Heart rate variability (HRV) may be useful for objective assessment of stress and pain in animals. The objective of this study was to describe the effect of anesthesia and surgery on HRV in dogs. We hypothesized that surgery would decrease HRV to a greater degree and for a longer duration than anesthesia alone. Four healthy male dogs (29 ± 2 kg) were instrumented for ambulatory ECG monitoring. Continuous ECG data was obtained for 1 day prior to, and 6 days following anesthesia alone (ANES) or anesthesia plus unilateral stifle arthrotomy (ANSX). The anesthetic protocol included xylazine (0.5 mg kg–1 IM), morphine (0.5 mg kg–1 IM), atropine (0.04 mg kg–1 IM), thiopental (10 mg kg–1 IV) and isoflurane in oxygen. A single dose of morphine (0.5 mg kg–1 IM) was administered at extubation. Time domain analysis of HRV was performed on 5 minutes epochs of artefact‐ and arrhythmia‐free ECG data obtained at 12 noon and 12 midnight on each of the seven experimental days. Mean RR interval, standard deviation of normal R‐R intervals (SDNN), and the standard deviation of successive differences in RR intervals (SDSD) were compared to baseline for ANES and ANSX. Pain scores obtained during the day were also evaluated. Significance was set at p < 0.01. There were no differences between groups for any baseline data. Mean RR interval did not differ from baseline on days 1–6 in ANES or ANSX. SDNN and SDSD values at noon were not different from baseline on days 1–6 in ANES or ANSX. At midnight on days 1 and 2, SDNN was significantly decreased from baseline in ANSX, and on day 1 a significant difference between groups existed. ANSX values of SDSD at midnight were significantly decreased from baseline and ANES on day 1. Pain scores for ANSX were significantly greater than baseline on days 1–3, and different from ANES on days 1–5. These results suggest that HRV is decreased following anesthesia plus surgery, and that changes in HRV may be associated with pain.  相似文献

Surveys for lobster larvae in offshore waters of the north‐eastern Gulf of Maine in 1983, 1987 and 1989 confirm that local hatching occurs mainly at depths <100 m over the banks, including Georges and Browns Banks. Detailed studies in the vicinity of Georges Bank in late July of both 1987 and 1989 indicate that the first and second moult stages were located primarily over the bank whereas stages III and IV lobster were collected both over and off the bank. At times stage IV lobster were more abundant off the bank than over it. The condition of stage III and IV lobster, as measured by a lipid index, was better off than over Georges Bank in 1988 and 1989 indicating a possible physiological advantage to being off the bank. In addition, the higher surface temperatures off Georges Bank would shorten larval development time to settlement. To determine the probable hatch sites of stage IV lobster collected off of Browns Bank in 1983 and off of Georges in 1987 and 1989, a 3‐D circulation model of the Gulf of Maine was used to simulate larval lobster drift backwards in time. In all cases, areas off Cape Cod, MA, and off Penobscot Bay, ME were suggested as the source of the larvae, although most of the larval trajectories never reached these near‐shore waters that are well‐known, larval hatching areas. The model‐projected larval release times match most closely the observed inshore hatch off Massachusetts but model uncertainties mean that coastal Maine cannot be ruled out as a source. Georges Bank is also a potential source because the present model does not take into account short‐term wind events, off‐bank eddy transport or the possibility of directed off‐bank larval swimming. Examination of weather records prior to and during our 1988 and 1989 sampling periods indicates that winds were not of sufficient intensity and duration to induce larval transport off Georges Bank. The shedding of eddies from the northern flank of Georges Bank into the Gulf of Maine are a relatively common phenomenon during summer but not enough is known about them to evaluate their contribution to possible cross‐bank transport of lobster larvae. Directed larval swimming is another possible source for the stage IV lobster found near Georges Bank. Plankton distributions across the northern frontal zone of Georges Bank in 1988 were used as proxies for the scarce larval lobsters. The more surface distribution of the microplankton, in particular, supports the possibility that wind and eddy events may be important in the transport of stage III and IV lobsters off of Georges Bank. Further studies are needed to evaluate these possible additional sources of advanced stage lobster larvae found off of the offshore banks.  相似文献

AIM: To validate the use of para-aminohippuric acid (PAH) as a marker for measuring blood flow in wethers consuming a mixed diet of locoweed and blue grama hay.

