The current model of apoptosis holds that upstream signals lead to activation of downstream effector caspases. We generated mice deficient in the two effectors, caspase 3 and caspase 7, which died immediately after birth with defects in cardiac development. Fibroblasts lacking both enzymes were highly resistant to both mitochondrial and death receptor-mediated apoptosis, displayed preservation of mitochondrial membrane potential, and had defective nuclear translocation of apoptosis-inducing factor (AIF). Furthermore, the early apoptotic events of Bax translocation and cytochrome c release were also delayed. We conclude that caspases 3 and 7 are critical mediators of mitochondrial events of apoptosis.  相似文献

Development of the thyroid gland of the Japanese eel (Anguilla japonica) was studied with the use of tank-reared fish. A single thyroid follicle was first found in larvae at 29 days post hatching (dph), total length (TL) 12 mm. Until reaching 25 mm in TL (100 dph), leptocephali had one or two follicles per individual. The inner colloid of the follicles was weakly immuno-positive against the anti-thyroxine (T4) antibody. The number of thyroid follicles and the immunoreactivity later increased as the larvae grew. Thyroid hormones (TH) T4 and triiodothyronine (T3) were not detected in premetamorphic larvae by radioimmunoassays, but became detectable during metamorphosis. The maximum level of T4 was seen in fish at the end of metamorphosis and in just-metamorphosed juveniles, whereas T3 reached the highest level during metamorphosis and declined toward the end of metamorphosis. The results indicated that the thyroid gland first became active during metamorphosis in the development of eel.  相似文献

Crop physiological traits of Liangyoupeijiu, a “super” hybrid rice variety recently bred in China, were compared with those of Takanari and Nipponbare in 2003 and 2004 in Kyoto, Japan. Liangyoupeijiu showed a significantly higher grain yield than Nipponbare in both years, and achieved a grain yield of 11.8 t ha−1 in 2004, which is the highest yield observed under environmental conditions in Kyoto. Liangyoupeijiu had longer growth duration and larger leaf area duration (LAD) before heading, causing larger biomass accumulation before heading than the other two varieties. Liangyoupeijiu had a large number of grains and translocated a large amount of carbohydrates from the vegetative organ to the panicle during the grain filling period. The three yield components measured were panicle weight at heading (P0), the amount of carbohydrates translocated from the leaf and stem to the panicle during the grain filling period (ΔT), and the newly assimilated carbohydrates during grain filling (ΔW). It was found that the sum of P0 and ΔT were strongly correlated with grain yield when all the data (n = 8) were combined (r = 0.876**). However, there was no significant difference in the radiation use efficiency (RUE) of the whole growth period between Liangyoupeijiu and Nipponbare for both years. Even though the growth duration was shorter, Takanari, an indica/japonica cross-bred variety, showed a similar yield to Liangyoupeijiu in both years. The mean RUE of the whole growth period was significantly higher in Takanari, 1.60 and 1.64 g MJ−1 in 2003 and 2004, respectively, than in Liangyoupeijiu, which had a RUE of 1.46 and 1.52 g MJ−1 in 2003 and 2004, respectively. The high grain yield of Takanari was mainly due to its high RUE compared with Liangyoupeijiu and its large P0 and ΔT. Our result showed that the high grain yield of Liangyoupeijiu was due to its large biomass accumulation before heading, which resulted from its large LAD rather than its RUE.  相似文献

Intensive rice farming in aerobic soil, referred to herein as aerobic rice, can greatly reduce the water input compared to that of flooded rice cultivation. The objective of this study was to compare the potential productivity of aerobic rice and flooded rice using high-yielding varieties at two locations in Japan in two successive years. In aerobic fields, the total amount of water supplied (irrigation plus rainfall) was 800–1300 mm. The soil water potential at 20-cm depth averaged between −15 and −30 kPa each growing season, but frequently reached −60 kPa. The average yield under aerobic conditions was similar to or even higher than that achieved with flooded conditions (7.9 t ha−1 in 2007 and 9.4 t ha−1 in 2008 for aerobic versus 8.2 t ha−1 for flooded). The average water productivity under aerobic conditions was 0.8–1.0 kg grain m−3 water, slightly higher than common values in the literature. The super-high-yielding cultivar Takanari achieved yields greater than 10 t ha−1 with no yield penalty under aerobic conditions in 3 out of 4 experiments. The favorable agronomic characteristic of Takanari was its ample sink capacity (grain number × grain weight). In conclusion, high-productivity rice cultivation in aerobic soil is a promising technology for water conservation. With continued breeding, future aerobic rice varieties will possess large numbers of spikelets and sufficient adaptation to aerobic conditions such that they will consistently achieve yields comparable to the potential yield of flooded rice.  相似文献

