The aim of the current study was to determine the possible effects of progestagen oestrous synchronization on vascular endothelial growth factor (VEGF) expression during sheep luteogenesis and the peri‐implantation period and the relationship with luteal function. At days 9, 11, 13, 15, 17 and 21 of pregnancy, the ovaries from 30 progestagen treated and 30 ewes cycling after cloprostenol injection were evaluated by ultrasonography and, thereafter, collected and processed for immunohistochemical evaluation of VEGF; blood samples were drawn for evaluating plasma progesterone. The progestagen‐treated group showed smaller corpora lutea than cloprostenol‐treated and lower progesterone secretion. The expression of VEGF in the luteal cells increased with time in the cloprostenol group, but not in the progestagen‐treated group, which even showed a decrease between days 11 and 13. In progestagen‐treated sheep, VEGF expression in granulosa‐derived parenchymal lobule capillaries was correlated with the size of the luteal tissue, larger corpora lutea had higher expression, and tended to have a higher progesterone secretion. In conclusion, the current study indicates the existence of deleterious effects from exogenous progestagen treatments on progesterone secretion from induced corpora lutea, which correlate with alterations in the expression of VEGF in the luteal tissue and, this, presumably in the processes of neoangiogenesis and luteogenesis.  相似文献

The use of heavy machinery is increasing in agriculture, which induces increased risks of subsoil compaction. Hence, there is a need for technical solutions that reduce the compaction risk at high total machine loads. Three field experiments were performed in order to study the effects of dual wheels, tandem wheels and tyre inflation pressure on stress propagation in soil. Vertical soil stress was measured at three different depths by installing probes into the soil horizontally from a dug pit. In one experiment, also the stress distribution below the tyre was measured. Beneath the dual wheels, vertical stresses at 0.15 and 0.3 m depth were lower between the two wheels than under the centre of each wheel, despite the gap between the wheels being small (0.1 m). At 0.5 m depth, vertical stress beneath the wheels was the same as between the two wheels. The stress interaction from the two wheels was weak, even in the subsoil. Accordingly, measured stresses at 0.3, 0.5 and 0.7 m depth were highest under the centre of each axle centre line of tandem wheels, and much lower between the axles. For a wheel load of 86 kN, tyre inflation pressure significantly affected stress at 0.3 m depth, but not at greater depths. Stress directly below the tyre, measured at 0.1 m depth, was unevenly distributed, both in driving direction and perpendicular to driving direction, and maximum stress was considerably higher than tyre inflation pressure. Calculations of vertical stress based on Boussinesq's equation for elastic materials agreed well with measurements. A parabolic or linear contact stress distribution (stress declines from the centre to the edge of the contact area) was a better approximation of the contact stress than a uniform stress distribution. The results demonstrate that stress in the soil at different depths is a function of the stress on the surface and the contact area, which in turn are functions of wheel load, wheel arrangement, tyre inflation pressure, contact stress distribution and soil conditions. Soil stress and soil compaction are a function of neither axle load nor total vehicle load. This is of great importance for practical purposes. Reducing wheel load, e.g. by using dual or tandem wheels, also allows tyre inflation pressure to be reduced. This reduces the risk of subsoil compaction.  相似文献

Recently, isolation and in vitro culture of putative spermatogonial stem cells (SSCs) in the domestic cat have been conducted. However, the cellular niche conditions that facilitate the establishment and long‐term maintenance of feline SSCs (FSSCs) have not been described. Therefore, we investigated the type of feeder cells used to stimulate colony formation and growth of FSSCs among the various factors in the FSSC niche. Spermatogonial stem cells isolated from feline testes were cultured on mitotically inactivated testicular stromal cells (TSCs) derived from cats, dogs and mice, and mouse embryonic fibroblasts (MEFs). The formation and growth of colonies derived from SSCs cultured on each type of feeder cell were identified at passage 0, and the morphology, alkaline phosphatase (AP) activity and expression of SSC‐specific genes in surviving colonies were investigated at passage 4. Among these diverse feeder cells, TSCs from cat showed the greatest colony formation, growth and maintenance of FSSCs, and SSC colonies cultured by passage 4 showed a typical dome‐shaped morphology, AP activity and expression of SSC‐specific genes (NANOG, OCT4, SOX2 and CD9). Accordingly, these results demonstrate that feline TSCs could be used as feeder cells to support the establishment and maintenance of SSCs from domestic cats.  相似文献