METHODS: Fourteen sheep, stratified by bodyweight (BW), were assigned to one of three treatments: 0.8 mg swainsonine (SW)/kg BW (HI), 0.2 mg SW/kg BW (LO), and no SW (Control). Sheep were fed various ratios of locoweed and blue grama hay to deliver SW treatments, for 28 days prior to infusion of PAH. Concentrations of SW and activities of alkaline phosphatase (Alk-P) and aspartate aminotransferase (AST) in serum were measured to confirm exposure to SW and subclinical intoxication. A single 20-ml injection of 5% PAH was delivered into the jugular vein after subclinical intoxication had been achieved. Blood samples were collected and serum analysed for PAH immediately prior to injection, then every 5 min from 5–30 min, and every 10 min from 30–60 min, following injection of PAH.

RESULTS: Effective delivery of SW was evident from the greater concentrations of SW measured in the serum of HI compared with LO animals (p<0.05). No significant differences were detected in the rate of elimination (range 0.097–0.108 L/min), elimination half-life (range 6.62–7.24 min), apparent volume of distribution for the central compartment (range 7.14–9.72 L), and clearance (range 0.73–0.92 L/min) of PAH, between treatments.

CONCLUSIONS: Subclinical intoxication with SW did not affect the pharmacokinetics of PAH. Thus, use of downstream dilution of PAH is a valid method to determine the rate of blood flow in nutrient flux experiments that involve consumption of locoweed.  相似文献

Endothelial seeding of vascular grafts has been shown to decrease graft thrombogenicity and prolong longevity when implanted in vivo. Previous studies have utilized anatomic grafts to study endothelialization and healing. Anatomic thoracoabdominal grafts do not allow for sequential biopsy for evaluation of individual grafts nor do they approximate the environment for long bypass grafts used in limb salvage. This study evaluated the use of an extra-anatomic aortic bypass graft to assess the healing of endothelial cell seeded expanded polytetrafluoroethylene (ePTFE). Radionuclide angiography was used to evaluate graft patency and quantify blood flow through the graft. Dogs underwent placement of an extra-anatomic 60 cm long, 8 mm internal diameter, graft seeded with autologous endothelium. Grafts were biopsied from 2 weeks up to 1 year. Radionuclide studies were performed postimplantation and following each graft biopsy. Graft placement and biopsies were well tolerated in all dogs. Biopsied segments of graft allowed for sequential studies of the healing of implanted grafts by scanning electron and light microscopy. Flow through the implanted graft was close to 50% of the total caudal abdominal aortic flow. No significant difference in graft flow was noted either between animals or overtime.  相似文献

The purpose of this study was to evaluate the effect of cooling ovarian tissue on pig pre-antral follicles. Ovaries were maintained in saline solution (0.9%) at 4 or 20 degrees C for 6, 12 or 18 h. After storage, pre-antral follicles were morphologically evaluated. While primordial follicles were not affected by the storage, the percentage of morphologically normal growing follicles was significantly reduced in ovarian tissue stored at 20 degrees C for 12 or 18 h. To test the viability of stored follicles, growing follicles isolated from ovaries stored at 4 degrees C for 18 h and at 20 degrees C for 6 h were cultured for 3 days. Follicles stored in either condition presented the same growth pattern in vitro as fresh follicles. We conclude that storage of pig ovaries at 4 degrees C for up to 18 h or at 20 degrees C for up to 6 h does not affect the morphology of growing follicles or their ability to grow in vitro.  相似文献

Abstract

AIM: To determine the amount of ergovaline and lysergic acid retained or excreted by geldings fed endophyte-infected seed containing known concentrations of these alkaloids, and the effects of exposure time on clinical expression of toxicosis.

