In 2008 and 2009 seasons, a sudden increase in Potato virus Y (PVY) incidence was recorded in foundation seed potatoes in Hokkaido, northern Japan. This increase was obvious during the field inspection and the postharvest indexing. Molecular typing revealed that besides the previously reported strains of PVYO and PVYNA‐N, the most common strain identified was the recombinant PVYNTN, with three characteristic recombinant junctions at the HC‐Pro, VPg and CP regions. No potato tuber necrotic ringspot disease (PTNRD) was observed in foundation seed potatoes in correlation with the presence of PVYNTN. Moreover, an isolate with a typical PVYNTN recombinant genome, namely Eu‐12Jp, did not induce PTNRD in 62 Japanese potato cultivars tested in both primarily and secondarily infected plants. Two cultivars carrying the extreme resistance gene Rychc were resistant to the infection with Eu‐12Jp, which presents potential sources of resistance to PVYNTN. Eu‐12Jp induced systemic mottle in potato cultivars Desiree and King Edward carrying resistance genes Ny and Nc, respectively, but induced a hypersensitive reaction in potato cultivar Maris Bard, with the Nz hypothetical resistance gene typical of the PVYZ strain group. Therefore, based on the genome structure and the reaction of the potato N resistance genes, Eu‐12Jp should be classified as PVYZ‐NTN, as described for isolates from Idaho, USA recently. This is the first report of PVYZ‐NTN in Japan and the sudden and increased occurrence of PVYNTN/PVYZ‐NTN represents a potential risk of PTNRD developing and increases the significance of PVY in Japan.  相似文献

The surface of micro crustaceans collected from the overlying water of a submerged paddy soil was observed in detail by scanning electron microscopy. Epibiotic bacteria were less diverse compared to free-living bacteria in the overlying water. Specific bacteria appeared to preferentially colonize certain parts of microcrustaceans. The epibiotic bacteria, the sites they colonized and their density depended on the kinds of microcrustaceans (Moina macrocopa, Cyclopidae, Cypretta sp., and Tanycypris sp.). These results suggested the presence of significant associations in the overlying water between specific groups of bacteria and microcrustaceans.  相似文献

The changes of Al species in the presence of montmorillonite (Mt) with aging were investigated using 27Al-nuclear magnetic resonance and inductively coupled plasma atomic emission spectroscopy after extraction with 1 mol L-1 KCl. Composition of the Al species in a hydroxyaluminosilicate (HAS) solution with a Si/Al molar ratio of 0.63 without Mt was not appreciably affected by 42 d of aging. In the absence of Mt, the concentration of Al13 ([Al04Al12(OH)24- H2O)12]7+) in the HAS solution with a Si/Al molar ratio of 0.09 and hydroxy-aluminum (HyA) solution decreased during 42 d of aging, suggesting that degradation (or polymerization) of Al13 took place upon aging. In the presence of Mt, Al13 was adsorbed onto Mt from the HyA and HAS(0.09) solutions. The adsorbed Al13 was partly recovered by 1 mol L-1 KCl from HyA- and HAS(0.09)Mt complexes after 42 d of aging, suggesting that at least a part of the adsorbed Al13 was exchangeable and the rest was considerably stabilized by adsorption onto Mt. The desorption ratios of Al from the HyA- and HAS(O.09)Mt complexes accounted for 25 to 30% and 6 to 8% of total Al adsorbed, respectively. The species of Al desorbed from these complexes consisted mainly of Al13 and AlNON. The AlNON was attributed to electrically asymmetric Al including HAS and/or polymer HyA ions.  相似文献

ABSTRACT:   The Mekong giant catfish is one of the largest freshwater fish in the world, and is endemic to the Mekong basin. Recently, the number of Mekong giant catfish has drastically decreased. In this study, seven microsatellite DNA markers of the Mekong giant catfish were developed for evaluation of the species' genetic diversity. Mendelian inheritance of four of seven markers was confirmed using offspring (F1) produced from one pair. Number of alleles per locus in the founder population ranged 1–4 with a mean of 2.8, and that in F1 ranged 1–2 with a mean of 1.6. The expected heterozygosity in the founder population ranged 0–0.66 with a mean of 0.38, and that in F1 ranged 0–0.5 with a mean of 0.28. The mean expected heterozygosity in the founder population is apparently lower than that in Pangasius bocourti and other freshwater species, but higher than that in endangered species. In the future, the microsatellite data in this study will be used in minimal kinship approaches, because these individuals constitute the basic seed for release into wild in an effort to conserve the native population of Mekong giant catfish.  相似文献

