The aim of this study was to assess clinical signs and altered pulmonary cell expression of cytokines related to eosinophil kinetics in horses with pulmonary eosinophilia. Pulmonary eosinophilia was detected by bronchoalveolar lavage (BAL) in a group of standardbreds in training. Horses had detailed clinical examination, bronchoscopy, endobronchial biopsy and BAL on three occasions at approximately 6 month intervals. During the second sampling period BAL eosinophils were significantly elevated (p>0.010), with five horses having from 5% to 37% eosinophils in BAL. Neither detailed clinical examination parameters nor gene expression of IL-4 and IL-5 mRNA (real-time-PCR) were associated with BAL eosinophilia. Pulmonary eosinophilia abated without treatment apart from deworming. It appears that pronounced lung eosinophilia in horses can be transient, abate without specific treatment, and in this instance, lack correlation to upregulation of expression of either IL-4 or IL-5.  相似文献

Centrifugation of a homogenate of white matter, in a solution of buffered sucrose containing salt, produces a floating layer of myelinated axons. When these are suspended in hypotonic buffer, the mnyelin swells and strips away from the axon. Axons are then separated from the myelin by centrifugation. The resulting preparation consists of a variable population of processes with lengths up to 200 micrometers and diameters between 0.3 and 5.0 micrometers. The axons contain neurofilaments and mitochondria, although no axolemma or neurotubules are evident. The preparation contains cerebroside and sulfatide, yet is essentially free of myelin.  相似文献

Measles virus generally produces acute illness. Rarely, however, persistent infection of brain cells occurs, resulting in a chronic and fatal neurological disease, subacute sclerosing panencephalitis (SSPE). Evidence indicates that expression of the measles virus matrix protein is selectively restricted in this persistent infection, but the mechanism underlying this restriction has not been identified. Defective translation of matrix messenger RNA has been described in one SSPE cell line. This report presents evidence that in a different SSPE tissue culture cell line IP-3-Ca, the matrix protein is synthesized but fails to accumulate. A general scheme is proposed to reconcile the different levels at which restriction of matrix protein has been observed.  相似文献

Precision irrigation involves the accurate and precise application of water to meet the specific requirements of individual plants or management units and minimize adverse environmental impact. Under precision irrigation applications, water and associated solute movement will vary spatially within the root zone and excess water application will not necessarily result in deep drainage and leaching of salt below the root zone. This paper estimates that 10% of the irrigated land area (producing as much as 40% of the total annual revenue from irrigated land) could be adversely affected by root zone salinity resulting from the adoption of precision irrigation within Australia. The cost of increases in root zone salinisation due to inappropriate irrigation management in the Murray and Murrumbidgee irrigation areas was estimated at AUD 245 million (in 2000/01) or 13.5% of the revenue from these cropping systems. A review of soil–water and solute movement under precision irrigation systems highlights the gaps in current knowledge including the mismatch between the data required by complex, process-based soil–water or solute simulation models and the data that is easily available from soil survey and routine soil analyses. Other major knowledge gaps identified include: (a) effect of root distribution, surface evaporation and plant transpiration on soil wetted patterns, (b) accuracy and adequacy of using simple mean values of root zone soil salinity levels to estimate the effect of salt on the plant, (c) fate of solutes during a single irrigation and during multiple irrigation cycles, and (d) effect of soil heterogeneity on the distribution of water and solutes in relation to placement of water. Opportunities for research investment are identified across a broad range of areas including: (a) requirements for soil characterisation, (b) irrigation management effects, (c) agronomic responses to variable water and salt distributions in the root zone, (d) potential to scale or evaluate impacts at various scales, (e) requirements for simplified soil–water and solute modelling tools, and (f) the need to build skills and capacity in soil–water and solute modelling.  相似文献

Commercial performance evaluations of surface irrigation are commonly conducted using infiltration functions obtained at a single inflow rate. However, evaluations of alternative irrigation management (e.g. flow rate, cut-off strategy) and design (e.g. field length) options using simulation models often rely on this single measured infiltration function, raising concerns over the accuracy of the predicted performance improvements. Measured field data obtained from 12 combinations of inflow rate and slope over two irrigations were used to investigate the accuracy of simulated surface irrigation performance due to changes in the infiltration. Substantial errors in performance prediction were identified due to (a) infiltration differences at various inflow rates and slopes and (b) the method of specifying the irrigation cut-off. Where the irrigation cut-off at various inflow rates was specified as a fixed time identified from simulations using the infiltration measured at a single inflow rate, then the predicted application efficiency was generally well correlated with the application efficiency measured under field conditions at the various inflow rates. However, the predictions of distribution uniformity (DU) were poor. Conversely, specifying the irrigation cut-off as a function of water advance distance resulted in adequate predictions of DU but poor predictions of application efficiency. Adjusting the infiltration function for the change in wetted perimeter at different inflow rates improved the accuracy of the performance predictions and substantially reduced the error in performance prediction associated with the cut-off recommendation strategy.  相似文献

