In leaves pretreated with an indole derivative [indole-3-acetic acid (IAA) tryptamine, or tryptophan], blast lesion formation was suppressed compared to those treated with distilled water (DW) as a control. Phenylalanine ammonia-lyase (PAL) activity and PAL expression were significantly enhanced in the IAA- or tryptophan-pretreated leaves, but not in tryptamine-pretreated leaves. This induction of resistance was inhibited by pretreatment with a PAL inhibitor, α-aminooxyacetic acid, in IAA- and in tryptophan-treated leaves, but not in tryptamine-treated leaves. This study strongly shows that the indole derivatives IAA tryptamine and tryptophan can enhance a disease resistance mechanism that is supported by different metabolic pathways.  相似文献

Chemical fungicides are mainly used to control Colletotrichum orbiculare, but this fungal pathogen usually develops resistance to the pesticides. Natural compounds are therefore desirable for controlling C. orbiculare. Here, a culture filtrate (CF) from ME202, a fungal strain isolated from Trifolium incarnatum in Shimane Prefecture in 2020, was tested in vitro and found to inhibit conidial germination of C. orbiculare. The inhibitory compound in ME202-CF was soluble in ethyl acetate. The ethyl acetate extract of ME202-CF (ME202-ECF) significantly inhibited in vitro conidial germination of C. orbiculare in a dose-dependent manner and lesion formation on cucumber leaves. The inhibitory compounds were heat stable at 40–121 °C and molecular mass?≥?500 and ≥?1000 Da. Thin layer chromatography–bioautography of ME202-ECF showed that the compounds inhibiting C. orbiculare growth had an Rf of 0.81 and 0.91. Furthermore, ME202 was found to be a member of the genus Cercospora through sequence analysis of the internal transcribed spacer and 5.8 S rDNA. These results suggest that secondary metabolites of Cercospora spp., such as ME202, can be used to develop new fungicides against anthracnose.

The variation in the root diameter of field-grown crops was directly measured under a microscope by manually removing soil particles from roots without washing the roots. The frequency of the root diameter at the mode (about 10 μm, mainly of root hairs) was higher in wheat and maize (about 60%) than in soybean and sugar beet (about 30%), and it was the lowest in potato (21%). The diameter of 88 to 97% of the roots larger than 20 μm ranged from 21 to 160 μm. The finer the roots, the higher was the frequency, except for potato roots in which the frequency of roots with a diameter of 21 to 40, 41 to 80, and 81 to 160 μm was about 30%. Since fine roots and root hairs account for the largest proportion of the crop root system, they should appropriately be quantified whenever morphological and physiological aspects of roots are analyzed.  相似文献

At Tlaltizapan, Mexico, 26 maize varieties with a wide range of various contrasting traits were grown at three plant densities (25, 000, 50, 000, and 100, 000 plants per ha) to clarify the factors currently limiting grain yield. The following results were obtained.

1. Long-duration varieties compared to short-duration varletles tended to be taller and to produce a larger number of leaves, larger LAl, larger TDW, and higher grain yield. No evidence was obtained to indicate that plant type was important for high-yielding varieties.

2. High grain yield was attributable mainly to the larger number of kernels per unit field area. The sink size is possibly the dominant factor controlllng the grain yield, indicating the importance of studies on the yield components.

For obtaining high-yielding varieties, it would be important to improve a strain so that a single ear became larger and more tolerant to barrenness at a high plant density.  相似文献

Measuring the phosphorus (P) solubility in animal manure compost (AMC) is important to estimate both the risk of P loss from agricultural land and the P availability for crops in agriculture that use AMC as fertilizer. Water-extractable phosphorus (WEP) has most often been used to estimate P solubility in AMC. A single water extraction with a high ratio of compost to water is usually used to determine WEP, but in many cases, this may not evaluate the maximum WEP. In this study, we extracted the P included in AMC by using a continuous extraction method with a large amount of water, and tested this approach for 16 AMCs: four cattle manure composts, five swine manure composts, five layer (chicken) manure composts, and two broiler (chicken) litter composts. The P dissolution patterns were fitted to kinetic models, and the maximum WEP (WEPmax) was determined by the coefficient of the non-linear regression equation. The WEPmax values corresponded to the sum of H2O- and sodium bicarbonate (NaHCO3)-extractable P measured using the modified Hedley sequential extraction method. The result also suggests that the maximum amount of WEP from AMC can be estimated rapidly using a sequential extraction (the Hedley method) that has been widely used to characterize P in manure or AMC. The maximum water-extractable magnesium (Mg) (WEMgmax) was significantly positively correlated with WEPmax (r = 0.854, P < 0.01). This suggests that WEPmax in the AMCs is affected by water-soluble magnesium compounds.  相似文献

