Freezing and thawing may substantially influence the rates of C and N cycling in soils, and soil frost was proposed to induce NO losses with seepage from forest ecosystems. Here, we test the hypothesis that freezing and thawing triggers N and dissolved organic matter (DOM) release from a forest soil after thawing and that low freezing temperatures enhance the effect. Undisturbed soil columns were taken from a soil at a Norway spruce site either comprising only O horizons or O horizons + mineral soil horizons. The columns were subjected to three cycles of freezing and thawing at temperatures of –3°C, –8°C, and –13°C. The control columns were kept at constant +5°C. Following the frost events, the columns were irrigated for 20 d at a rate of 4 mm d–1. Percolates were analyzed for total N, mineral N, and dissolved organic carbon (DOC). The total amount of mineral N extracted from the O horizons in the control amounted to 8.6 g N m–2 during the experimental period of 170 d. Frost reduced the amount of mineral N leached from the soil columns with –8°C and –13°C being most effective. In these treatments, only 3.1 and 4.0 g N m–2 were extracted from the O horizons. Net nitrification was more negatively affected than net ammonification. Severe soil frost increased the release of DOC from the O horizons, but the effect was only observed in the first freeze–thaw cycle. We found no evidence for lysis of microorganisms after soil frost. Our experiment did not confirm the hypothesis that soil frost increases N mineralization after thawing. The introduction of an alien top predator, the brown treesnake (Boiga irregularis), has resulted in severe losses of native vertebrate populations in Guam. Among these are important pollinators and seed dispersers. This study is a first attempt to document cascading effects on vertebrate-pollinated native plant species in Guam. We investigated flower visitation, seed set and germination in two native plants, the mangrove tree Bruguiera gymnorrhiza and the forest tree Erythrina variegata var. orientalis. Both species are bird-pollinated. Studies were conducted on two Mariana islands, Guam (with high density of snakes) and Saipan (with nearly no snakes). Visitation rates by birds were high on Saipan, but zero on Guam. Insects and lizards visited flowers to a low extent on both islands. Only lizards were potential effective pollinators. Seed set of both species were significantly higher on Saipan compared to Guam, and for B. gymnorrhiza, seedling recruitment was significantly higher on Saipan. Hence, these bird-pollinated species appear highly dependent on bird visitors for reproduction. The eradication of flower-visiting birds by the invasive treesnake thus secondarily results in broken mutualistic interactions, which may, in turn, result in a lower recruitment of native plants. Thus, the treesnake affects not only potential prey species, but its effects cascade through the entire ecosystem on Guam. Thirteen samples of authentic absinthe dating from the preban era (i.e., prior to 1915) were analyzed for parameters that were hypothesized as contributing to the toxicity of the spirit, including naturally occurring herbal essences (thujone, pinocamphone, fenchone), methanol, higher alcohols, copper, and antimony. The total thujone content of preban absinthe was found to range between 0.5 and 48.3 mg/L, with an average concentration of 25.4 +/- 20.3 mg/L and a median concentration of 33.3 mg/L. The authors conclude that the thujone concentration of preban absinthe was generally overestimated in the past. The analysis of postban (1915-1988) and modern commercial absinthes (2003-2006) showed that the encompassed thujone ranges of all absinthes are quite similar, disproving the supposition that a fundamental difference exists between preban and modern absinthes manufactured according to historical recipes. Analyses of pinocamphone, fenchone, base spirits, copper, and antimony were inconspicuous. All things considered, nothing besides ethanol was found in the absinthes that was able to explain the syndrome "absinthism". 相似文献

