AbstractThe carbon (C) budget of managed grassland in a cool-temperate region of Japan was estimated using a combination of eddy covariance and the biometric method for five years, to evaluate the effect of manure application. Chemical fertilizer was applied to the fertilizer (F) plot at a rate of 79 ± 20 kg N ha?1 yr?1. In the manure (M) plot, dairy cattle manure was applied at a rate of 10 Mg fresh matter ha?1 yr?1 (1923 ± 407 kg C ha?1 yr?1, 159 ± 68 kg N ha?1 yr?1). There was no significant difference in seasonal gross primary production (GPP) and harvest between the treatment plots, indicating that both fertilizer and manure can increase the biomass production. Annual net ecosystem production (NEP) and ecosystem respiration (RE) was significantly different between the treatment plots. The difference in RE, and between M and F plots approximates heterotrophic respiration of manure (RHm), which ranged from 0.9 to 1.3 Mg C ha?1 yr?1. Average annual RHm was 1.1 ± 0.4 Mg C ha?1 yr?1, and accounted for 56% of the total amount of applied manure C. The annual net biome production (NBP) in the M plot (from 0.0 to 1.5 Mg C ha?1 yr?1) was significantly higher than in the F plot (?1.4 to 0.5 Mg C ha?1 yr?1). The long-term effect of manure application combined with chemical fertilizer did not reduce grass production compared with chemical fertilizer only; however, manure application decreased the NEP throughout manure decomposition, and long-term manure application enhanced the NBP. 相似文献

Neoculin occurring in an edible tropical fruit is a heterodimeric protein which has both sweetness and a taste-modifying activity that converts sourness to sweetness. Both the primary and the overall tertiary structures of neoculin resemble those of monocot mannose-binding lectins. This study investigated differences in biochemical properties between neoculin and the lectins. Structural comparison between the mannose-binding sites of lectins and the corresponding regions of neoculin showed that there is at least one amino acid substitution at each site in neoculin, suggesting a reason for the lack of its mannose-binding ability. This was consistent with hemagglutination assay data demonstrating that neoculin had no detectable agglutinin activity. DNA microarray analysis indicated that neoculin had no significant influence on gene expression in Caco-2 cell, whereas kidney bean lectin (Phaseolus vulgaris agglutinin) greatly influenced various gene expressions. These data strongly suggest that neoculin has no lectin-like properties, encouraging its practical use in the food industry. 相似文献

Leaf area index (LAI) is one of the major determinants of crop photosynthesis. The objectives of this study were to clarify the relationship between LAI development and crop growth in diverse rice genotypes grown under widely different climate conditions and to develop a model explaining genotypic and environmental variation in LAI dynamics based on environmental and plant factors. Cross-locational experiments were conducted with nine different rice genotypes at eight locations in Asia covering a wide climate range under irrigated conditions with sufficient nitrogen application. The LAI observed at the heading stage ranged from 0.85 to 8.77 among the genotypes grown at the eight locations. A fairly stable allometric relationship was observed between LAI development and above-ground biomass growth during the period from transplanting to 2 weeks before heading over all the genotypes, sites and years (r = 0.91). The allometric relationship was, however, under the influence of leaf nitrogen content per unit leaf area (LNC, g m−2 leaf) and air temperature. On the basis of these results, we modeled the LAI development as a function of relative crop growth rate (RGR), LNC and air temperature. The rate of LAI decrease associated with leaf senescence was also described as a function of LNC. 相似文献

The concept of aerobic culture is to save water resource while maintaining high productivity in irrigated rice ecosystem. This study compared nitrogen (N) accumulation and radiation use efficiency (RUE) in the biomass production of rice crops in aerobic and flooded cultures. The total water input was 800–1300 mm and 1500–3500 mm in aerobic culture and flooded culture, respectively, and four high-yielding rice cultivars were grown with a high rate of N application (180 kg N ha−1) at two sites (Tokyo and Osaka) in Japan in 2007 and 2008. The aboveground biomass and N accumulation at maturity were significantly higher in aerobic culture (17.2–18.5 t ha−1 and 194–233 kg N ha−1, respectively) than in flooded culture (14.7–15.8 t ha−1 and 142–173 kg N ha−1) except in Tokyo in 2007, where the surface soil moisture content frequently declined. The crop maintained higher N uptake in aerobic culture than in flooded culture, because in aerobic culture there was a higher N accumulation rate in the reproductive stage. RUE in aerobic culture was comparable to, or higher than, that in flooded culture (1.27–1.50 g MJ−1 vs. 1.20–1.37 g MJ−1), except in Tokyo in 2007 (1.30 g MJ−1 vs. 1.37 g MJ−1). These results suggest that higher biomass production in aerobic culture was attributable to greater N accumulation, leading to higher N concentration (N%) than in flooded culture. Cultivar differences in response to water regimes were thought to reflect differences in mainly (1) early vigor and RUE under temporary declines in soil moisture in aerobic culture and (2) the ability to maintain high N% in flooded culture. 相似文献