Summary

Genetic variation and resulting variable groupings in tea (Camellia sinensis) and its wild Camelia relatives were assessed using Random Amplified Polymorphic DNA (RAPD) and Amplified Fragment Length Polymorphic (AFLP) markers. Numerous polymorphic bands were generated, of which 266 unambiguous ones were scored. The highest level of polymorphism as determined by the expected heterozygosity (Hav) was detected with AFLPs. Three major groups were recognized in the germplasm based on the parsimony method of cluster analysis. Two of the groups corresponded to varieties assamica and sinensis while the third group consisted of a heterogeneous mix of tea cultivars and related wild species. The Taiwan yamacha, unique tea cultivars grown predominantly in the highlands of Taiwan for the production of pan fired semi-fermented (Oolong and Pouchong) tea clustered in this group. Analysis of phenotypic diversity based on the generated RAPD and AFLP profiles revealed that population diversity (H), decreased in the order; Wild Camellia >India > China > Kenya > Sri-lanka > Vietnam > Japan > Taiwan. Analysis of molecular variance (AMOVA) revealed that most variation resided among individuals within populations (72%). Calculation of genetic distances and nested AMOVA further revealed a significant degree of structuring among the populations based on the country of origin.  相似文献

An ethyl acetate extract of a culture filtrate (ECF) from an unidentified fungal isolate O821 was evaluated for antifungal activity against the rice pathogen Magnaporthe oryzae. The O821-ECF significantly inhibited spore germination, appressorium formation, and mycelial growth of M. oryzae, and its antifungal activity was heat-stable. It also significantly suppressed the number and size of blast lesions. In an analysis of the ITS sequence of this isolate, it shared similarities with species of the fungus Biscogniauxia. These results suggest that isolate O821 of the genus Biscogniauxia produces a heat-stable antifungal compound(s) in its culture filtrate.  相似文献

There is strong evidence that oxidative stress participates in the etiology of neurodegenerative diseases such as Parkinson's, and Alzheimer's diseases. Moreover, emotional stress effects in the central nervous system play a vital role in homeostasis. The protective effect of anthocyanins on the cerebral oxidative stress was studied using the whiskers cut model. In mice, such treatment causes psychological or emotional distress leading to oxidative stress in tissues. To investigate the in vivo antioxidant activity of anthocyanins, an extract of Vaccinium myrtillis L., an anthocyanin mixture, was orally administered (100 mg/kg of body weight.) to mice for 7 days, and then psychological stress was assessed by cutting off their whiskers. Whisker removal increased both protein carbonyl formation and lipid peroxidation in the brain, heart, kidney, and liver. Further, the levels of oxidative markers showed regional differences in the brain. Concomitantly, dopamine neurotransmitter levels were altered in both the midbrain and the brain cortex. Orally administered anthocyanins were also active in the brain, suppressing stress-induced cerebral oxidative stress and dopamine abnormalities in distressed mice. These effects of anthocyanin treatment suggest their possible usefulness for the treatment of cerebral disorders related to oxidative stress.  相似文献

The effects of the dietary medicinal herbs Massa medicata, Crataegi fructus (Cf), Artemisia capillaries, Cnidium officinale and their mixture (HM), on growth and resistance against Vibrio anguillarum were examined using larval red sea bream, Pagrus major. The methanol extracts of Cf and HM inhibited the proliferation of seven infectious bacterial strains including the genera Aeromonas, Edwasiella, Photobavterium, Pseudomonas and Vibrio. Rotifers enriched with emulsified bonito oil containing extracts of Cf and HM lowered the bacterial counts in their body on TCBS medium for the genus Vibrio in comparison with rotifers enriched by emulsified bonito oil alone. Although statistically not significant, larvae fed rotifer fortified with Cf from 3 to 20 days after hatching showed the best growth and final body weight followed by those given rotifer with HM and control enrichment. However, final body length of larvae fed rotifer with Cf was significantly larger than larvae fed on rotifers enriched with other herb extracts. After a challenge test with V. anguillarum, survival of larvae fed rotifer with Cf and HM was significantly higher than for the control treatment. These results revealed that Cf and HM in rotifer is useful to promote growth and resistance against V. anguillarum in red sea bream larvae, providing a new technology for mass production of disease‐resistant fry and fingerlings.  相似文献

A cell line (PL38PB) was established from blood samples of a 6-month-old pig that was diagnosed with lymphoma with CD5 expression. Histopathological examination revealed neoplastic lesions in the spleen, liver and lymph nodes. Tumor cells were immunohistochemically positive for CD20 and immunoglobulin heavy chains (μ, γ and α). Membranous CD5 and cytoplasmic Immunoglobulin M (IgM), ​Immunoglobulin G (IgG) and ​Immunoglobulin A (IgA) were detected in PL38PB cells by flow cytometry. In addition, the cytoplasm of PL38PB cells were positive for IgM, IgG and IgA by immunofluorescent. However, no Ig secretion was detected in culture supernatant by Ouchterlony gel diffusion method. Results suggest that PL38PB cells express three Ig isotypes that are produced but not secreted.  相似文献