Tree girdling has been experimentally used to stop the allocation of carbohydrates from the canopy to tree roots and their associated ectomycorrhizal (EM) mycelia. We used three already established girdling experiments in northern Sweden, one in a Pinus sylvestris stand and two in Picea abies stands, to determine the effect of tree girdling on soil-living oribatid mites. These mites often feed on fungal hyphae, but it is not known to what extent they feed on EM or saprotrophic fungi. We hypothesised that a presumed decline in EM fungi after girdling would strongly reduce EM-feeding specialists and, correspondingly, reduce total abundance and species richness of oribatids in girdled plots. Tree girdling resulted in a significant decline in total abundance of oribatid mites in the two spruce stands, which was assumed to be linked to the decline in EM fungi, but not in the pine stand. Species richness decreased in girdled plots in one of the spruce forests. The decline in total abundance in girdled spruce stands was primarily dependent on a significant and consistent population decrease in Oppiella nova, which was the most abundant oribatid mite in the ungirdled spruce plots. Its abundance after girdling was only 8–18% of that in ungirdled spruce plots. In the pine stand, O. nova had much lower abundance than in the spruce stands, and despite a tendency to decline in number after girdling also in the pine stand, it had a minor effect on total abundance. These results suggest that O. nova may be dependent on EM fungi in spruce forest soils, whereas the dependence on EM fungi in pine forest soils is less evident. 相似文献

There is increasing evidence that reactive oxygen species arising from several enzymatic reactions are mediators of inflammatory events. Plant preparations have the potential for scavenging such reactive oxygen species. Flavans and procyanidins are bioavailable and stable during the process of cooking. This study used conditions that mimicked digestion of Vitis vinifera seed powder in the stomach (acidic preparation) and small intestine (neutral preparation). The flavonoids of these two preparations were released during simulated digestion and were determined with HPLC analysis. Biochemical model reactions relevant for the formation of reactive oxygen species in vivo at inflammatory sites were used to determine the antioxidant properties of the two preparations. The inhibition of the indicator reaction for the formation of reactive oxygen species represents a potential mechanism of the physiological activity of the corresponding preparation. The results of this work show clearly that the polyphenols released during the simulated digestion of the two preparations have good scavenging potential against superoxide radicals, hydroxyl radicals, and singlet oxygen. They protect low-density lipoprotein against copper-induced oxidation due to the copper-chelating properties and their chain-breaking abilities in lipid peroxidation. 相似文献

Plant response to one type of stress can be affected by simultaneous exposure to a second stress, for example when abiotic and biotic stresses occur together. Ten rice genotypes comprising those with bacterial blight (BB) resistance (R) genes, drought quantitative trait loci (QTLs) plus a BB R gene, and BB susceptible genotypes, were subjected to mild and moderate drought stress and plants were inoculated with two Xoo strains (PXO99 and PXO145) to simulate the challenges rice crops face under simultaneous stress of drought and BB. Plant height and dry shoot biomass were significantly reduced by drought stress treatments. The BB disease lesion lengths varied according to rice genotypes and PXO99 Xoo multiplication and spread in planta was higher compared to that of PXO145, which generally decreased under mild drought stress. Rice genotype IRBB7 (Xa7) showed less Xoo spread and a reduced Xoo multiplication under drought stress compared to the well-watered control with PXO145. In contrast, in genotypes with a different BB R gene and/or drought QTLs [IRBB4 (Xa4), IR87705–6-9-B (Xa4 + qDYT 2.2 ), IR87707–445-B-B-B (Xa4 + qDYT 2.2 + qDYT 4.1 ) and IR87707–446-B-B-B (Xa4 + qDYT 2.2 + qDYT 4.1 )], Xoo multiplication and spread in planta was higher with drought stress. This study has shown that drought stress affected rice response to the BB pathogen and the response varied according to the rice genotype. It is concluded that evaluating rice varieties under combined abiotic and biotic stresses will be the best strategy to determine biotic stress resistance durability under climate change. 相似文献

