 Rice reflectance was measured to determine the spectral regions most sensitive to leaf blast infection with a multispectral radiometer. As disease severity increased, reflectance also increased in the 400–500 nm (blue), 570–700 nm (red), and 900–2000 nm regions but decreased in the 500–570 nm and 700–900 nm regions. The increased reflectance in the blue and red regions may be attributed to decreased chlorophyll and carotenoid contents in response to the blast infection. The maximum and minimum reflectance differences occurred at 680 nm and 760 nm for the nondiseased and diseased rice, respectively. The spectral location of maximum sensitivity was 675 nm regardless of disease severity. Rice reflectance ratios were evaluated as indicators of leaf blast severity. Two ratios, R550/R675 (reflectance at 550 nm divided by reflectance at 675 nm), and R570/R675 quantified the significant disease severity. These wavelengths were selected based on the sensitivity minima and maxima. The ratios of nondiseased rice plants varied depending on growth stage. Repro22 is an N-ethyl-N-nitrosourea (ENU)-induced mutation in mice showing depletion of both male and female germ cells. In the present study, we investigated the male phenotypes of the mutant mouse at the adult stage. The repro22/repro22 homozygous mice showed reduced body weights as well as markedly reduced testis weights. Histological examination of the testes at 4 and 10 months of age showed no germ cells in the seminiferous tubules of the affected testis while a number of Sertoli cells were observed in the tubules. In addition to the germ cell depletion, the testes of the affected mouse contained expanded intertubular spaces that were filled by Leydig cell-like interstitial cells. These interstitial cells were confirmed to be Leydig cells by immunohistochmical staining using anti-3beta-HSD antibody. The estimated number of Leydig cells in the affected testes at 10 months of age increased approximately 2 fold compared with those of normal testes. Furthermore, the plasma testosterone levels of the affected mice at 10 months of age were significantly higher than those of the normal mice. These findings indicated that the repro22/repro22 mouse developed hyperplasia of Leydig cells that was presumably caused by the absence of germ cells in the seminiferous tubules.  相似文献

This study is the first investigation on Japanese isolates of Prototheca zopfii from bovine mastitis and the cow-barn surroundings by molecular characterization to clarify routes of infection for bovine protothecal mastitis. We performed isolation of Prototheca from cow-barn surroundings (drinking water, sewage and feces) and milk samples from cases of bovine mastitis. Genotypes of the 32 isolates of P. zopfii from cow-barn surroundings and 67 isolates from mastitis were analyzed by genotype-specific PCR assays and restriction fragment length polymorphism (RFLP) assays. All mastitis isolates were identified as P. zopfii genotype 2. Conversely, 29 isolates from cow-barn surroundings were identified as P. zopfii genotypes 1 and 3 isolates as genotype 2, respectively. Given these results, both genotypes of P. zopfii could exist in cow-barn surroundings, but no sites were identified as frequent sources of P. zopfii genotype 2. P. zopfii isolates should thus be further explored with regard to genotype to clarify the reservoir of etiological agents in bovine Prototheca mastitis.  相似文献

To understand the development of cold hardiness in highbush blueberry plants (Vaccinium corymbosum L.), we investigated seasonal changes in the protein compositions of current-year and overwintered shoots. Electrophoresis using SDS-polyacrylamide gels indicated that the amounts of a few proteins increased in autumn, in association with an enhancement of cold hardiness. Of these proteins, 65- and 60-kDa proteins were confirmed to be dehydrins by western blotting. While the levels of most of the accumulated proteins decreased in April, a 27-kDa protein maintained its level in the overwintered stem during spring. The amino acid sequence deduced from a cDNA for this protein showed significant similarities with known chitinases. The stem segments from overwintered shoots sampled in July showed higher cold hardiness against sub-zero temperature than those from current-year shoots, and the bark tissue of overwintered shoots had higher cold resistance than that of current-year shoots. The 27-kDa protein (BC27) was mostly found in the bark tissue of stems. Appreciable accumulation of this protein in floral buds was not detected. The purified BC27 protein exerted a cryoprotective effect on protoplasts subjected to a freeze–thaw treatment. These results suggest that the stems of highbush blueberry develop a system of cold hardiness different from that of the buds and apices, and that the accumulation of chitinase is involved in tolerance to low temperature in winter or unseasonably low temperature in spring.  相似文献

