Near-infrared Fourier transform Raman (FT-Raman) spectroscopy was employed to study the molecular structure of edible zein films/coatings, which were fabricated directly from zein protein. The secondary structure of zein protein was mainly in alpha-helix and remained unaltered during film formation as evidenced by the vibrational modes of amide I at 1656 cm(-1) and amide III at 1274 cm(-1). Raman results indicated that hydrophobic interaction played an important role in the formation of zein film and disulfide bonding might be responsible for the structural stability of zein protein during film formation. To enhance its antimicrobial property, an antimicrobial zein film was manufactured by incorporating zein protein with benzoic acid whose structure was then characterized by FT-Raman. It showed that physical entrapment or hydrophobic interaction was crucial to the incorporation of benzoic acid with zein protein, and the secondary structure of the antimicrobial film was still maintained in alpha-helical form. In addition, FT-Raman exhibits its preference in directly determining the thickness of zein films/coatings. By correlating the Raman intensity ratio of nu(1003) to nu(84) (I(1003/84)) versus the thickness of zein film, a linear relationship with high coefficient (R(2) = 0.9927) was obtained, which was then used pragmatically to determine the thickness of zein coatings on apple. It showed that the FT-Raman result (thickness = 0.27 +/- 0.01 mm) was consistent with that of classical micrometric measurement (thickness = 0.28 +/- 0.02 mm). In order to investigate sparing effects of vitamin C on vitamin E for juvenile hybrid tilapia, fish were fed diets with or without vitamin E each supplemented with either adequate or a high (3× adequate) level of two ascorbate sources,
-ascorbic acid (AA) and its derivative,
-ascorbyl-2-monophosphate-Mg (C2MP-Mg). Each diet was fed to three replicate groups of fish initially weighing a mean value of 0.54±0.01 g/fish in a closed-recirculated rearing system for 8 weeks. Weight gain and feed efficiency of fish fed the vitamin E-deficient diet supplemented with adequate levels of either AA or C2MP-Mg were significantly lower (P<0.05) than fish fed the other diets. Plasma and liver -tocopherol concentrations were lower in fish fed vitamin E-deficient diets regardless of the level or source of supplemented ascorbate. Gill -tocopherolxxxxxxxx concentration was highest in fish fed vitamin E-adequate diets, intermediate in fish fed vitamin E-deficient diets supplemented with a high level of AA or C2MP-Mg and lowest in fish fed a vitamin E-deficient diet supplemented with an adequate level of AA or C2MP-Mg. Fish fed a vitamin E-deficient diet supplemented with an adequate level of AA or C2MP-Mg had higher hepatic thiobarbituric acid-reactive substances than fish fed the other diets. These data suggest that the high supplementation level (3× adequate) of ascorbate could spare vitamin E in diets for hybrid tilapia and that both AA and C2MP-Mg exhibits the same sparing effect. 相似文献

Soil moisture plays a key role in water and energy exchange in the land hydrologic process. Effective soil moisture information can be used for many applications in weather and hydrological forecasting, water resources, and irrigation system management and planning. However, to accurate modeling of soil moisture variation in the soil layer is still very challenging. In this study, in situ and remote sensing information of near-surface soil moisture is assimilated into the Noah land surface model (LSM) to estimate deep-layer soil moisture variation. The sequential Monte Carlo-Particle Filter technique, being well known for capability of modeling high nonlinear and non-Gaussian processes, is applied to assimilate surface soil moisture measurement to the deep layers. The experiments were carried out over several locations over the semi-arid region of the US. Comparing with in situ observations, the assimilation runs show much improved from the control (non-assimilation) runs for estimating both soil moisture and temperature at 5-, 20-, and 50-cm soil depths in the Noah LSM. 相似文献

Recently, the incidence of colon cancer has been rapidly increasing in previously low-risk countries other than the Western world. Since dietary factors are thought to be key components involved in high risk colon cancer, the current trend for colon cancer prevention is toward dietary intervention. To explore if whole oat functions as a chemoprevention agent, an inflammation-related mouse colon cancer model, initiated with 1, 2-dimethylhydrazine (DMH), followed by dextran sodium sulfate (DSS), was performed to evaluate the preventive effect of whole oat containing diets. The result indicated middle and high dose whole oat diets significantly reduced the number of aberrant crypt foci (ACF) as well as colon tumors. Further, human colon carcinoma cells were subcutaneously inoculated into BALB/cAnNg-Foxn1 nude mice to measure the growth inhibition on whole oat diets. Low, middle and high dose whole oat diets significantly decreased the tumor volumes by 13%, 17% and 43%, respectively, indicating a dose dependent inhibitory effect. Meanwhile, 38% and 54% reductions in tumor weights were observed in middle and high dose whole oat diets. Together, the evidence suggests whole oat helps protect against colon cancer development and could be a good chemoprevention agent taken as a daily supplement. 相似文献

