T cell receptor (TCR) and costimulatory receptor (CD28) signals cooperate in activating T cells, although understanding of how these pathways are themselves regulated is incomplete. We found that Homer2 and Homer3, members of the Homer family of cytoplasmic scaffolding proteins, are negative regulators of T cell activation. This is achieved through binding of nuclear factor of activated T cells (NFAT) and by competing with calcineurin. Homer-NFAT binding was also antagonized by active serine-threonine kinase AKT, thereby enhancing TCR signaling via calcineurin-dependent dephosphorylation of NFAT. This corresponded with changes in cytokine expression and an increase in effector-memory T cell populations in Homer-deficient mice, which also developed autoimmune-like pathology. These results demonstrate a further means by which costimulatory signals are regulated to control self-reactivity. 相似文献

Mercury has contaminated rivers worldwide, with health consequences for aquatic organisms and humans who consume them. Researchers have focused on aquatic birds as sentinels for mercury. However, trophic transfer between adjacent ecosystems could lead to the export of aquatic mercury to terrestrial habitats. Along a mercury-contaminated river in Virginia, United States, terrestrial birds had significantly elevated levels of mercury in their blood, similar to their aquatic-feeding counterparts. Diet analysis revealed that spiders delivered much of the dietary mercury. We conclude that aquatic mercury pollution can move into terrestrial habitats, where it biomagnifies to levels in songbirds that may cause adverse effects. Rivers contaminated with mercury may pose a threat to the many bird species that feed on predatory invertebrates in adjacent riparian habitats. 相似文献

Naked metallic and semiconducting single-walled carbon nanotubes (SWNTs) were dissolved in organic solutions by derivatization with thionychloride and octadecylamine. Both ionic (charge transfer) and covalent solution-phase chemistry with concomitant modulation of the SWNT band structure were demonstrated. Solution-phase near-infrared spectroscopy was used to study the effects of chemical modifications on the band gaps of the SWNTs. Reaction of soluble SWNTs with dichlorocarbene led to functionalization of the nanotube walls. 相似文献

NKT cells represent a distinct lineage of T cells that coexpress a conserved alphabeta T cell receptor (TCR) and natural killer (NK) receptors. Although the TCR of NKT cells is characteristically autoreactive to CD1d, a lipid-presenting molecule, endogenous ligands for these cells have not been identified. We show that a lysosomal glycosphingolipid of previously unknown function, isoglobotrihexosylceramide (iGb3), is recognized both by mouse and human NKT cells. Impaired generation of lysosomal iGb3 in mice lacking beta-hexosaminidase b results in severe NKT cell deficiency, suggesting that this lipid also mediates development of NKT cells in the mouse. We suggest that expression of iGb3 in peripheral tissues may be involved in controlling NKT cell responses to infections and malignancy and in autoimmunity. 相似文献

Detailed studies of elementary chemical processes on well-characterized single crystal surfaces have contributed substantially to the understanding of heterogeneous catalysis. Insight into the structure of surface alloys combined with an understanding of the relation between the surface composition and reactivity is shown to lead directly to new ideas for catalyst design. Local control of the domain orientation in diblock copolymer thin films can be obtained by the application of electric fields on micrometer-length scales. Thin films of an asymmetric polystyrene-polymethylmethacrylate diblock copolymer, with cylindrical polymethylmethacrylate microdomains, were spin-coated onto substrates previously patterned with planar electrodes. The substrates, 100-nanometer-thick silicon nitride membranes, allow direct observation of the electrodes and the copolymer domain structure by transmission electron microscopy. The cylinders aligned parallel to the electric field lines for fields exceeding 30 kilovolts per centimeter, after annealing at 250°C in an inert atmosphere for 24 hours. The time dependence of the orientation of Earth's inner core relative to the mantle was determined using a recently discovered 10-degree tilt in the axis of symmetry of the inner core's seismic-velocity anisotropy. Two methods of analyzing travel-time variations for rays traversing the inner core, on the basis of 29 years of data from the International Seismological Centre (1964-1992), reveal that the inner core appears to rotate about 3 degrees per year faster than the mantle. An anomalous variation in inner-core orientation from 1969 to 1973 coincides in time with a sudden change ("jerk") in the geomagnetic field. 相似文献

The aim of this work (from the FP6 project TRACE) was to develop methods based on the use of geochemical markers for the authentication of the geographical origin of cereal samples in Europe (cf. EC regulations 2081/92 and 1898/06). For the first time, the potential usefulness of combining n(87Sr)/n(86Sr) and δ13C, δ15N, δ18O and δ34S isotopic signatures, alone or with key element concentrations ([Na], [K], [Ca], [Cu] and [Rb], progressively identified out of 31 sets of results), was investigated through multiple step multivariate statistics for more than 500 cereal samples collected over 2 years from 17 sampling sites across Europe representing an extensive range of geographical and environmental characteristics. 相似文献

The elevated accumulation of certain essential long-chain polyunsaturated fatty acids (PUFA) during reproductive maturation can place a significant demand on lipid metabolism. As lipid metabolism is closely linked with protein metabolism, protein intake during maturation may significantly influence transport proteins and enzyme activity which is responsible for supplying the essential fatty acids for gamete development. The present study investigated the influence of increased protein intake on the accumulation of certain long-chain PUFA in two species of commercially valuable sea urchin, Paracentrotus lividus and Psammechinus miliaris during maturation. Sea urchins were fed three macroalgal diets over a 90-day period. These diets included the red alga Palmaria palmata, which had been grown in seawater enriched with two levels of ammonia-N, which produced a high protein content (41%) or low protein content (32%). The third diet (or reference diet) used the brown macroalga Laminaria saccharina with a 23% protein content. The fatty acid composition of the gonads was determined by GC. Feeding the high protein diet had no effect on the maturation and total lipid in the gonad compared with the low protein diet, however, it did significantly increase the levels of docosahexaenoic acid DHA 22:6n-3 in the mature gonad tissue in P. miliaris and P. lividus. In contrast, only P. miliaris exhibited higher levels of arachidonic acid ARA 20:4n-6 when fed the high protein diet. The present study suggests that elevated protein levels can promote the accumulation of DHA and ARA in gonadal tissue during maturation, although the extent of the influence is species-specific and highlights the importance of understanding the complexities and inter-connectivity between protein and lipid metabolism when producing commercially valuable species. 相似文献