METHODS: Mature geldings (n=10) received diets containing either endophyte-free (E-) or endophyte-infected (E+) tall fescue seed during three experimental phases. The first phase (Days ?14 to ?1) was an adaptation phase, to allow all horses to adapt to a diet containing E- tall fescue seed. The second (Days 0 to 3) was the initial exposure phase to E+ tall fescue seed, used for the delivery of ergovaline and lysergic acid at 0.5 and 0.3 mg/kg of diet, respectively, to test the initial effects of exposure on routes and amounts of elimination of alkaloid. During this phase, half the geldings were exposed to an E+ diet while the rest served as controls by remaining on the E- diet. Once assigned to treatments, geldings remained on the same diet through the third phase (Days 4 to 21), which served as the extended exposure phase. Total outputs of faeces and urine were collected within each phase, to determine retention of ergovaline and lysergic acid and nutrient digestibility. Serum was collected weekly and analysed for activities of enzymes and concentrations of prolactin. Bodyweights (BW) and rectal temperatures were recorded weekly.

RESULTS: BW, rectal temperature, enzyme activities and concentrations of prolactin in serum, and nutrient digestibility were not affected by treatment. Total intake of ergovaline by geldings on the E+ diet was 3.5 and 3.6 (SE 0.20) mg/day, and 2.1 and 2.3 (SE 0.11) mg/day were not accounted for in initial and extended phases, respectively. Lysergic acid was excreted in the urine (4.0 and 4.9 (SE 0.97) mg/day) and faeces (2.5 and 2.7 (SE 0.35) mg/day) at greater amounts than that consumed (2.0 and 1.9 (SE 0.09) mg/day) during the initial and extended exposure phases, respectively. Animals exposed to E+ seed for a period of 20 days appeared to excrete more (1.5 vs 1.2 mg/day; SE 0.08; p=0.03) ergovaline in the faeces than those exposed for only 4 days.

CONCLUSIONS: Exposure time to the ergot alkaloids had a limited effect on the route of elimination or the amounts of ergovaline or lysergic acid excreted by horses. The primary alkaloid excreted was lysergic acid, and urine was the major route of elimination. These data will aid future research to improve animals′ tolerance to toxic endophyte-infected tall fescue.  相似文献

Recordings of the middle latency of the auditory evoked potential (MLAEP) were made in eight conscious ponies. These traces were compared to those made under halothane anaesthesia with and without paralysis of the skeletal muscles. Recordings were also made from percutaneous electrodes placed along the neck with the same stimulus used for the auditory evoked potentials. The results of these experiments were used to deduce the origin of latencies in the auditory evoked potential occurring between 10 and 25 ms after the stimulus. The MLAEP was found to contain two positive peaks between the latencies of 10 and 25 ms. The first of these two peaks was not abolished by halothane anaesthesia or muscle paralysis. The second of these two peaks was abolished by halothane anaesthesia in all but one animal. In this animal the second peak was abolished by muscle paralysis. No peaks of corresponding latency were recorded from the percutaneous electrodes except from one electrode position at the caudal neck in one pony. The first peak of the middle latency auditory evoked potential seen in conscious ponies appeared to be of central nervous orign. The second peak appeared to be of muscular origin, possibly from the external auditory muscles. The second peak may be analogous to the post-auricular waveform described in man.  相似文献

Femoral head and neck excision was performed in 169 dogs over a 7 year period. A written questionnaire completed by 56 owners, evaluating a total of 72 joints, was the basis for retrospective evaluation of the efficacy of the procedures used. There were four surgical groups: femoral head and neck excision (FHNE) in dogs with body weights over 14 kg, the same procedure in dogs with body weights less than 14 kg (SMALL), femoral head and neck excision with muscle interposition (MI), and the wedge resection technique (WRT). Although differences in results were noted, there were no statistically significant differences between any of the groups for the percentage of leg use during normal activity (p greater than 0.05), mean postoperative time until leg use, use of the leg or hopping while running, or lameness with exercise (p greater than 0.10). The results of this study suggest that there is little advantage in performing the more time-consuming and traumatic muscle interposition and wedge resection procedures.  相似文献

