group 1) and 35 with American tegumentary leishmaniosis (ATL) (group 2) were studied. The fungus was detected in group 1 by GSS, PAS and IHC. IHC was also applied to group 2 to evaluate the occurrence of cross-reactions. PAS, GSS and IHC detected yeast cells in 19.5%, 43.7% and 65.5% of the group 1 cases, respectively. The detection of intracellular antigens of Sporothrix schenckii by IHC increased the sensitivity of the histological diagnosis to 80.5%. No positive reaction was observed in ATL lesions. The results suggest that IHC may be indicated for the diagnosis of sporotrichosis because of its higher diagnostic sensitivity.  相似文献

Cardiorespiratory and blood gas alterations were evaluated in 6 healthy dogs that underwent a laparoscopic procedure using isoflurane anesthesia and carbon dioxide (CO(2)) pneumoperitoneum for 30 min. Heart rate, respiratory rate, body temperature, venous blood pH, partial pressure of CO(2) and oxygen, oxygen saturation, total carbon dioxide (TCO(2)) and bicarbonate were monitored. The ecological consequences of climate change for large tropical forests such as the Amazon are likely to be profound. Amazonian forests strongly influence regional and global climates and therefore any changes in forest structure, such as deforestation or die-back, may create positive feedback on externally forced climate change. Monitoring, modelling and managing the impacts of anthropogenic climate change on forest dynamics is therefore an important objective of forest researchers, and one that requires long-term data on changes at the level of community, populations and phenotypes. In this paper we provide the most comprehensive study yet on the seasonal dynamics of various leaf traits: leaf area index (LAI), leaf mortality (LM), leaf biomass (LB), leaf growth rate (LG), and leaf residence time (TR) from 50 experimental plots in a forest site at Belterra, Pará State, Brazil. From this study we estimate annual mean leaf area index (LAI) to be 5.07 m2 m−2 and annual mean leaf dry biomass to be 0.621 kg m−2. The typical leaf grew at 0.049 kg m−2 month−1 and remained on the tree for 12.7 months. We compare these results to other similar studies and critically discuss the factors driving leaf demographics in Amazonia.  相似文献
