Fatty acids have been used in marine biogeochemistry as food chain biomarkers, but in freshwater these studies are rare. In order to evaluate the fatty acid potential as biomarkers in freshwater, their profile was analyzed during vitellogenesis in two fish species, in both waterfall and reservoir environments of the Paraíba do Sul River Basin. Detrivorous Hypostomus affinis and omnivorous Geophagus brasiliensis seem to elongate and desaturate polyunsaturated fatty acids (PUFA) and transfer them to the ovaries’ phospholipids. Waterfall Geophagus brasiliensis have more highly unsaturated fatty acids in the liver, but in the reservoir, accumulation mainly occurs in muscle and ovary triglycerides, suggesting trophic opportunism and a plasticity during vitellogenesis. In Hypostomus affinis, PUFA alteration occurs only in the reservoir, suggesting a high phytoplankton occurrence. Eutrophication and water speed is reflected in Hypostomus affinis ovaries by higher PUFAn3 and bacterial fatty acids. As in marine environments, analysis of mono- and polyunsaturated fatty acids during vitellogenesis can be used as a tool in food chain studies in freshwater.  相似文献

Cowpea is widely cultivated in arid and semi‐arid regions of the world where salinity is a major environmental stress that limits crop productivity. The effects of moderate salinity on growth and photosynthesis were examined during the vegetative phase of two cowpea cultivars previously classified as salt‐tolerant (Pitiúba) and salt‐sensitive (TVu). Two salt treatments (0 and 75 mm NaCl) were applied to 10‐day‐old plants grown in nutrient solution for 24 days. Salt stress caused decreases (59 % in Pitiúba and 72 % in TVu) in biomass accumulation at the end of the experiment. Photosynthetic rates per unit leaf mass, but not per unit leaf area, were remarkably impaired, particularly in TVu. This response was unlikely to have resulted from stomatal or photochemical constraints. Differences in salt tolerance between cultivars were unrelated to (i) variant patterns of Cl? and K+ tissue concentration, (ii) contrasting leaf water relations, or (iii) changes in relative growth rate and net assimilation rate. The relative advantage of Pitiúba over TVu under salt stress was primarily associated with (i) restricted Na+ accumulation in leaves paralleling an absolute increase in Na+ concentration in roots at early stages of salt treatment and (ii) improved leaf area (resulting from a larger leaf area ratio coupled with a larger leaf mass fraction and larger specific leaf area) and photosynthetic rates per unit leaf mass. Overall, these responses would allow greater whole‐plant carbon gain, thus contributing to a better agronomic performance of salt‐tolerant cowpea cultivars in salinity‐prone regions.  相似文献

The physiological state of fish before transportation is an important aspect to be observed when reduction in losses due to mortality is aimed. Various preparative procedures for transport are reported, but little is known about the use of additives in fish diet. In this study, we aimed to evaluate the effect of dietary turmeric (Curcuma longa) supplementation on the tolerance of juvenile Astyanax aff. bimaculatus to transportation stress. The experimental design consisted of six treatments: 0, 20, 40, 60, 80 and 100.0 g kg?1 turmeric. Fish weighing 0.83 ± 0.04 g were distributed in 24 80‐L aquaria at a density of 0.5 fish L?1 and were fed to satiation three times a day for 60 days. After this period, fish were transported for 24 h in plastic bags (15 fish per bag). Using a discontinuous linear response plateau (LRP) model, the lowest estimated concentration of turmeric that maximized the survival rate was 16.67 g kg?1. Quadratic effects of turmeric on blood lactate levels were observed, and the estimated concentration of turmeric required to minimize blood lactate levels was 64.7 g kg?1. We observed quadratic effects of turmeric on superoxide dismutase activity and the concentration of malondialdehyde in fish gills and the turmeric concentrations required to minimize the transport stress effects on these variables were 42.50 and 83.33 g kg?1, respectively. Thus, dietary supplementation with turmeric enhanced the tolerance of juvenile A. aff. bimaculatus to transportation stress.  相似文献

With this study, we aimed to assess the safety of using Curcuma longa in fish feed. Juvenile Astyanax aff. bimaculatus (0.83 ± 0.04 g) were kept in 24 80‐L aquaria, at a density of 0.5 fish L?1 for 60 days. Six diets supplemented with 0.0, 20.0, 40.0, 60.0, 80.0 and 100.0 g kg?1 were evaluated. Histopathological analysis of the gills showed that there was an increasing linear effect of turmeric on the frequency of lamellar aneurysm, hypertrophy of mucous cells, oedema and necrosis. There was a quadratic effect of turmeric supplementation on the frequency of fusion and disruption of secondary lamellae, vacuolation and hypertrophy of the epithelial cells, and the estimated values to minimize these variables were 4.63, 25.93 and 24.46 g kg?1 respectively. There was a quadratic effect of turmeric on the gills index (IBRA), and the estimated value to minimize this index was 30.23 g kg?1. No effect of turmeric on the frequency of histopathological changes and organ index in fish liver was observed. Thus, we conclude that turmeric is not hepatotoxic for A. aff. bimaculatus; however, its use is safe at concentrations up to 30.23 g kg?1 diet as a function of its gill toxic effect at higher doses.  相似文献