Résumé— Une dermite pyogranulomateuse provoquée par le protozoaire Neospora caninum a été diagnostiquée chez un Golden Retriever de 12 ans. Le nombreux nodules fistulisés étaient localisés au niveau de la tête et du thorax. De nombreux tachyzoites de N. caninum ont été retrouvés dans les biopsies des nodules et le diagnostic a été confirmé pas immunohistologie et examen en microscopie électronique. Le chien avait un titre de sérologie Neospora caninum de 3200 par immunofluorescence indirecte. Après un traitement à base de clindamycine pendant 45 jours, les lésions cutanées ont disparu. Le chien est probablement mort à cause d'un lymphosarcome. Il existait aussi une infection latente àToxoplasma gondii. les Neospora caninum n'ont pas pu être retrouvés par des techniques biologiques ni en culture ou par inoculation de souris à partir de prélèvements nécropsiques. Seuls des tachyzoites dégenérés ont pu être observés histologiquement. Ces observations montrent que la néosporose peut être envisagée dans le diagnostic différentiel des dermites pyogranulomateuses du chien et que la clindamycine est un médicament efficace pour traiter la néosporoe canine. El animal presentaba varios nódulos en la piel de la cabeza y tórax. Se observaron numerosos taquizoitos de N. caninum en los cortes histológicos de tejido obtenido mediante biopsia de dichos nódulos y el diagnóstico fue confirmado por tinción inmunohistológica y por microscopia electrónica. El perro mostró un titulo de anticuerpos contra N. caninum de 1:3,200 en la prueba de fluorescencia indirecta. Las lesiones cutáneas se resolvieron tras un tratamiento con hidroclorido de clindamicina durante 45 dias. El perro murió posteriormente a causa de un linfoma y presentaba también una infestación latente por Toxoplasma gondii. No se pudo demostrar la presencia de Neospora caninum mediante bioensayos en cultivos celulares ni en ratones inoculados con tejido canino obtenido en la necrospia. Tan solo se pudieron observar taquizoitos degenerados de N. caninum en tejido cutáneo obtenido en la necrospia. Estos hallazgos indican que se debe incluir neosporosis en el diagnóstico diferencial de dermatitis piogranulomatosas en el perro y que la clindamicina puede ser un fármaco eficaz para el tratamiento de la neosporosis cutánea. [Canine cutaneous neosporosis: clinical improvement with clindamycin (Neosporosis cutánea canina: mejora clinica con clindamicina). Abstract— Pyogranulomatous dermatitis caused by the protozoan parasite Neospora caninum was diagnosed in a 12-year-old Golden Retriever dog. Multiple draining nodules were located in the skin of the head and thorax. Numerous tachyzoites of N. caninum were found in histologic sections of the biopsy tissue from the cutaneous nodules and the diagnosis was confirmed by immunohistochemieal staining and by electron microscopic examination. The dog had a 1:3200 serum antibody titer to N. caninum in the indirect fluorescent antibody test. The cutaneous lesions resolved after a 45-day treatment with clindamycin hydrochloride. The dog eventually died because of lymphosarcoma and also had a latent infection with Toxoplasma gondii. Neospora caninum could not be demonstrated by bioassays in cell culture or mice inoculated with canine tissue obtained at necropsy. Only degenerating N. caninum tachyzoites were seen in skin tissue taken at necropsy. These observations indicate that neosporosis should be considered in the differential diagnosis of pyogranulomatous dermatitis in dogs and that clindamycin may be an effective drug for treating cutaneous neosporosis.  相似文献