Impact of crop rotation, fertilizer treatments and herbicidal applications on the long term maintenance of soil fertility
V. Comparative studies regarding chemical and physical factors of the yield potential of a soil over a ten year experimental period
In order to examine the influence of different crop management practices such as crop rotation, fertilizer treatments and herbicide applications on the yield potential of a soil, the Crop Science Department of the Swiss Federal Institute of Technology (ETH Zurich) conducted a long term field experiment commencing 1973. The experimental location was at the Swiss Federal Research Station for Farm Management and Agricultural Engeneering at Tanikon/TG (northeastern Switzerland).
After the ten year period, chemical and physical soil properties were not found to be substantially affected by the various experimental treatments. Organic carbon and total nitrogen were more influenced by fertilizer treatments than by crop rotation and herbicide applications. Both cation exchange capacity and pH proved to be fairly stable values.
Aggregate stability and organic matter exhibited similar profiles, although soil bulk density was affected by crop rotation. The volume of large and medium sized pores was similarly affected in that the corn-dominated crop rotation resulted in a quantifiable impact upon soil compaction.
Depending upon weed control efficiency, the grain yields of the winter wheat test crop clearly exhibited the effects of crop rotation whereas the lowest yields resulted from the corn-dominated crop rotation.  相似文献

Tarsal radiographs of 134 trotters were made prior to training. Radiographs were evaluated for changes commonly associated with spavin. Spavin, if present, was graded as mild, moderate, or severe. Sone of the 134 horses were radiographed annually for one, two, or three years. Radiographs of the 134 horses were compared with radiographs of 141 additional horses in which spavin had been diagnosed clinically. Only 66 of the 134 trotters (50.4%) had normal tarsal radiographs at the time of initial examination; the other 65 had some radiographic evidence of spavin. The incidence and severity of radiographic changes identified in a group of horses in which spavin was diagnosed clinically. Thus, the present results suggest that tarsal radiography is of little value in establishing the diagnosis and prognosis of spavin in trotters.  相似文献

Restriction endonuclease fingerprints of infectious laryngotracheitis virus (ILTV) DNA from 13 Pennsylvania field isolates, embryo-propagated and tissue-culture-propagated vaccine strains, and three reference strains were compared. These comparisons were made to evaluate the possible contribution of mutation of ILTV vaccine strains to recent outbreaks of infectious laryngotracheitis (ILT) in Pennsylvania. Six different restriction enzymes were used to generate the fingerprints. Differences in DNA banding patterns were revealed between the currently used ILTV vaccine strains and six of the 13 field isolates. Even greater DNA banding pattern differences were found between the older ILTV reference strains and the vaccine strains. The ILTV DNA fingerprints generated in the present study suggest that at least five different strains of ILTV have contributed to the outbreaks of ILT that have occurred since 1987 in Pennsylvania.  相似文献

The inhalation of grain dust by grain workers is responsible for a large number of pulmonary pathophysiologies. These problems may be acute or chronic and may be mediated by the chronic activation of the immune system. Constant inflammatory states in the lung may eventually lead to tissue damage and respiratory deficit. This study was designed to measure the changes in the relative number of inflammatory cells in peripheral blood, bronchoalveolar spaces, and lung interstitium that occur in response to intratracheally instilled airborne spring wheat dust in rats. It was found that 6h after instillation with dust, neutrophils were present in greater numbers in the blood and bronchoalveolar spaces than in lung interstitium. After 24h, there appeared to be a larger number of neutrophils in the lung interstitium in dust-instilled animals than in saline-instilled controls. These results indicate that intratracheal instillation of grain dust initiates an acute inflammatory reaction, and that there is an initial influx of neutrophils into the air spaces of the lung followed by transit of these cells into the lung interstitium.  相似文献