In recent years, there has been considerable interest in lectins from marine invertebrates. In this study, the biological activities of a lectin protein isolated from the eggs of Sea hare (Aplysia kurodai) were evaluated. The 40 kDa Aplysia kurodai egg lectin (or AKL-40) binds to D-galacturonic acid and D-galactose sugars similar to previously purified isotypes with various molecular weights (32/30 and 16 kDa). The N-terminal sequence of AKL-40 was similar to other sea hare egg lectins. The lectin was shown to be moderately toxic to brine shrimp nauplii, with an LC50 value of 63.63 µg/mL. It agglutinated Ehrlich ascites carcinoma cells and reduced their growth, up to 58.3% in vivo when injected into Swiss albino mice at a rate of 2 mg/kg/day. The morphology of these cells apparently changed due to AKL-40, while the expression of apoptosis-related genes (p53, Bax, and Bcl-XL) suggested a possible apoptotic pathway of cell death. AKL-40 also inhibited the growth of human erythroleukemia cells, probably via activating the MAPK/ERK pathway, but did not affect human B-lymphoma cells (Raji) or rat basophilic leukemia cells (RBL-1). In vitro, lectin suppressed the growth of Ehrlich ascites carcinoma and U937 cells by 37.9% and 31.8%, respectively. Along with strong antifungal activity against Talaromyces verruculosus, AKL showed antibacterial activity against Staphylococcus aureus, Shigella sonnei, and Bacillus cereus whereas the growth of Escherichia coli was not affected by the lectin. A replicate heart-cut column-switching HPLC method combined with two switching valves was newly developed for the simultaneous determination of three antihypertensive peptides (Ala-Phe, Tyr-Pro, and Trp-Tyr) in tryptic casein hydrolysate in one run-in assay. After a first separation on an octadecyl silane (ODS) column, heart-cuts of each peptide were individually separated on a subsequent analytical ODS column: 26% acetonitrile for Ala-Phe and Tyr-Pro (32% for Trp-Tyr) in 0.1% trifluoroacetic acid containing 10 mM sodium 1-octanesulfonate at 0.8 mL/min. Ala-Phe, Tyr-Pro, and Trp-Tyr in casein hydrolysate were determined within 70 min to be 0.377 +/- 0.037 mg/g, 2.50 +/- 0.26 mg/g, and 0.096 +/- 0.008 mg/g, respectively. 相似文献

Spontaneous iron accumulation in hepatocytes was observed in a 7-week-old female Han Wistar GALAS rat. Very fine yellowish brown pigments, which showed a positive reaction with Berlin Blue stain, were apparent in the cytoplasm close to the bile canaliculi, with a diminishing periportal-to-centrilobular gradient. There were also differences in distribution between and within lobes. Transmission electron microscopy revealed cytosolic ferritin and pericanalicular siderosomes in hepatocytes. No degeneration or necrotic changes were observed, and non-hepatocyte cells did not demonstrate any obvious accumulation of iron. There were no abnormalities in the animal other than this finding in the liver. 相似文献

We investigated the expansion of NO3 ?-contaminated groundwater in the Sichuan Basin, China. Nitrogen concentrations and isotopic ratios of NH4 + and NO3 ? were analyzed in groundwater and rain collected from four areas in this basin in order to evaluate the sources of nitrogen pollution. NH4 + in rain, for which δ15N values are strongly negative to slightly positive ?13.4 to + 2.3‰, appears to originate from fertilizers and excretory waste. NO3 ? in rain (δ15N: ?10.2 to ?4.4‰) was attributed to NO x from automobile exhaust gas. In the studied area, well water sampled from farmyards was found to have the highest δ15NNO3 (average: +9.7 ± 4.7‰), indicating contamination by domestic sewage as animal excrement. The lowest δ15NNO3 (?0.2 ± 3.7‰), found in spring water, indicates that the studied groundwater samples are widely affected by air contaminants (mainly as nitrogen oxides) resulted from fuel combustions. The δ15NNO3 (+3.7 ± 2.1‰) values of well water from farmland are between these levels, suggesting that NO3 ? contamination results primarily from cultivation using nitrogen fertilizers, although the contribution from animal excrement cannot be excluded. These results demonstrate that the studied groundwater is widely polluted by locally derived nitrogen sources. 相似文献

The canine Bcl-xL gene was cloned and sequenced. Canine Bcl-xL cDNA clone was 1252 bp in length, and encoded 233 deduced amino acides. The predicted canine Bcl-xL amino acid sequence shared 99.6%, 97.0%, 97.9%, 98.7% and 98.3% homology with that of human, mouse, rat, sheep and pig Bcl-xL, respectively. RT-PCR analysis revealed that canine Bcl-xL mRNA was constitutively expressed in CL-1 (canine lymphoma) and GL-1 (canine B cell leukemia) cell lines. 相似文献