Gelatinolytic enzymes were partially purified from the skeletal muscle of red sea bream Pagrus major and characterized to obtain information on post mortem tenderization of fish muscle. Four gelatinolytic activities, G1 (90 kDa), G2 (65 kDa), G3 (60 kDa), and G4 (100 kDa), were detected in the Q Sepharose column. G1, the major gelatinolytic enzyme, and G4 were identified as serine proteinases from results of inhibitor spectrum and substrate specificity. By contrast, G2 and G3 were found to be metalloproteinases since these were inhibited by ethylenediamine tetraacetic acid and o-phenanthroline, and activated by 4-aminophenylmercuric acetate. The optimum pH and temperature of these enzymes were in the ranges of 7–9 and 20–40°C, respectively.  相似文献

The composition of sediment pore water was determined for ponds constructed on clayey Ultisols at Auburn, Alabama. Pore water was anaerobic and contained much higher concentrations of ferrous iron (Fe2+), soluble reactive phosphorus (SRP), total phosphorus (TP), total ammonia-nitrogen (TAN), and sulfide (S2−) than surface or bottom waters. Concentrations of SRP and TP in pore water were higher in ponds with high soil phosphorus concentrations than in a new pond with less soil phosphorus. Increased concentrations of organic matter in soil or larger inputs of feed to ponds favored greater microbial activity in soils and higher concentrations of TAN in pore water. The pH of pore water was 6.5–7.0, and pH was apparently controlled by the equilibrium:

Movement of Fe2+, SRP, and S2− from pore water into pond water apparently was prevented by the oxidized layer of soil just below the soil-water interface. Pond managers should concentrate on maintaining this oxidized layer to reduce the tendency for toxic substances to diffuse into the pond water.  相似文献

以日本海域捕获的宝石石斑鱼(Epinephelus areolatus)、黑边石斑鱼(E.fasciatus)、赤点石斑鱼(E.akaara)、尾纹九棘鲈(Cephalopholis urodeta)、蜂巢石斑鱼(E.merra)和纹波石斑鱼(E.ongus)成鱼为对象,比较其性腺发育和脑垂体结构以及垂体中FSHβ和LHβ免疫信号的分布。结果表明:宝石石斑鱼、黑边石斑鱼、赤点石斑鱼和尾纹九棘鲈性腺发育为未成熟阶段,蜂巢石斑鱼和纹波石斑鱼性腺发育为成熟阶段。6种石斑鱼的脑垂体位于间脑腹面,由神经垂体(NH)和腺垂体(AH)组成。腺垂体从左至右进一步细分为前外侧部(RPD)、中外侧部(PPD)和中间部(PI)。在宝石石斑鱼、赤点石斑鱼和纹波石斑鱼中NH结构被PPD隔开分为上下两部分,而在黑边石斑鱼、尾纹九棘鲈和蜂巢石斑鱼中NH为一个整体。6种石斑鱼脑垂体中FSHβ和LHβ细胞的免疫信号主要分布在PPD和PI区域,且FSHβ信号强度均较LHβ强,推断在石斑鱼性腺发育成熟前后,FSHβ较LHβ重要。为石斑鱼的资源保护及人工繁殖方面提供基础生物学资料和理论依据。  相似文献

Clearance of Theileria sergenti-infected bovine red blood cells (Bo-RBCs) from the blood circulation of severe combined immune deficiency (SCID) mice was studied to help understand the mechanisms of anemia developing in cattle infected with T. sergenti. For the clearance test, Bo-RBC samples having 2%, 58%, and 76% parasitemia and, as a control, parasite-free Bo-RBCs were prepared in the Bo-RBC-SCID mouse model. The T. sergenti-infected Bo-RBCs and the uninfected control Bo-RBCs were separately labeled with two, green and red, fluorescent dyes, mixed together, and injected intravenously into SCID mice. The blood samples collected at various time points were observed under a fluorescent microscope, and the numbers of green and red fluorescing RBCs were counted differentially to determine the clearance rates of T. sergenti-infected and uninfected Bo-RBCs. This test clearly demonstrated that the Bo-RBC samples having higher parasitemias were cleared faster from the blood circulation of SCID mice. The results suggest that the intravascular clearance system in SCID mice may have a mechanism by which T. sergenti-parasitized and non-parasitized Bo-RBCs are recognized and cleared differentially.  相似文献
