Oxidative stress has been recognized as an important factor in the pathophysiology of preeclampsia. It has been reported that the expression of xanthine oxidase (XO) in the cytotrophoblast and plasma hydrogen peroxide (H2O2) level are significantly higher in preeclamptics than in control women. The aim of this study was to clarify the biological influence of reactive oxygen species (ROS) produced by XO on extravillous trophoblast (EVT) cells. TCL1 cells, a human immortalized EVT cell line, were incubated with xanthine and XO (X/XO). We then measured the cell number, urate level of the culture media and the apoptotic cell ratio. Similar experiments were performed with additional administration of allopurinol, catalase, L-NAME or D-NAME, and with administration of H2O2 in substitution for X/XO. We assessed the effects of H2O2 on invasion ability, tube-like formation and protein expression of HIF1A and ITGAV of TCL1. Finally, the apoptotic cell ratio using primary cultured trophoblasts was measured following exposure to H2O2. X/XO decreased the relative cell number and increased the urate level and apoptotic cell ratio significantly. Elevation of the urate level and apoptotic cell ratio was attenuated by allopurinol and catalase, respectively. L-NAME and D-NAME had no influence on these effects. H2O2 also decreased the relative cell number. Pretreatment with H2O2 significantly inhibited the invasion ability, tube-like formation and HIF1A and ITGAV of TCL1. H2O2 also induced apoptosis in primary cultured trophoblasts. In conclusion, ROS produced by XO induced apoptosis and affected EVT function including invasion and differentiation.  相似文献

Cattle exhibit a range of carcass defects, including blood splash (BLS), intramuscular edema (INE), muscle steatosis (MUS), bruising (BR), trim loss (TRL) and others (OTH). These defects lower the carcass value and can result in significant economic loss to producers. We estimated the incidence, relationship with inbreeding coefficients and genetic parameters of carcass defects in Japanese Black cattle using 561 619 carcass records from Kagoshima, Japan during April 1988 through March 2011. The defect incidence ranged from 0.22% for TRL to 5.73% for BR. The incidence of MUS and BR increased from 1.21% to 6.57% and from 1.06% to 9.31%, respectively. The incidence of INE peaked at 7.44% in 1999 and decreased thereafter. We observed a positive linear relationship between the defect incidence and the inbreeding coefficients in MUS, BR and TRL (P < 0.01). The heritabilities estimated by univariate animal model with Gibbs sampling for BLS, INE, MUS, BR and TRL were 0.24, 0.06, 0.18, 0.05 and 0.02, respectively. The contribution of farm variance to phenotypic variance was negligible (0.01 to 0.04). Significant genetic correlations of TRL were estimated with MUS (0.63) and BR (0.63). Our results suggest that genetic factors contribute to the incidence of BLS and MUS.  相似文献

Mass mortality events are ubiquitous in nature and can be caused by, for example, diseases, extreme weather and human perturbations such as contamination. Despite being prevalent and rising globally, how mass mortality in early life causes population-level effects such as reduced total population biomass, is not fully explored. In particular for fish, mass mortality affecting early life may be dampened by compensatory density-dependent processes. However, due to large variations in year-class strength, potentially caused by density-independent variability in survival, the impact at the population level may be high in certain years. We quantify population-level impacts at two levels of mass mortality (50% and 99% additional mortality) during early life across 40 fish species using age-structured population dynamics models. The findings from these species-specific models are further supported by an analysis of detailed stock-specific models for three of the species. We find that population impacts are highly variable between years and species. Short-lived species that exhibit a low degree of compensatory density dependence and high interannual variation in survival experience the strongest impacts at the population level. These quantitative and general relationships allow predicting the range of potential impacts of mass mortality events on species based on their life history. This is critical considering that the frequency and severity of mass mortality events are increasing worldwide.  相似文献

Herein, we developed a triploidization method using spawned eggs collected immediately after spawning of eastern little tuna (ELT), Euthynnus affinis. ELT broodstock induced the spawning by hormonal treatment in the tank, with the resulting spawned eggs being used for the triploidy induction. Under optimal conditions, the mean ± SEM triploidization and hatching rates were 97.2 ± 2.8% and 84.5 ± 10.3%, respectively. Although triploid ELT showed growth performance equivalent to that of diploids, the triploids died at a higher rate than the diploids during 2–4 weeks post‐hatching when triploids and diploids were reared in the same tank. Therefore, we propose that it would be necessary in a practical operation to use triploid‐only ELT seedlings to avoid selective cannibalism by the diploids. The ELT triploids exhibited an all‐female phenotype. Because previous studies have reported that female triploids show a greater probability of sterility than male triploids, this characteristic could be a major advantage. Since this triploidization method, using spawned eggs, can be performed without handling the broodstock, it is possible to avoid the physical damage caused by the process of artificial insemination, making it possible to repeatedly produce triploid populations without valuable broodstock loss. Thus, we have developed an efficient method to produce ELT triploids, although further study is essential to evaluate sterility of the triploid ELT.  相似文献

