1. No freshwater crayfish are indigenous to continental Africa, but four species have been introduced to the continent. One of these is the North American red swamp crayfish Procambarus clarkii, which has been introduced into several African countries, mainly for aquaculture, and has had demonstrable impacts where it has escaped captivity. In South Africa, the documentation of this species in farm dams near Dullstroom and the adjacent Crocodile River in 1988 resulted in an eradication attempt in 1994, with unknown results. 2. In order to evaluate the status of P. clarkii in South Africa, dams on the previously invaded farm and the Crocodile River were sampled four times between December 2015 and June 2016 using visual surveys, trapping, dipnetting and electrofishing. This yielded a single reproductively active male P. clarkii from one of the farm ponds, while many other native aquatic species were found in high numbers. 3. It is clear from this study that P. clarkii was not eradicated in South Africa and that individuals have been surviving in the wild (i.e. outside captivity or cultivation) for at least 28 years in the location where it was introduced. Containment and eradication of the species are proposed as management actions, which have major importance in preventing undesirable further spread or translocation of this species into new aquatic environments in South Africa. 相似文献

The objective of this study was to simplify two-step addition of cryoprotectant for vitrification of bovine embryos by developing a one-step procedure. Survival was calculated as a percentage of non-vitrified controls developed from the same batch of oocytes. In experiment 1, bovine blastocysts were vitrified following one- or two-step addition of cryoprotectant. Exposure of embryos to cryoprotectant in one-step resulted in survival rates not significantly lower (p > 0.1) than those obtained by two-step addition (85% vs 98%, respectively). Based on these results, experiments 2-4 were designed to test one-step addition of cryoprotectant more rigorously. Experiment 2 exposed day 7 blastocysts to 6, 7 or 8 M ethylene glycol for 2.5 or 3.5 min. At 24 h post-vitrification, survival of embryos was similar, irrespective of ethylene glycol concentration or exposure time (6 M 38%, 7 M 51%, 8 M 59%; 2.5 min 54%, 3.5 min 45%). In experiment 3, blastocysts were exposed to 7 M ethylene glycol for shorter times (30 or 60 s); 30 s exposure resulted in decreased survival (8% vs 31%, p < 0.05). Experiment 4 concerned one-step addition of cryoprotectant to day 6 bovine morulae, exposed to 7 M ethylene glycol for 1 or 1.5 min. There was no difference in survival between exposure times of 1 or 1.5 min (28% vs 45%, respectively; p > 0.1). It is unclear why many embryos survive vitrification with one-step addition of cryoprotectant, but others do not. Although, one-step addition of cryoprotectant simplifies the vitrification procedure, survival rates were inadequate for routine cryopreservation of in vitro-produced bovine embryos. 相似文献

The objective of this study was to determine the influence of powdered coconut water (ACP‐318®) diluted in high glucose (11.0 mm ) TCM199 in the achievement of nuclear in vitro maturation (IVM) of canine oocytes. Cumulus oocyte complexes (COCs) (n = 632) were randomly allocated into three experimental groups named as group 1 (control group), group 2 (5% powdered coconut water) and group 3 (10% powdered coconut water). The percentage of meiotic resumption (MR) (GVBD to MII) was 39.1% (81/207), 50.2% (108/215) and 46.6% (98/210) for groups 1, 2 and 3 respectively (p < 0.05). There were no differences in MR rates among groups 2 and 3. The medium with ACP‐318® slightly enhanced the nuclear maturation of canine oocytes when a comparison was established with rates of maturation exhibited by oocytes in the experimental group 1 without ACP‐318® (p < 0.05). The results suggest that oocytes’ nuclear morphology integrity and meiosis achievement were positively influenced when exposed to high glucose TCM199 supplemented with 5% powdered coconut water. Further investigation must be performed for a better understanding of powdered coconut water influence in cellular events during IVM of dog oocytes. 相似文献