We have elucidated genetic relationships of Vietnamese native pigs (VNP) using preliminarily collected samples by a single‐nucleotide polymorphism (SNP) array. In order to confirm our previous results and compare with the results of a previous study using microsatellite (MS) markers, we aimed to characterize genetic diversity and population structure in wider varieties (24 breeds from 21 Provinces) of VNP across the country using 20 polymorphic MS markers recommended by ISAG/FAO (International Society for Animal Genetics/Food and Agriculture Organization) for diversity study. In this study, we collected 1,136 DNA samples of the VNPs and three exotic breeds. Our results revealed that the average number of alleles and allelic richness across the loci in VNPs were 10.0 and 7.6, which were higher than those of exotic breeds. Genomic components among VNPs were subjected to the sampling locations. Interestingly, Co Binh Thuan showed remarkable genetic feature compared to the other VNPs, because the habitation of Co Binh Thuan was relatively far from the other breeds. The results of this study provided useful information for exploitation, conservation, and development trends of the VNP breeds. More recently, African swine fever caused significant damage to most of the VNP populations. Therefore, our findings will help a reconstruction scheme of the VNP genetic resources.  相似文献

The function and/or morphological features of the vomeronasal olfactory system remain unclear in aquatic animals, although the system appeared first in urodeles based on phylogenic data. We examined the lectin binding patterns in the olfactory bulb of a semi-aquatic urodele, the Japanese red-bellied newt, Cynops pyrrhogaster, using 22 different lectins. Eleven of the lectins showed specific binding to the nerve fibres and glomeruli in the olfactory bulb. Among these, Wheat germ agglutinin, pokeweed and peanut agglutinin preferentially bound the main olfactory bulb, reflecting variation in the expression of glycoconjugates between the main and accessory olfactory bulbs. By contrast, the types of lectins bound to the Cynops olfactory bulb were considerably different from those reported in other urodele families. These results suggest a histochemical distinction between the main and accessory olfactory bulbs, and that glycoconjugate expression may differ significantly among urodele families.  相似文献

Interleukin-2 (IL-2) binds to two distinct receptor molecules, the IL-2 receptor alpha (IL-2R alpha, p55) chain and the newly identified IL-2 receptor beta (IL-2R beta, p70-75) chain. The cDNA encoding the human IL-2R beta chain has now been isolated. The overall primary structure of the IL-2R beta chain shows no apparent homology to other known receptors. Unlike the IL-2R alpha chain, the IL-2R beta chain has a large cytoplasmic region in which a functional domain (or domains) mediating an intracellular signal transduction pathway (or pathways) may be embodied. The cDNA-encoded beta chain binds and internalizes IL-2 when expressed on T lymphoid cells but not fibroblast cells. Furthermore, the cDNA gives rise to the generation of high-affinity IL-2 receptor when co-expressed with the IL-2R alpha chain cDNA.  相似文献

In the interleukin-2 (IL-2) system, intracellular signal transduction is triggered by the beta chain of the IL-2 receptor (IL-2R beta); however, the responsible signaling mechanism remains unidentified. Evidence for the formation of a stable complex of IL-2R beta and the lymphocyte-specific protein tyrosine kinase p56lck is presented. Specific association sites were identified in the tyrosine kinase catalytic domain of p56lck and in the cytoplasmic domain of IL-2R beta. As a result of interaction, IL-2R beta became phosphorylated in vitro by p56lck. Treatment of T lymphocytes with IL-2 promotes p56lck kinase activity. These data suggest the participation of p56lck as a critical signaling molecule downstream of IL-2R via a novel interaction.  相似文献
