We genotyped Setaria italica Dehydration-responsive element-binding 2 (SiDREB2) gene, which had been reported to be associated with dehydration stress tolerance, in 588 accessions of foxtail millet from various parts of Eurasia and other regions by a dCAPS marker. Of these, 480 accessions were genotyped in ribosomal DNA (rDNA), polyphenol oxidase (PPO) gene and Heading date 1 (HD1) gene in our previous studies, and 108 accessions from India were newly used in this study and genotyped in these genes in addition to the SiDREB2 gene. We compared the geographical distribution of the SiDREB2 genotypes with that of these three genes. Accessions from countries in South Asia including India and Sri Lanka were the most variable in the SiDREB2 gene followed by Korean accessions and Japanese accessions, suggesting that Indian accessions and East Asian accessions are useful as genetic resources for dehydration stress tolerance. This study also showed that although Indian accessions are not so diverse in rDNA and in transposon display markers previously studied, they are diverse in adaptive genes such as SiDREB2, PPO and HD1 genes.  相似文献

BACKGROUND

Diagnosis of acute intoxication with organophosphate (OP) or carbamate (CM) pesticides in humans is achieved by measuring plasma butyrylcholinesterase (BuChE) activity. However, BuChE activity is not an ideal biomarker in experimental animal models. The aim of this study was to establish an experimental mouse model for evaluating exposure to OP and CM pesticides by monitoring BuChE activity using chimeric mice in which the liver was reconstituted with human hepatocytes.

RESULTS

A single oral administration of acephate (300 mg/kg), chlorpyrifos (10 mg/kg), fenobucarb (300 mg/kg) or molinate (250 mg/kg) in chimeric mice led to inhibition of >95%, > 95%, 28% and 60% of plasma BuChE activity after 7, 0.5, 0.5 and 7 h, respectively. Dose‐dependent decreases in plasma BuChE activity were also observed for acephate and chlorpyrifos. A 5‐day repeated‐dose study with 10 or 30 mg/kg acephate found a constitutive reduction in plasma BuChE activity to 80% and 70% of pre‐dose levels, respectively.

CONCLUSION

Changes in plasma BuChE activity in chimeric mice with humanized liver clearly reflected the exposure levels of OP and CM pesticides. These results suggest that the humanized‐liver mouse model may be suitable for estimating levels of exposure to these pesticides in humans. © 2017 The Authors. ABSTRACT

To successfully afforest coastal forest belts ensuring high disaster prevention, growth bases for them have been constructed by piling up soil in the low wetlands along the Kujukuri coastline. Ground surfaces in such bases are often covered with water because of soil compaction, leaving them susceptible to stagnant water. Water stagnation in soil is problematic, potentially interfering with afforesting coastal forests. Therefore, row deep tillage was conducted for parts of growth bases to combat the poor physical properties of the existing soil. Here, we surveyed soil profiles and measured vertical soil hardness distribution in two forest stands with piling up soil to evaluate the effectiveness of tillage for man-made soil. Soil hardness measurements indicated that the vertical areas with ‘soft’ and ‘hard/consolidated’ soil alternately appeared in growth base profiles. Generally, soil of the dense and very hard layers was apt to be formed by strong compaction of the filled-up soil because of heavy machinery usage during growth base preparation. Such dense and hard soils in the untilled areas of the profiles were also observed in this study. By contrast, it was confirmed that row deep tillage drastically improved soil physical properties, i.e., decreased hardness and increased water permeability, because the sequential hardened subsoil layers were well-broken-up. Moreover, it was observed many thick and large roots penetrated deeper layers with deep tillage areas. These results suggest that row deep tillage of hardened soil is quite effective at securing the areas and providing the physical conditions for deeply penetrated roots into deeper soils, which increases healthy root development. They show the effects of soil hardness reduction and water permeability improvement have been maintained for two decades, at least, after construction. These findings will be useful for alleviating some problems of soil compaction, water stagnation, and tree-growth hindrance that have been encountered on afforestation sites with man-made soil.  相似文献

BackgroundMiniature Dachshunds (MD) are predisposed to lymphoma with disease onset of young age and long‐term survival.ObjectivesTo compare clinical features and survival time of lymphoma in MD and non‐MD.AnimalsOne hundred and eight MDs with lymphoma and 149 non‐MD breed dogs with lymphoma were included in the study.MethodsThis was a retrospective multicenter observational study. Lymphoma was classified based on signalment, histopathology/cytology, and anatomical site of the disease. For each type of lymphoma, median survival time was analyzed by Kaplan‐Meier estimates and life table analysis. Prognostic factors for large‐cell gastrointestinal lymphoma (LGIL) were analyzed using Cox regression.ResultsGastrointestinal lymphomas were more common in MDs (53/108) compared to non‐MDs (41/149). The multicentric lymphoma was most common in non‐MD breed dogs (74/149) compared to MDs (33/108). The median age that dog developed lymphoma in MD and non‐MD were both 10 years old; however, lymphomas were more frequently observed in younger dogs (<4 years) in MDs (20/108) compared to non‐MDs (9/149; P = .002). Seventy percent were diagnosed with B‐cell with median age of diagnosis was 3 (1‐14) years. Mott cell differentiation was observed in 6 dogs. Age <4 years and B‐cell phenotype were significant factors for longer survival time in MD with LGIL.Conclusions and Clinical ImportanceLymphomas in MDs involved gastrointestinal lesions at higher frequency compared to other dog breeds examined. B‐cell lymphoma was more common in early‐onset LGIL in MD and cases that involved Mott cell differentiation were observed. Awareness of this specific presentation of lymphoma in dogs will possibly affect the treatment decision process for the owners of MD with LGIL.  相似文献

The determination of damaged starch and diastatic activity in flour was studied using a flow-injection analysis (FIA) biosensor system. The system consisted of an oxygen electrode and an immobilized enzyme column containing purified glucoamylase and glucose oxidase immobilized on activated aminopropyl glass beads. The biosensor system has an optimum pH between 6.5 and 7.5 and an optimum temperature of 35°C for glucose measurement. The response of the FIA biosensor was linear up to 1.000 g/L of glucose with a lower detection limit of 0.025 g/L. Each assay took about 20 min, and the system showed good reproducibility (r = 0.998, n = 8). When applied to the measurement of damaged starch and diastatic activity in wheat flour, the results obtained agreed with those obtained using the conventional methods of measurement. This biosensor system is a rapid practical alternative for the measurement of damaged starch and diastatic activity in wheat flour.  相似文献