The distribution of T cells and T cell subsets was examined within the human central nervous system in active lesions from seven patients with chronic multiple sclerosis. The monoclonal antibodies anti-T11, anti-T4, and anti-T8 were used to detect total (whole) T cells, helper T cells, and suppressor-cytotoxic T cells, respectively, and a monoclonal antibody against human Ia was used for macrophages and B cells. Lesion progression was associated with large numbers of T4+ cells at the lesion margin and these extended great distances into the adjacent normal-appearing white matter. T8+ cells were most commonly concentrated around the lesion margin and displayed a preferential perivascular distribution. Within the lesion center, only a few T cells were found. Ia+ macrophages were most numerous within the centers of active lesions and were always present in the adjacent normal white matter. The monoclonal antibodies to T cells did not cross-react with glial cells including oligodendrocytes. These results indicate that T4+ cells are actively involved in lesion extension and Ia+ cells, in demyelination.  相似文献

The deer ked (Lipoptena cervi) is a haematophagous parasitic fly of cervids that spread to Finland in the early 1960's. Presently its northern distribution limit lies at approximately 65°N and it is gradually spreading northwards. In Finland the principal host species has been the moose (Alces alces), but the deer ked is about to establish contact with another potential host, the semi-domesticated reindeer (Rangifer tarandus tarandus) causing possible threats to reindeer health and management. The aim of this study was to investigate if the deer ked would have an influence on the welfare of the reindeer. Eighteen adult reindeer were divided into three experimental groups: the control group and two infected groups with 300 deer keds per reindeer introduced in August-September. One of the infected groups was treated with subcutaneous ivermectin in November. To gather comprehensive data on potential health hazards caused by the deer ked a wide array of physiological variables was measured during and at the end of the experiment in December. The keds caused no clear changes in the complete blood count, plasma clinical chemistry, amino acids, endocrinology, energy stores, enzyme activities or tissue fatty acid profiles of the host. The haematological, clinical chemical and endocrinological values displayed changes that could be related to the seasonal physiological adaptations of the species. In conclusion, at the duration and intensity of infection that were employed, the effects of the deer ked on the measured physiological variables of the reindeer were insignificant.  相似文献

Many seabird species are experiencing population declines, with key factors being high adult mortality caused by fishery by-catch and predation by introduced predators on nesting islands. In the Mediterranean, both of these pressures are intensive and widespread. We studied the adult survival of an endemic Mediterranean seabird, the Yelkouan shearwater (Puffinus yelkouan), between 1969–1994 and 2007–2010 in Malta and between 2004–2010 in France using mark–recapture methods. Mean annual survival probabilities for breeding adults were below 0.9 for all colonies and periods. Between 1969–1994, annual survival for adults of unknown breeding status was on average 0.74 (95% confidence interval: 0.69–0.80) in Malta, possibly as a result of various human disturbances (including illegal shooting), light pollution and fisheries by-catch. Over the period 2004–2010, we found strong support for variation in adult survival probabilities between breeders and non-breeders, and islands with and without introduced predators in France. Survival probabilities for non-breeders (0.95, 0.81–1.0) appeared to be higher than for breeders (0.82, 0.70–0.94), but were imprecise partly due to low recapture probabilities. In Malta, we found evidence for heterogeneity in survival probabilities between two unknown groups (probably breeders and non-breeders), and seasonal variation in survival probability. Birds were more likely to survive the period including the peak breeding season than an equally long period during which they roam widely at sea. Although annual adult survival probability was still low (0.85, 0.58–1.0), colony protection measures appear to have reduced mortality at nesting cliffs. A population model indicated that colonies in France and Malta would currently require continuous immigration of 5–12 pairs per year to maintain stable populations. Our estimates of adult survival probabilities over the past four decades are consistent with overall population declines. Threats to Yelkouan shearwaters require immediate management actions to avoid ongoing population declines in the western Mediterranean.  相似文献

Abstract –  In a previous study on the male roach, Rutilus rutilus , we found a positive correlation between sexual ornamentation, breeding tubercles (BTs) on lateral sides and parasite resistance. As a continuation of that study, we examined another measure of sexual ornamentation – number of BTs on front head – of male roach from five populations. BTs on head correlated positively with fish size in two populations and with condition in one population, but not with parasite resistance (proportion of dead parasites) in any of the populations. Moreover, head BTs correlated negatively with the number of two parasite species ( Rhipidocotyle campanula and Myxobolus mülleri ) in two populations, but positively with the number of a third parasite ( Raphidascarus acus ) in one population. In addition, a negative correlation with spleen size was found in one population. The present results suggest that head BTs of male roach – although the patterns vary between populations – may signal condition and parasite load, but not resistance, in contrast to lateral BTs. Therefore, our results are in line with the hypothesis of multiple-message sexual ornaments.  相似文献