Objective To determine if an association existed between sickness, mortality and bullers in a western Canadian feedlot.
Design A retrospective epidemiological study.
Animals 78,445 male cattle that entered a 24,000-head feedlot in western Canada from 1991 to 1993.
Procedure Animal health records for bullers were collected and analysed to see if they were at greater risk of sickness and mortality than other steers, and to see if pens with a high prevalence of bullers also had a high prevalence of sickness and mortality.
Results The prevalence of bullers increased with increasing age of cattle on arrival at the feedlot (R = 0.36; P < 0.001). Sickness and mortality decreased with increasing age of cattle on arrival. However, sickness and mortality in bullers relative to other steers actually increased with increasing age on arrival suggesting an interaction existed between sickness and bullers. Bullers were significantly (P < 0.05) more likely to get sick and to die than other steers. In all cases, there was a strong temporal association between sickness and bullers, with sickness and bullers mostly occurring within the first 30 days of the feeding period. On average, pens of cattle with a high prevalence of bullers did not have a correspondingly high prevalence of sickness or mortality.
Conclusion This study suggests that sickness is an effect modifier of dominance behaviour and therefore bullers in feedlot steers. Bullers should always be checked for signs of sickness and treated accordingly. Further research is needed to investigate the effects of sickness on dominance behaviour in pens of feedlot cattle. 相似文献

This study was conducted: (i) to observe the features and levels of blue colour impregnation in morphologically selected immature canine cumulus oocyte complexes (COCs) stained with the brilliant cresyl blue (BCB) dye, as indicators of quality, and integrity of nuclear oocyte chromatin configuration before in vitro maturation (IVM); (ii) to observe the relationship between the influence of serum progesterone (SP) concentrations from ovary donors and BCB staining of immature dog oocytes. The results showed that out of 138 canine COCs, germinal vesicle (GV) stage prevailed in BCB+ oocytes at percentages of 67.4% (60/89), which were statistically higher than those observed in BCB+/− (52.2%; 23/44) and BCB− (20%; 1/5) oocytes (p = 0.023). Oocytes BCB+ were interpreted as those having completed their growth and therefore possessing the capacity to mature and develop in vitro . Ooplasm and cumulus cells (CCs) of canine oocytes were BCB staining independent. Ooplasm blue colour staining reaction varied between grown oocytes, revealing different levels of glucose-6-phosphate dehydrogenase activity among and within oocytes. Additionally, SP profile of ovary donors was not a relevant indicator for selection of oocytes screened with the BCB stain. Similar numbers of high quality oocytes were observed to be BCB+, BCB+/− and BCB− between groups of females with SP varying from 0 to 2.5 ng/ml (n = 5), and those with SP varying from 2.6 to 16.7 ng/ml (n = 4) (p = 0.680). It may be inferred that bitches with low and high SP profiles have grown oocytes in their ovaries, as determined by the BCB absorbance in their ooplasms. 相似文献

Potassium (K) is required in high doses by the banana (Musa sp.) plant and interacts with other nutrient elements in which banana tissues are maintained under in vitro condition as a consequence modifications in the plant metabolism take place mainly in nitrogen (N) compounds, such as proteins, amino acids, and secondary compounds. When K is present in concentrations lower than that required, diamines such as putrescine and poliamines are formed. This metabolic disorder can also be correlated with the presence of different inorganic N forms, such as nitrate (NO3) and ammonium (NH4), and the ratios between both ions as well. In order to follow the physiological performance of the interrelationships, K/putrescine and of the NO3/NH4 ratio in the tissue of banana vitroplantlets, shoot apex of two banana cvs. Nanica and Prata Ana were maintained in modified MS medium in the presence of six different doses of K: 5, 10, 15, 20, 25, and 30 mM. After the period of tissue proliferation the cultures were transferred to rooting media containing the same different K doses. Dry matter, K, putrescine, and spermidine contents and their accumulation were determined in the shoots and roots of the vitroplantlets and in the shoot apex of the expiant donor cultivar as well as the corresponding values for the whole vitroplantlets calculated. The data were statistically analyzed. The contents and accumulations of putrescine and spermidine in banana tissues were enhanced as K concentration decreased in the medium: four times (0.19% of the dry matter) for cv. Nanica and eight times (0.25% of the dry matter) for cv. Prata Ana. This behavior was not only related to the K depletion but to the NO3/NH4 ratio as well. 相似文献