We developed both a cryopreservation method for Japanese sea cucumber spermatozoa and an artificial fertilization method using post‐thaw spermatozoa. Twenty per cent dimethyl sulfoxide (DMSO), 16% foetal bovine serum, and 64% artificial seawater were suitable cryodiluent, and the diluent was pre‐cooled to 0°C. Semen was diluted with the solution and enclosed in a 250 μl straw, cooled to ?50°C at 10.4 ± 0.4°C/min, and immediately immersed in liquid nitrogen. Although this method showed the highest post‐thaw motility in all the conditions we examined, its post‐thaw motility was still less than approximately 15%. Artificial fertilization was carried out by adding post‐thaw semen with a cryodiluent to the oocytes. The fertilization rate of 200 oocytes/ml seawater increased with the amount of post‐thaw semen from 1 to 5 μl but showed a significant decrease at 25 μl. This decrease was considered to be due to DMSO in the cryodiluent, because the fertilization rate of the fresh semen decreased sharply when the DMSO concentration around the oocytes was 1.0% or more. Further improvement in increasing post‐thaw motility and lowering the cryoprotectant concentration is necessary for commercial‐scale artificial fertilization.  相似文献

Mycelial protoplasts of two compatible monokaryons ofLentinula edodes were prepared, and electro-fusion was undertaken. Sixteen dikaryons isolated after the electrofusion were tested for their fruiting abilities in comparison to the two dikaryons obtained by reciprocal mycelial mating. There were significant differences in fruiting body yield among the fusants, and some of them produced more fruiting bodies than did normally mated dikaryons. Although these dikaryons were divided into five genetically different groups by isozyme analyses and a cluster analysis, we could not find clear differences among the groups in terms of fruiting body yield, mycelial growth rate, or degree of wood decay. It appeared that fruiting body yield correlated positively with the mycelial growth rate but negatively with the degree of wood decay.  相似文献

We prepared carpet cleaners containing three wood oils extracted from Thujopsis dolabrata Sieb. et Zucc. var. hondai Makino, Chamaecyparis obtusa Endl., and Chamaecyparis taiwanensis Masamune et Suzuki and studied their effects on mites and perfumerists' impressions. The oil concentrations were set at 0.1%, 0.2%, 0.4%, 0.8%, and 1.6%. The effects on Dermatophagoides pteronyssinus Trouessart were investigated. The sensory evaluations were conducted by seven male perfumerists using the SD method and they were asked to describe freely their impressions of the scents. These results showed that: (1) all three types of wood oil had a significant effect on making D. pteronyssinus inactive at 0.1% concentration; and (2) the wood oil of T. dolabrata evoked refreshing, natural, rich, and intellectual feelings; C. obtusa oil evoked vivacious, rich, and intellectual feelings; and C. taiwanensis evoked refreshing, natural, rich, rough, masculine feelings. According to the test subjects, T. dolabrata evoked woody and earthy impressions; C. obtusa evoked woody, citrus, and pine-resin impressions; and C. taiwanensis evoked woody, citrus, and medical impressions. Many issues remain to be investigated, but the inclusion of wood oil in carpet cleaners offered both good mite control and a pleasant aroma to humans. This study suggests a new potential for using these wood oils.  相似文献

Square lumber specimens of laser-incised Douglas fir (Pseudotsuga menziesii Franco) were treated with steam before dipping. Two types of steam (saturated steam and superheated steam), three steam-injection times (5, 10, and 20 min), four different time intervals (moving time) between steam treatment and dipping (immediate, 3, 10, and 30 min), and four different dipping times (0.5, 1, 3, and 12 h) were used in the study. The maximum absorption was 480 kg/m3 when saturated steam was injected for 20 min and the specimen was immediately dipped into liquid for 12 h. Samples treated with this condition not only absorbed the maximum amount of liquid but also penetrated over 83.4% and 87.3% of the total area along and across the grain, respectively. The optimum conditions were then applied to laser-incised sugi (Cryptomeria japonica D. Don) and Japanese larch (Larix leptolepis Gordon) where the absorption of liquid was 415 and 187 kg/m3, respectively. It was shown that initial moisture content below the fiber saturation point was good for passive impregnation. The absorption of liquid and its distribution in wood indicates that it can be a good preservative treatment method for impermeable woods.  相似文献
