In this study, the expressions of VEGF in dog follicles were detected by immunohistochemistry and the effects of VEGF treatment on the primordial to primary follicle transition and on subsequent follicle progression were examined using a dog ovary organ culture system. The frozen‐thawed canine ovarian follicles within slices of ovarian cortical tissue were cultured for 7 and 14 days in presence or absence of VEGF. After culture, the ovaries were fixed, sectioned, stained and counted for morphologic analysis. The results showed that VEGF was expressed in the theca cells of antral follicles and in the granulosa cells nearest the oocyte in preantral follicle but not in granulosa cells of primordial and primary follicles; however, the VEGF protein was expressed in CL. After in vitro culture, VEGF caused a decrease in the number of primordial follicles and concomitant increase in the number of primary follicles that showed growth initiation and reached the secondary and preantral stages of development after 7 and 14 days. Follicular viability was also improved in the presence of VEGF after 7 and 14 days in culture. In conclusion, treatment with VEGF was found to promote the activation of primordial follicle development that could provide an alternative approach to stimulate early follicle development in dogs.  相似文献

Cross-laminated timber (CLT) panels consist of several layers of lumber stacked crosswise and glued together on their faces. Prototype sugi CLT floor panels were manufactured and bending tests were carried out under the different parameters of lumber modulus of elasticity (MOE), number of layers, thickness of lumber and thickness of CLT panels. On the basis of above tests, bending stiffness and moment carrying capacity were predicted by Monte Carlo method. MOE of lumber was measured by using grading machine and tensile strength of lumber was assumed to be 60 % of bending strength based on the obtained bending test. Bending stiffness EI of CLT panels could be estimated by adopting composite theory and equivalent section area. Experimental moment carrying capacity showed 12 % higher value than the calculated moment carrying capacity by average lumber failure method, and also showed 45 % higher value than the calculated moment carrying capacity by minimum lumber failure method due to the reinforcement of the outer layer by the neighboring cross layer.  相似文献

ABSTRACT:   To utilize fisheries waste products as food materials with functional properties, shrimp head protein hydrolysates (SHPH) from three species of shrimp, that is, Northern pink shrimp ( Pandalus eous ), Endeavour shrimp ( Metapenaeus endeavouri ) and Black tiger shrimp ( Penaeus monodon ), were produced by enzymatic hydrolysis using endopeptidase derived from Bacillus subtilis and exopeptidase derived from Aspergillus oryzae at a level of 0.1% (w/w). SHPH were rich in protein (90–91%) and amino acids (71–84%) but little fat (0.01–0.02%). The average molecular weight of SHPH was 300–1400. The effect of 5% SHPH (dry basis) addition on the state of water and denaturation of lizard fish myofibrils (Mf) during the dehydration process was evaluated by the desorption isotherm and the Ca-ATPase activity, and compared with the effect of sodium glutamate (Na-Glu). SHPH decreased the water activity and the Ca-ATPase inactivation, and increased monolayer sorbed water and multilayer sorbed water of Mf, although these effects of SHPH were smaller than those of Na-Glu. These findings suggest that the SHPH suppressed dehydration-induced denaturation of myofibrillar protein by stabilizing the hydrated water surrounding myofibrils.  相似文献

A major alginate lyase, FlAlyA, was purified from the periplasmic fraction of an alginate-assimilating bacterium, Flavobacterium sp. strain UMI-01. FlAlyA showed a single band of ~30 kDa on SDS-PAGE and exhibited the optimal temperature and pH at 55 °C and pH 7.7, respectively. Analyses for substrate preference and reaction products indicated that FlAlyA was an endolytic poly(mannuronate) lyase (EC 4.2.2.3). A gene fragment encoding the amino-acid sequence of 288 residues for FlAlyA was amplified by inverse PCR. The N-terminal region of 21 residues except for the initiation Met in the deduced sequence was predicted as the signal peptide and the following region of six residues was regarded as propeptide, while the C-terminal region of 260 residues was regarded as the polysaccharide-lyase-family-7-type catalytic domain. The entire coding region for FlAlyA was subjected to the pCold I—Escherichia coli BL21(DE3) expression system and ~eight times higher yield of recombinant FlAlyA (recFlAlyA) than that of native FlAlyA was achieved. The recFlAlyA recovered in the periplasmic fraction of E. coli had lost the signal peptide region along with the N-terminal 3 residues of propeptide region. This suggested that the signal peptide of FlAlyA could function in part in E. coli.  相似文献

In the North Nile Delta of Egypt, the impacts of overplanting paddy rice on water delivery performance have not been discussed quantitatively. Further, the amount of water that could have been saved if farmers would follow the planned area is unknown. In this study, water delivery performance was assessed by comparison of actual paddy rice planting and the government’s planned conditions. For both conditions, performance indicators relating to adequacy, equity, and dependability were analyzed across six locations in conjunction with the branch canal water level in 2013 and 2014. Based on the difference between the actual water supply and planned water demand, the amount of water that could have been saved for downstream uses was calculated. The average adequacy for the investigation period was good at one location, fair at 2 locations, and poor at 3 locations in both years. Further, adequacy under both actual and planned conditions was poor in late July at all locations. The planned adequacy and dependability downstream and equity among locations improved compared to the actual condition in both years. Under the condition that paddy rice area is the upper limit planned by the government, about 12.3% and 9.6% of water could be potentially saved in each year. The difference between actual and planned water delivery performance is caused by the branch canal’s low water level. Control of overplanting paddy rice and coordination of water distribution among water user associations would improve stable water level in the canal and, eventually, water delivery performance.

Two consecutive trials were conducted to investigate the effects of photoperiod manipulation on growth rate, food intake and feed conversion efficiency (Trial 1), and the digestibility of nutrients and energy (Trial 2) in red sea bream, Pagrus major (body weight 19–120 g). Fish were exposed to four photoperiods (6L:6D, 12L:12D, 16L:8D and 24L:0D) with light intensity 1500 lx on the water surface. The fish were fed with a commercial diet to apparent satiation. In Trial 2, 0.5% chromic oxide (Cr2O3) was used as an inert marker. Significantly higher weight gain and specific growth rates were observed in fish exposed to a 24L:0D photoperiod followed by 16L:8D, 6L:6D and 12L:12D photoperiods (P < 0.05). Food intake and feed conversion efficiency (FCE) were also significantly higher in fish exposed to 24L:0D followed by 16L:8D, 6L:6D and 12L:12D photoperiods (P < 0.05). Fish exposed to 24L:0D and 16L:8D photoperiods showed significantly higher lipid and energy digestibility than those exposed to a 12L:12D photoperiod (P < 0.05). The results demonstrated that the enhancement of growth performances under 16L:8D and 24L:0D photoperiods were attributed to improved appetite, greater food intake and higher feed conversion efficiency as well as higher digestibility.  相似文献
