Purpose
Nitrous oxide (N2O) is a potent greenhouse gas and, in grazed grassland systems where animals graze outdoor pastures, most of the N2O is emitted from animal urine nitrogen (N) deposited during grazing. Recently, ammonia-oxidizing archaea (AOA) were found to be present in large numbers in soils as well in the ocean, suggesting a potentially important role for AOA, in addition to ammonia-oxidizing bacteria (AOB), in the nitrogen cycle. The relationship between N2O emissions and AOB and AOA populations is unknown. Response of microbial metabolism (growth, substrate utilization, energetic metabolism) to fertilization by N and P and resulting changes in soil‐organic‐matter (SOM) decomposition (priming effect) were studied in grassland soils with relatively high organic‐matter content. Treatments with and without glucose addition were studied to simulate difference between rhizosphere and bulk soil. Our expectation was that fertilization would decrease soil respiration in both treatments due to an increased efficiency of microbial metabolism. At first, fertilization activated microbial metabolism in both treatments. In glucose‐nonamended soils, this was connected with a short‐term apparent priming effect but if glucose was available, the higher energetic demand was covered by its mineralization in preference against SOM, causing significant SOM savings as compared to unfertilized soils. After a relatively short period of 1–3 d, however, the phase of deprived microbial metabolism occurred in both treatments, which was characterized by lower soil respiration in fertilized than in unfertilized soils. Fertilization further decreased net microbial growth following glucose addition, shortened turnover time of microbial biomass and changed the partitioning of assimilated glucose within microbial biomass (decreased accumulation of storage compounds and increased the proportion of mineralized glucose). As a result, fertilization reduced soil respiration mainly due to a deprivation of microbial metabolism. The rate and range of microbial response to fertilization and also the amount of saved soil C were larger in the soil with higher SOM content, likely driven by the higher content of microbial biomass. 相似文献

Conspecific negative density-dependent establishment, in which local abundance negatively affects establishment of conspecific seedlings through host-specific enemies, can influence species diversity of plant communities, but the generality of this process is not well understood. We tested the strength of density dependence using the United States Forest Service's Forest Inventory and Analysis database containing 151 species from more than 200,000 forest plots spanning 4,000,000 square kilometers. We found that most species experienced conspecific negative density dependence (CNDD), but there was little effect of heterospecific density. Additionally, abundant species exhibited weaker CNDD than rarer species, and species-rich regions exhibited stronger CNDD than species-poor regions. Collectively, our results provide evidence that CNDD is a pervasive mechanism driving diversity across a gradient from boreal to subtropical forests. 相似文献

The Gram-positive bacterium Staphylococcus pseudintermedius is regarded as the major cause of canine bacterial pyoderma. Despite its clinical importance, there is only very limited knowledge about the pathogenesis of S. pseudintermedius infection and the specific bacterial virulence factors involved in causing disease. Using a whole-genome approach, we have previously identified 18 predicted cell-wall-anchored surface proteins representing possible virulence factors in a clinical isolate of S. pseudintermedius (strain ED99). They were designated S. pseudintermedius surface proteins A-R (SpsA-SpsR). The present study tested three of the putative Sps proteins (SpsD, SpsL and SpsO) for their ability to mediate adherence of bacteria to canine corneocytes. The three proteins were expressed on the surface of the nonpathogenic surrogate host Lactococcus lactis, a Gram-positive bacterium that does not adhere to canine corneocytes. Adherence assays were performed using corneocytes from different healthy canine donors (n = 5), and bacterial cells were quantified using computerized image analysis. Two of the proteins, SpsD and SpsO, mediated adherence of L. lactis to canine corneocytes, suggesting that they contribute to S. pseudintermedius pathogenesis and may represent novel therapeutic targets to combat infection. 相似文献

A 40 yr-old female white rhinoceros (Ceratotherium simum) suffered from chronic nail-bed abscesses. Due to worsening of clinical signs, the animal's nonsteroidal anti-inflammatory treatment was switched to firocoxib. Approximately 7 days after this change, the animal developed multifocal vesicles and bullae along the lateral aspects of the thorax and abdomen, the dorsum, and the proximal limbs. Cytology and culture did not identify an infectious etiology. Histologically, the lesions consisted of a severe, subacute vesiculobullous dermatitis with intraepidermal to subepidermal clefting with areas of individual keratinocyte necrosis and minor neutrophilic epidermal infiltrates. Ampicillin concentrations in pulmonary epithelial lining fluid (PELF) and plasma was studied after single intravenous ampicillin administration (15mg/kg) or single intragastric administration of its prodrug, pivampicillin (19.9mg/kg) to horses and discussed in relation to minimum inhibitory concentrations (MIC) of common equine respiratory pathogens. After intravenous administration, elimination of ampicillin was fast and not detectable in plasma after 12h in three out of six horses. Pivampicillin was absorbed well in non-fasted horses with an oral bioavailability of 36%. The degree of penetration of ampicillin into PELF, as described by the AUC(PELF)/AUC(plasma) ratio from 0 to 12h was 0.40 after intravenous administration and 1.00 after pivampicillin administration. In horses, ampicillin administered either intravenously or orally, in the form of pivampicillin, can provide clinically relevant drug concentrations in PELF for at least 12h, when treating susceptible equine respiratory pathogens (e.g. streptococci). Treatment of other bacterial pathogens requires susceptibility testing and possibly more frequent dosing, depending of minimum inhibitory concentrations (MIC) values. 相似文献

Comparison of taper functions between two planted and coppiced eucalypt clonal hybrids, South Africa
In addition to regeneration through seed, certain eucalypts are able to regenerate via the production of coppice shoots following
felling, which can then be selectively thinned over time and managed as a coppice stand for the commercial production of timber.
Little information could be found if tree form differs between coppiced (where one or two stems had been left per stump) and
planted eucalypts, or whether different volume taper models would need to be developed. To determine if this was necessary,
rotation-end stem taper data was collected from an Eucalyptus grandis × E. urophylla and an E. grandis × E. camaldulensis trial to compare volume taper equations for planted versus coppiced commercially grown Eucalyptus clonal hybrids. For treatment comparisons, taper data were collected from the parent crop (1R), the replanted crop (2R),
as well as from coppiced stands where either a single (Cop_Sngl) or double stem (Cop_Dbl) had been left per stump. Stem taper functions used for treatment comparisons indicated differences between clones, as well as between treatment, with
the models based on single stems (1R, 2R or Cop_Sngl) being significantly different from those containing two stems (Cop_Dbl).
Despite any non-significant model differences (for example between 1R, 2R and Cop_Sngl single stem crops) the percentage magnitude
of any bias in utilizable volume differences when comparing the individual models (between a 6.31% over-prediction to a 3.8%
under-prediction) still needs to be taken into account as these differences may have importance in terms of the volume and
