The analysis of proanthocyanidin cleavage products after acid-catalysis in the presence of excess phloroglucinol was investigated. In the developed analytical method, a solution of 0.1 N HCl in methanol, containing 50 g/L phloroglucinol and 10 g/L ascorbic acid was prepared. The proanthocyanidin of interest was reacted in this solution (5 g/L) at 50 degrees C for 20 min, and afterward combined with 5 volumes of 40 mM aqueous sodium acetate before analysis by reversed-phase HPLC using an aqueous acetic acid and methanol gradient. This procedure was used to investigate the composition of proanthocyanidins isolated from the seed and skin tissue of Vitis vinifera L. berries. The results compared favorably to results obtained when benzyl mercaptan was used as the trapping nucleophile, indicating that phloroglucinol is an effective reagent for this analysis.  相似文献

Parkinson's disease (PD) has been proposed to result from the interaction of aging and environment in susceptible individuals. The prevalence of PD in Quebec was 84 per 100 000 in 1985. The regional variation observed has been linked with several environmental factors. We intended to verify the existence of a relation between the development of PD with exposure to 3 metal pollutants: Fe, Mn, and Al. The choice of these metals rests on their potential presence in the environment caused by the industrial activity in the region studied, and on the fact that it theoretically favors the prevalence of the illness. Environmental exposure to these metals and the resulting risks were evaluated using red spruce as a bio-indicator. The metal concentrations in tree rings were determined by neutron activation analysis. These concentrations permitted a temporal estimation of contamination in each of the territories studied, from 1938 to 1987. The risk of environmental exposure was assumed to be proportional to the concentration of the metal. The calculated risks combined with the history of residence allowed a comparison between parkinsonians and controls. It appeared that environmental exposure to the metals was not significantly different between those two groups.  相似文献

This study investigated the effect of management on -glucosidase, -glucosaminidase, alkaline phosphatase, and arylsulfatase activities and the microbial community structure in semiarid soils from West Texas, USA. Surface samples (0–5 cm) were taken from a fine sandy loam, sandy clay loam, and loam that were under continuous cotton ( Gossypium hirsutum L.) or in cotton rotated with peanut ( Arachis hypogaea L.), sorghum ( Sorghum bicolor L.), rye ( Secale cereale) or wheat ( Triticum aestivum L.), and had different water management (irrigated or dryland), and tillage (conservation or conventional). The enzyme activities were higher in the loam and sandy clay loam than in the fine sandy loam. Soil pH was not affected by management, but the soil organic C and total N contents were generally affected by the different crop rotations and tillage practices studied. The trends of the enzyme activities as affected by management depended on the soil, but in general crop rotations and conservation tillage increased the enzyme activities in comparison to continuous cotton and conventional tillage. The soil enzyme activities were significantly correlated with the soil organic C ( r -values up to 0.90, P< 0.001), and were correlated among each other ( r -values up to 0.90, P <0.001). There were differences in the fatty acid methyl ester profiles between the fine sandy loam and the sandy clay loam and loam, and they reflected the differences in the enzyme activities found among the soils. For example, a 15:0 ranged from 1.61±0.25% in cotton-peanut/irrigated/no-till in the fine sandy loam to 3.86±0.48% in cotton-sorghum/dryland/conservation tillage in the sandy clay loam. Ovine vulvitis was experimentally reproduced by intravaginal inoculation of a mixture of bacterial strains of the histophilus/haemophilus group isolated from field cases. Analysis of selection experiments using mixed model methodology   总被引:2,自引:0,他引:2  
Use of mixed model techniques to estimate genetic variance and selection response is illustrated by simple examples. A minimum variance quadratic unbiased estimator (MIVQUE) of genetic variance using a reduced animal model is derived. Properties of the mixed model estimator of response are discussed and illustrated with results from Monte Carlo simulation. The mixed model estimator of response requires knowledge of the base population heritability. When the latter is not known, simulation results suggest that using a MIVQUE estimate obtained from the data yields estimates of response in good agreement with the true response. If a number of conditions are satisfied, the mixed model estimator of response partitions the phenotypic trend into its genetic and environmental components, without need for a control population. These conditions are unlikely to hold in long-term selection experiments. More work is needed to understand the implications of finite numbers of loci or the presence of unaccounted natural selection opposing artificial selection, for example, on the properties of the mixed model estimator of response.  相似文献