Wheat is affected by many diseases, in Germany eight fungal diseases are recorded during the cultivar registration process. For a commercially successful cultivar, therefore, at least moderate resistances to important diseases, like yellow rust (YR) and Fusarium head blight (FHB), are necessary. Additionally, in 2013 a regional stem rust (SR) epidemic occurred in Central Germany for the first time for decades. Our objective was to analyze the resistance of 36 commercially grown winter wheat cultivars to YR, FHB, and SR in three individual and one combined inoculation. Appreciable disease severities were achieved for YR and FHB at three to four locations in two years (= seven location × year combinations), for SR at one to two locations in two years (= three location × year combinations). Wheat cultivars showed a significant genotypic variation for all diseases with high heritabilities (0.90–0.95). Interaction between inoculation treatments (individual vs. combined) and wheat genotype was not significant for each of the three diseases. Accordingly, correlations between both inoculation treatments were very high (R2?=?0.95–0.99). Several cultivars showed multi-disease resistance (MDR) to YR, FHB, and SR. In conclusion, resistance ranking among genotypes was not changed when plants were challenged with all three pathogens together compared to factorial inoculations of only one of them. Substituting factorial inoculation trials by multi-pathogen inoculation makes it more efficient to select for MDR in practical breeding programs. 相似文献

A decrease of the concentration of the synthetic avenanthramide N‐(4′‐hydroxy‐(E)‐cinnamoyl)‐5‐hydroxyanthranilic acid in a buffered slurry of milled oat groats (Avena sativa L.) was temperature and pH‐dependent, with a maximum rate at 30°C and pH 9. The reaction was inhibited in the presence of 2‐mercaptoethanol, acetic acid and at high temperature; suggestive of enzymatically catalyzed nature. Among eight different synthetic avenanthramides tested, the tentative enzyme had highest affinity for avenanthramides comprising caffeic and p‐coumaric acids and lowest for those comprising sinapic and ferulic acids. The activity was found in samples from several oat cultivars and was equally pronounced in both bran and endosperm flour of oats. Steeping of oat grains did not influence the reaction. 相似文献

Canine meningiomas are currently graded using the human grading system. Recently published guidelines have adapted the human grading system for use in dogs. The goal of this study was to validate the new guidelines for canine meningiomas. To evaluate the inter-observer agreement, 5 veterinary surgical pathologists graded 158 canine meningiomas following the human grading system alone or with the new guidelines. The inter-observer agreement for histologic grade and each of the grading criteria (mitotic grade, invasion, spontaneous necrosis, macronucleoli, small cells, hypercellularity, pattern loss and anaplasia) was evaluated using the Fleiss kappa index. The diagnostic accuracy (sensitivity and specificity) was assessed by comparing the diagnoses obtained with the 2 grading systems with a consensus grade (considered the reference classification). The consensus histologic grade was obtained by agreement between 4 experienced veterinary neuropathologists following the guidelines. Compared with the human grading alone, the canine-specific guidelines increased the inter-observer agreement for: histologic grade (κ = 0.52); invasion (κ = 0.67); necrosis (κ = 0.62); small cells (κ = 0.36); pattern loss (κ = 0.49) and anaplasia (κ = 0.55). Mitotic grade agreement remained substantial (κ = 0.63). The guidelines improved the sensitivity in identifying grade 1 (95.6%) and the specificity in identifying grade 2 (96.2%) meningiomas. In conclusion, the new grading guidelines for canine meningiomas are associated with an overall improvement in the inter-observer agreement and higher diagnostic accuracy in diagnosing grade 1 and grade 2 meningiomas. 相似文献

The three main oat avenanthramides, N‐(4′‐hydroxy)‐(E)‐cinnamoyl‐5‐hydroxyanthranilic acid ( Bp ), N‐(4′‐hydroxy‐3′‐methoxy)‐(E)‐cinnamoyl‐5‐hydroxyanthranilic acid ( Bf ), and N‐(3′,4′‐dihydroxy)‐(E)‐cinnamoyl‐5‐hydroxyanthranilic acid ( Bc ), and their corresponding cinnamic acids, p‐coumaric ( P ), ferulic ( F ), and caffeic ( C ), were investigated for stability to pH, temperature, and UV‐light treatment. The retention of the avenanthramides after processing of oat‐based food products was also analyzed. The avenanthramide Bc and the cinnamic acid C were sensitive to alkali and neutral conditions, especially in combination with heat treatment, whereas the other compounds studied were more stable. The cinnamic acids but not the avenanthramides were isomerized when irradiated with UV‐light. The avenanthramides were restored after processing of oat‐based products. 相似文献
