A three-year survey (1997-99) was carried out on organically reared sheep flocks throughout Sweden. The aim was to determine the prevalence and intensity of nematode infections and to establish relationships between sheep management practices and parasite infections. Faecal samples from ewes and lambs were collected from 152 organic flocks around lambing-time and during the grazing-period for analysis. Results were compared with the different management practices that farmers use to prevent parasitism in their flocks. A high proportion of the flocks was infected with nematodes. The most prevalent species were Haemonchus contortus, Teladorsagia circumeincta, Trichostrongylus axei. T. colubriformis and Chabertia ovina and infections progressively increased during summer in lambs grazing on permanent pastures. Severity of parasitic infection in lambs was highly dependent on egg output from the ewes. H. contortus was found in 37% of the flocks, even at latitudes approximating the Polar Circle. Nematodirus battus was recorded for the first time in Sweden during the course of this study. Lambs turned out onto permanent pasture showed higher nematode faecal egg counts (epg) than lambs that had grazed on pastures, which had not carried sheep the previous year. This beneficial effect of lambs grazing non-infected pastures persisted if the ewes were treated with an anthelmintic before turn-out and if the lambs were kept on pastures of low infectivity after weaning. In lambs, the prevalence and the magnitude of their egg counts were higher during autumn in flocks where lambs were slaughtered after 8 months of age, compared with flocks where all lambs were slaughtered before this age. These results will be used in providing advice to farmers of ways to modify their flock management in order to minimise the use of anthelmintics, but at the same time efficiently produce prime lambs.  相似文献

Purpose To study retinal function in cats homozygous and heterozygous for a recessively inherited rod‐cone degeneration. Methods Dark‐adapted electroretinograms (ERGs) were performed on early affected, heterozygous (ophthalmoscopically normal), and clinically normal, nonrelated cats. Responses to blue stimuli over a 3.9‐log unit range were recorded. Results Lower b‐wave amplitudes than normal were observed in heterozygotes and early affected cats. The amplitudes of the heterozygotes took an intermediate position between normal and early affected cats. Normalized amplitude/intensity data suggest a normal dynamic range in carriers. B‐wave implicit times in carriers were comparable to those of normal cats. Conclusions These results show that heterozygotes have an altered retinal function, although they are ophthalmoscopically normal. It is difficult to electrophysiologically differentiate heterozygotes from affected cats with the very early stage of retinal degeneration.  相似文献

Objective To understand how progressive rod cone degeneration due to a mutation in CEP290 affects the pupillary light reflex (PLR) in domestic cats. Animals studied Domestic cats identified as either normal wildtype (WT; n = 6), or homozygous for the rdAc mutation in CEP290 and having early stage retinal degeneration (stage 2, S2; n = 4), or advanced retinal degeneration (S4; n = 6). Methods The effect of light on pupil size was measured over a series of 10‐s pulses of white and chromatic light in cats lightly sedated with medetomidine. Results In WT cats, the PLR was characterized by a pronounced initial constriction that rapidly re‐dilated during the stimulus (pupil escape), to a stable or sustained constriction. There was then a marked constriction at stimulus offset. Each component of the PLR was retained in affected cats, but with progressively reduced irradiance sensitivity from early to advanced retinal disease. Conclusions The PLR of cats had multiple phases, with a remarkably high‐amplitude ‘paradoxical’ off‐constriction even in the absence of retinal disease. In rdAc cats, reduced irradiance sensitivity was consistent with progressive loss of rod and cone function. Based on previously characterized retinal pathology, this suggests the visual streak of the retina has a proportionally large contribution to PLR input. These findings support the hypothesis that the efficacy of planned therapeutic trials can be determined by careful evaluation of the PLR in cats.  相似文献

对celB基因标记法检测苜蓿根瘤菌竞争性的可行性进行了研究。通过供体大肠杆菌HAMBI2356与根瘤菌接合的方法成功地将celB基因导入到4株苜蓿根瘤菌株Y611、BB1811、BB2211和WZ622中,然后对标记基因可能会对标记菌株产生的影响进行了一系列检测,结果表明标记基因在标记菌株内能稳定传代,而且对标记菌株的生长、竞争性及有效性都没有显著影响,从而证明celB基因标记技术可以作为一种简便、直观而又有效的检测手段用于追踪根瘤菌株在土壤中的竞争性。  相似文献

The total thermal conductivity lambda of resorcinol-formaldehyde aerogel monoliths has been measured as a function of density rho in the range from rho = 80 to 300 kilograms per cubic meter. A record-low conductivity value in air at 300 K of lambda approximately 0.012 watt per meter per kelvin was found for rho approximately 157 kilograms per cubic meter. Caloric measurements under variation of gas pressure as well as spectral infrared transmission measurements allowed the determination of solid conductivity, gaseous conductivity, and radiative conductivity as a function of density. The development of such low conductivity materials is of great interest with respect to the substitution of environmentally harmful insulating foams made from chlorofluorocarbons.  相似文献