Fifteen cows among a herd of 50 suckler cows and calves rapidly lost body condition and became dull and anorexic after grazing pasture containing bog asphodel (Narthecium ossifragum) during the summer of 1989. The affected cows had evidence of kidney damage characterised by elevated plasma urea and creatinine concentrations. Eleven cows died and diffuse renal tubular necrosis was present in three cows which were examined post mortem. Similar renal lesions were reproduced experimentally by feeding bog asphodel to a healthy calf.  相似文献

Recombinant bovine somatotropin (bST) was administered at 0, 10.3, or 20.6 mg per cow per day to 32 Holsteins for 38 wk. Fifteen currently treated cows had been treated in the previous lactation. Eighteen hematological variables and blood concentration of insulin-like growth factor I (IGF-I) were measured at five sample periods. The objectives of the study were to test the effects of bST treatments on hematological profiles and to relate blood IGF-I concentration to these variables. Results indicated little influence (P greater than .10) of previous bST treatments on hematological profiles measured in the current lactation. Current bST treatments, however, altered hematocrits (P = .10), mean corpuscular volume (P = .03), mean corpuscular hemoglobin (P = .009), and fraction of blood lymphocytes (P = .09). A week x bST treatment interaction also contributed to variation in hematocrit (P = .09), mean corpuscular hemoglobin (P = .05), fraction of neutrophils (P = .02), and fraction of lymphocytes (P = .04). Blood IGF-I concentration influenced fractions and counts of neutrophils (P = .06, .09), lymphocytes (P = .04, .02), and monocytes (P = .08, .05). Further in vivo and in vitro studies are warranted because this preliminary evidence suggests that bST and(or) IGF-I contribute to regulation of hematopoiesis in mature dairy cows.  相似文献

Effects of age, season and sire on the occurrence of common diseases of Holstein-Friesian cows were evaluated in a two and a half year study. The data were from 2875 lactation records from 2008 cows in 32 commercial dairy herds within a 50 mile radius of Guelph, Ontario.Age patterns for all disease conditions were determined using χ2 or quadratic regression analyses. Linear increases with age were observed for milk fever, mastitis requiring local therapy only, cystic ovaries and foot and leg problems. Quadratic relationships with age were observed for ketosis (clinical and subclinical), mastitis requiring systemic therapy, subclinical mastitis, dystocia, respiratory diseases and traumatic reticulo-peritonitis. No consistent age patterns were found for retained placentas, reproductive tract infections, abomasal displacements, teat injuries, digestive disorders and abortions.Seasonal patterns were evaluated using a formal statistical test for the seasonality of events. Significant peaks were found in the risk of ketosis and cystic ovaries during the winter, and in the risk of teat injuries in the late winter or early spring. Seasonal patterns, more complex than a simple unimodal pattern, were observed for mastitis requiring local therapy only, traumatic reticulo-peritonitis and abortions.Heritabilities were determined for 9 disease conditions. All heritabilities were low or 0 with the exception of clinical ketosis (h2 = 0.31).  相似文献

Lambs premunized with a relatively less pathogenic (RLP) isolate of Haemonchus contortus were challenge exposed each day with 500 3rd-stage larvae (L3) of a normally pathogenic isolate of the same worm, starting on day 9 of the premunition-induced infection. In 6-month-old lambs, there was a fourfold decrease in egg production by the premunizing RLP isolate with challenge exposure compared with egg production in lambs given only the premunizing RLP isolate and not challenge exposed. Premunized lambs were able to carry larger worm burdens with less hemoglobin loss than were nonpremunized challenge-exposed control lambs. Although challenge exposure had affected parasitic egg production by the 9-day-old premunizing RLP worms, the same young infective isolates were able to survive and protect lambs against the additional parasitism.  相似文献