Tilapia aquaculture is one of the fastest‐growing segments of fish production in Brazil. Nile tilapia (Oreochromis niloticus) is largely cultivated in the state of Parana, where Streptococcus agalactiae is the cause of severe disease outbreaks. The objective of this paper was to evaluate an inactivated S. agalactiae vaccine in tilapia for the control of streptococcal disease outbreaks. Tilapia, weighing approximately 20 g each, were intraperitoneally (i.p.) inoculated with 0.1 mL of the vaccine at a dose of 2.0 × 108 colony‐forming unit (CFU) mL?1. One group of tilapia (treatment 1) received one vaccine dose, and the other group of tilapia (treatment 2) received two doses, with an interval of 21 days. The control group was i.p. inoculated with 0.1 mL tryptic soy broth fish?1. Immunized and control tilapia were i.p. challenged with 0.1 mL of 3.0 × 107 CFU mL?1 at 30 days post vaccination. The fish were monitored daily for disease signs and for mortality for 16 days post challenge. A statistically significant difference (P=0.0045) was found between the mortality of treatments 1 and 2. The value of relative per cent of survival of 83.6% and 96.4%, respectively, indicate that this vaccine was efficient in Nile tilapia.  相似文献

The aim of the present study was to evaluate the effect of turmeric (Curcuma longa) as additive in the diet for Astyanax aff. bimaculatus. Fish (0.83 ± 0.04 g) were fed, for 60 days, with six diets containing 0.0, 20.0, 40.0, 60.0, 80.0, and 100.0 g turmeric kg?1 feed. There was an increasing linear effect of turmeric on the thickness of the muscular layer, and height and width of the folds of the intestine. In the liver, a quadratic effect was observed of turmeric on the percentage of hepatocyte cytoplasm and a decreasing linear effect on the percentage of sinusoid capillaries. A quadratic effect was also observed of turmeric on the liver glycogen. There was no effect of turmeric on the antioxidant activity in the liver, carcass composition or productive performance of the fish. Thus, we concluded that Curcuma longa has trophic effects on the epithelium and the muscular layer of the intestine of A. aff. bimaculatus. Additionally, low levels of Curcuma longa cause increased deposition of liver glycogen and high levels cause reduction.  相似文献

The silver catfish, Rhamdia quelen , is endemic to North, Central and South America with high aquaculture potential and wide acceptance in the market. Breeder fish were subjected to induced reproduction through hypophysation using a crude common carp pituitary extract. Egg characteristics, oocyte surface ultrastructure and histology of larval ontogenesis until whole yolk resorption were described for the first time for this species. Oocytes and semen were obtained by manual extrusion, and fertilization was conducted using the dry method. After fertilization, eggs were kept in incubators at 24 °C. The embryonic development was monitored using a stereomicroscope every 10 min until hatching. To analyse the larval development, larvae samples were collected from incubators daily until the fifth day, fixed in Bouin's fluid and subjected to routine histological techniques. The oocyte extrusion occurred 8 h after the second hormone dose at 26 °C. The oocytes were spherical, non-adhesive and yellow, with a diameter of 1471.75±47.63 μm. Scanning electron microscopy revealed a thin jelly coat covering the zona radiata in the animal pole around the micropyle. The blastopore closure occurred within 8 h after fertilization, and the fertilization rate was 79.9±5.2% at 24 °C. Embryonic development was completed within 25 h 30 min after fertilization. The complete resorption of the yolk and the formation of the digestive system organs and the mouth opening occurred on the fifth day, indicating a need for exogenous feeding. The results of this study provide information important for improvement in R. quelen culture and management.  相似文献

Haematological response and growth performance over 150 days, and resistance to a low-temperature stress of Nile tilapia fed diets with increasing folic acid (FA) levels were evaluated. The experiment was conducted in a completely randomized design with eight FA levels (0.0, 0.5, 1.0, 2.0, 3.0, 4.0, 5.0 and 6.0 mg kg−1 feed) supplemented in purified diets (32.0% CP and 13 398 kj DE kg−1). One hundred and ninety-two fingerlings were randomly assigned to 32 net cages distributed in eight 1000 L aquaria with a physical and biological filter and a temperature control system (26.0 ± 1.0 °C). For cold-induced stress, fish were transferred to 24 30 L-aquaria with individual biofilters and aeration. The water temperature was gradually reduced until it reached 13 °C. Haematological parameters evaluated before and after cold stress were total erythrocytes and leucocytes count, differential leucocyte, haemoglobin, haematocrit, total plasmatic protein and haematometric indices. Growth performance parameters were mean weight gain, feed conversion ratio and survival. Dietary FA supplementation did not influence erythropoiesis under normal temperature conditions; cold stress impaired erythropoiesis, causing hypochromic microcytic anaemia and leucopoiesis, and also neutrophilia. Growth performance is influenced by folate and supplementation between 0.5 and 1.0 mg FA kg−1 diet, which makes up for nutritional demands, guaranteeing production and health under appropriate temperature conditions.  相似文献

The correlation between chlorophyll content and quantitative color parameters was investigated in order to find an indirect method for predicting green pigment in ripening soybean seeds. Five Brazilian soybean varieties harvested at different maturity stages (R(6) to R(8) according to the scale of Fehr & Caviness) and dried under two conditions (in oven at 40 degrees C with circulating air and at ambient temperature around 25 degrees C) were analyzed in two consecutive years. The slow-dried seeds at 25 degrees C lost chlorophyll faster, whereas drying at 40 degrees C did not result in yellowing of seeds. High and significant linear correlations between a value and total chlorophyll were obtained over the whole maturation period and on both conditions of drying. From an industrial point of view it appears that a value, obtained by the CIE-L*a*b* method, seems to be a good tool to be applied for quality control and classifying soybean seeds for different purposes.  相似文献