ABSTRACT The NSs protein of Watermelon silver mottle virus (WSMoV) was expressed by a Zucchini yellow mosaic virus (ZYMV) vector in squash. The expressed NSs protein with a histidine tag and an additional NIa protease cleavage sequence was isolated by Ni(2+)-NTA resins as a free-form protein and further eluted after sodium dodecyl sulfate-polyacrylamide gel electrophoresis for production of rabbit antiserum and mouse monoclonal antibodies (MAbs). The rabbit antiserum strongly reacted with the NSs crude antigen of WSMoV and weakly reacted with that of a high-temperature-recovered gloxinia isolate (HT-1) of Capsicum chlorosis virus (CaCV), but not with that of Calla lily chlorotic spot virus (CCSV). In contrast, the MAbs reacted strongly with all crude NSs antigens of WSMoV, CaCV, and CCSV. Various deletions of the NSs open reading frame were constructed and expressed by ZYMV vector. Results indicate that all three MAbs target the 89- to 125-amino-acid (aa) region of WSMoV NSs protein. Two indispensable residues of cysteine and lysine were essential for MAbs recognition. Sequence comparison of the deduced MAbs-recognized region with the reported tospoviral NSs proteins revealed the presence of a consensus sequence VRKPGVKNTGCKFTMHNQIFNPN (denoted WNSscon), at the 98- to 120-aa position of NSs proteins, sharing 86 to 100% identities among those of WSMoV, CaCV, CCSV, and Peanut bud necrosis virus. A synthetic WNSscon peptide reacted with the MAbs and verified that the epitopes are present in the 98- to 120-aa region of WSMoV NSs protein. Four food grade additives-sodium ascorbate, sodium bicarbonate, sodium carbonate-10-hydrate, and ferrous sulfate-7-hydrate-were selected as the basic ingredients to formulate the controlled atmosphere agents which could effectively remove oxygen and release carbon dioxide. The mathematical models giving the relationships between the formulations and the responses (oxygen and carbon dioxide contents) were developed using response surface methodology (RSM). Within 8-24 h, the oxygen and carbon dioxide contents of all tested formulations could reach constant levels, in the ranges of 2-9% and 0-41%, respectively. Products fermented with lactic acid bacteria may show antihypertensive effects via substances such as angiotensin I-converting enzyme inhibitor (ACEI) and γ-aminobutyric acid (GABA). It was previously found that milk fermented with Lactobacillus paracasei subsp. paracasei NTU 101 (101FM) or Lactobacillus plantarum NTU 102 (102FM) has ACEI and GABA activities. This study aimed to investigate the antihypertensive effects of 101FM and 102FM orally administered to spontaneously hypertensive rats (SHRs). Eight hours after a single oral administration or after 8 weeks of weekly (chronic) administration, 101FM and 102FM significantly decreased systolic and diastolic blood pressures in the SHRs. Microscopic examination of aortic tissue demonstrated that 101FM and 102FM reduced the disorganization of the media layer. These findings suggest that orally administered 101FM and 102FM have antihypertensive effects, possibly via ACEI and GABA activity, in SHRs. Therefore, 101FM and 102FM may be useful ingredients in physiologically functional foods to prevent hypertension. 相似文献

In this study, waterborne polyurethane (WPU) resins consisted of carboxylic acid and side-chained EO (ethylene oxide) functional groups were prepared via a prepolymer ion-hybrid reaction process. After dipping the WPU on fabrics, the dipped fabrics showed promising deodorization properties for both basic and acidic odors and also showed good wash durability because of the presence of those characteristic groups. Basic odors were neutralized via carboxylic acid groups on the WPU, whereas acidic odors were absorbed by the EO groups through hydrogen bonding. Fourier transform infrared spectroscopy (FT-IR) was used to examine the functional groups of carboxylic acid and EO groups, before and after dipping the WPU on the fabrics. The deodorization properties of the dipped fabrics were associated with the content of the functional groups existing on the WPU molecular chain. Scanning electron microscope (SEM) analysis also revealed that the dipped fabrics had a promising wash durability and deodorization properties after 5 times of laundering because WPU particles were still strongly adhered to the fabrics. More side-chain EO groups, more acetic acid molecules were absorbed; while more-COOH groups in polymer chain, more ammonium molecules were absorbed. Infectious pancreatic necrosis virus (IPNV) causes an acute, contagious disease in a number of economically important fish species (Pilcher & Fryer 1980). It is a member of the Birnaviridae (Brown 1986).