ABSTRACT:   Larval false clown anemonefish of about 12 h after hatching were fed newly-hatched brine shrimp nauplii in natural (control) and copper-added seawater (40–640 µg–Cu/L) at 26.5 ± 0.5°C for 14 days. Survival rate of the fish increased with increasing copper concentrations up to 160 µg–Cu/L. The rates at 80 and 160 µg–Cu/L were 65 and 80%, respectively, and were significantly higher than that of the control (30%). A positive effect of copper addition on the survival rate was also observed in the other rearing experiment with the fish from seven different spawning chances. Growth rate of the survival fish was not affected by copper concentrations. The copper concentration of the survived fish increased with increasing copper concentrations in the rearing water.  相似文献

The blue shark (Prionace glauca) is the most frequently captured shark in pelagic oceanic fisheries, especially pelagic longlines targeting swordfish and/or tunas. As part of cooperative scientific efforts for fisheries and biological data collection, information from fishery observers, scientific projects and surveys, and from recreational fisheries from several nations in the Atlantic and Indian Oceans was compiled. Data sets included information on location, size and sex, in a total of 478,220 blue shark records collected between 1966 and 2014. Sizes ranged from 36 to 394 cm fork length. Considerable variability was observed in the size distribution by region and season in both oceans. Larger blue sharks tend to occur in equatorial and tropical regions, and smaller specimens in higher latitudes in temperate waters. Differences in sex ratios were also detected spatially and seasonally. Nursery areas in the Atlantic seem to occur in the temperate south‐east off South Africa and Namibia, in the south‐west off southern Brazil and Uruguay, and in the north‐east off the Iberian Peninsula and the Azores. Parturition may occur in the tropical north‐east off West Africa. In the Indian Ocean, nursery areas also seem to occur in temperate waters, especially in the south‐west Indian Ocean off South Africa, and in the south‐east off south‐western Australia. The distributional patterns presented in this study provide a better understanding of how blue sharks segregate by size and sex, spatially and temporally, and improve the scientific advice to help adopt more informed and efficient management and conservation measures for this cosmopolitan species.  相似文献

Effects of daily feeding frequency, water temperature, and stocking density on the growth of tiger puffer, Takifugu rubripes, fry were examined to develop effective techniques to produce tiger puffer in a closed recirculation system. Fish of 4, 14, and 180 g in initial body weight were fed commercial pellet diets once to five times a day to apparent satiation each by hand for 8 or 12 wk at 20 C. Daily feeding frequency did not affect the growth of 14‐ and 180‐g‐size fish. However, the daily feed consumption and weight gain of the 4‐g‐size fish fed three and five times daily were significantly higher than those of fish fed once daily (P < 0.05). Fish of 4 and 50 g in initial body weight were reared with the pellet diet at 15–30 C for 8 wk. The weight gain of fish increased with increasing water temperature up to 25 C and decreased drastically at 30 C for both sizes. Similar trends were observed for feed efficiency, although 4‐g fish had highest efficiency at 20 C. Effects of stocking density on growth were examined with fish of 8, 13, and 100 g in initial body weight. Fish were reared with the pellet diet for 8 or 16 wk at 20 C. Fish were placed in floating net cages in the culture tank, and the stocking density was determined based on the total weight of fish and volume of the net cage. Fish of 8 g in body weight grew up to 35–36 g during the 8‐wk rearing period independent of the stocking density of 8, 15, and 31 kg/m3 at the end of rearing. Final biomass per cage reached 32, 60, and 115 kg/m3 for 13‐g‐size fish, and 10, 18, and 35 kg/m3 for 100‐g‐size fish, and the growth of the fish tended to decrease with increasing stocking density for both sizes.  相似文献

The ecdysone receptor (EcR) possesses the remarkable capacity to adapt structurally to different types of ligands. EcR binds ecdysteroids, including 20-hydroxyecdysone (20E), as well as nonsteroidal synthetic agonists such as insecticidal dibenzoylhydrazines (DBHs). Here, we report the crystal structures of the ligand-binding domains of Heliothis virescens EcR/USP bound to the DBH agonist BYI09181 and to the imidazole-type compound BYI08346. The region delineated by helices H7 and H10 opens up to tightly fit a phenyl ring of the ligands to an extent that depends on the bulkiness of ring substituent. In the structure of 20E-bound EcR, this part of the ligand-binding pocket (LBP) contains a channel filled by water molecules that form an intricate hydrogen bond network between 20E and LBP. The water channel present in the nuclear receptor bound to its natural hormone acts as a critical molecular adaptation spring used to accommodate synthetic agonists inside its binding cavity.  相似文献

Benzoylphenylureas (BPUs) were discovered as novel type insecticides about a half century ago; many analogs have been launched as insecticides and acaricides. BPUs are known to inhibit chitin synthesis in insects and other arthropods, but they have no effect against microorganisms such as fungi. We designed new chitin synthesis inhibitors based on the hypothesis that biomolecules that play important roles in cellulose and chitin biosynthesis are similar. In the full automatic modeling system (FAMS), the cellulose synthase was selected as a template three-dimensional structure. Thus, we focused on the structure of cellulose synthase inhibitor, isoxaben, to develop new chemistry. The 1,1-diethylethyl [-C(CH3)(CH2CH3)2] group of isoxaben was changed to a 4-substituted phenyl group bearing Cl, Et, or Ph. These compounds significantly inhibited chitin synthesis in the cultured integument of the rice stem borer Chilo suppressalis. The activity of the 4-ethylphenyl analog was enhanced 30-fold by adding piperonyl butoxide to the culture medium.  相似文献