An unexpected biochemical strategy for chain initiation is described for the loading module of the polyketide synthase of curacin A, an anticancer lead derived from the marine cyanobacterium Lyngbya majuscula. A central GCN5-related N-acetyltransferase (GNAT) domain bears bifunctional decarboxylase/S-acetyltransferase activity, both unprecedented for the GNAT superfamily. A CurA loading tridomain, consisting of an adaptor domain, the GNAT domain, and an acyl carrier protein, was assessed biochemically, revealing that a domain showing homology to GNAT (GNAT(L)) catalyzes (i) decarboxylation of malonyl-coenzyme A (malonyl-CoA) to acetyl-CoA and (ii) direct S-acetyl transfer from acetyl-CoA to load an adjacent acyl carrier protein domain (ACP(L)). Moreover, the N-terminal adapter domain was shown to facilitate acetyl-group transfer. Crystal structures of GNAT(L) were solved at 1.95 angstroms (ligand-free form) and 2.75 angstroms (acyl-CoA complex), showing distinct substrate tunnels for acyl-CoA and holo-ACP(L) binding. Modeling and site-directed mutagenesis experiments demonstrated that histidine-389 and threonine-355, at the convergence of the CoA and ACP tunnels, participate in malonyl-CoA decarboxylation but not in acetyl-group transfer. Decarboxylation precedes acetyl-group transfer, leading to acetyl-ACP(L) as the key curacin A starter unit.  相似文献

Point-of-care analyzers (POCAs) for quantitative assessment of bile acids (BAs) are scarce in veterinary medicine. We evaluated the Fuji Dri-Chem Immuno AU10V analyzer and v-BA test kit (Fujifilm) for detection of feline and canine total serum BA concentration. Results were compared with a 5th-generation assay as reference method and a 3rd-generation assay, both run on a bench-top analyzer. Analytical performance was assessed at 3 different concentration ranges, and with interferences. For method comparison, samples of 60 healthy and diseased cats and 64 dogs were included. Linearity was demonstrated for a BA concentration up to 130 µmol/L in cats (r = 0.99) and 110 µmol/L in dogs (r = 0.99). The analyzer showed high precision near the lower limit of quantification of 2 µmol/L reported by the manufacturer. Intra- and inter-assay coefficients of variation were < 5% for both species and all concentrations. Interferences were observed for bilirubin (800 mg/L) and lipid (4 g/L). There was excellent correlation with the reference method for feline (rs = 0.98) and canine samples (rs = 0.97), with proportional biases of 6.7% and −1.3%, respectively. However, a large bias (44.1%) was noted when the POCA was compared to the 3rd-generation assay. Total observed error was less than total allowable error at the 3 concentrations. The POCA reliably detected feline and canine BA in clinically relevant concentrations.  相似文献

This study was based on laboratory incubations of mor humus from two N fertilized stands of Norway spruce in Sweden (Skogaby and Stråsan), which had received repeated N additions (100 kg N ha−1 yr−1 as (NH4)2SO4 at Skogaby and 35, 73 and 108 kg ha−1 yr−1 as NH4NO3 at Stråsan) during 8 and 24-29 years, respectively. The aim was to investigate long-term N effects on the mineralization of C and production of DOC. Mor humus (Oe and Oa) was incubated in columns at 20 °C for 49 days. Columns were leached once a week with artificial throughfall solution, which was analyzed for DOC, total N, NH4+-N and NO3-N. Prior to each leaching event, CO2 evolution from the columns was determined. C-to-N ratios in the N-treated Oe layers at Stråsan (21-24) and Skogaby (24) were significantly lower than those of the controls (Stråsan, 32; Skogaby, 28). The cumulative amount of CO2-C showed a significant treatment effect in the Oe layer at Skogaby, i.e. 18 and 29 mg C g−1 C in the N treatment and control, respectively. At Stråsan, the cumulative CO2-C was significantly lower in the N3 treatment compared to the control in both layers (33 compared to 74 mg C g−1 C in the Oe layer and 16 compared to 35 mg C g−1 C in the Oa layer). Neither the DOC nor the DON production showed any significant treatment effects at the two sites. However, DOC was lower in the fertilized Oe layer at Skogaby throughout the incubation. The leaching of DON was highest in the Oe layers at both sites, and DON increased with time at Skogaby while there was a decreasing trend at Stråsan. The DOC-to-DON ratio tended to be lower in the fertilized Oe layers at both sites. The NH4+ leaching at Skogaby decreased in the N-treated Oe and Oa layers. At Stråsan, NH4+ from the Oe layer increased in N2 and control. The NO3 leaching was low and constant in both Skogaby layers. At Stråsan, NO3 increased in the Oe layer of N1. Cumulative CO2 was positively correlated to C-to-N ratio (r2=0.71,p<0.01) and to cumulative DOC (r2=0.63,p<0.05) in the Oe layer at Stråsan. Our conclusion was that long-term N additions caused decreased C-to-N ratios and decreased CO2 evolution rates. The correlation between CO2 and C-to-N ratio in the Oe layers at Stråsan may be due to a changed quality of the fertilized forest floor material and presence of more N efficient microorganisms.  相似文献
