To investigate the effect of CO2 laser incising under five drying methods on drying characteristics of Sugi lumber, the squares (120 mm × 120 mm) of Sugi lumber with length of 650 mm were used. A half of samples were incised by CO2 laser with incising density of 2,500 holes/m2. Five types of drying methods were used: microwave drying, steam injection drying, and three combinations of microwave heating and steam injection drying. Steam injection drying was conducted by injecting superheated steam of 120 °C through a perforated plate heated to 140 °C of an injection press. Microwave was irradiated with the power of 3 kW at frequency of 2.45 GHz. The results indicated that incising helps heat through a specimen and thus the whole temperature raised rapidly, which was up to threefolds compared to that of no-incised one. Incised specimens dried by a combination of microwave heating for 1 h and steam injection showed the highest drying rate, which was up to 5.3 %/h. Incising and microwave heating contributed positively to dry lumber under more uniform distribution of moisture content and to reduce surface and internal checks. Incised specimen dried by microwave showed the most uniform distribution of moisture content without surface and internal checks.  相似文献

In this study, the ability of passive impregnation of Douglas fir (Pseudotsuga menziesii Franco) lumber was compared with a conventional full cell method. With an incising density of 10,000 holes/m2 carried out by CO2 laser, square Douglas fir lumber was treated by both passive impregnation and full cell method for 6 h and 50 min. Different incising densities were also used in passive impregnation to examine the optimum incising density. It was observed that there was no significant difference between these two methods regarding absorption of liquid, moisture content after dipping and penetrated area both at cross and longitudinal sections. However, the value was higher in case of full cell method. Regarding different incising densities, absorption and penetration of liquid was similar for 10,000 and 7,500 holes/m2 but it was low for 5,000 holes/m2. Therefore, it can be concluded that an incising density of 7,500 holes/m2 is the optimum for passive impregnation. The absorption of liquid and its distribution in wood indicates that this non-pressure passive impregnation method can be a good preservation method for impermeable lumber and can be an alternative to full cell method.  相似文献

To investigate the influence of heating and drying history on the microstructure of dry wood, in addition to the dynamic viscoelastic properties, CO2 adsorption onto dry wood at ice.water temperature (273 K) was measured, and the micropore size distribution was obtained using the Horvath-Kawazoe (HK) method. Micropores smaller than 0.6 nm exist in the microstructures of dry wood, and they decreased with elevating out-gassing temperature and increased again after rewetting and drying. Dry wood subjected to higher temperatures showed larger dynamic elastic modulus (E′) and smaller loss modulus (E″). This is interpreted as the result of the modification at higher temperature of the instability caused by drying. Drying history influenced the number of micropores smaller than 0.6 nm in dry wood not subjected to high temperature, although the difference in the number of micropores resulting from the drying history decreased with increasing out-gassing temperature. A larger number of micropores smaller than 0.6 nm exist in the microstructure of dry wood in more unstable states, corresponding to smaller E′ and larger E″ than in the stable state. Consequently, unstable states are considered to result from the existence of temporary micropores in the microstructures of dry wood, probably in lignin. To investigate the changes in microstructures of wood with elapsed time in the environment, CO2 adsorption onto dry wood was measured at ice-water temperature (273 K) for samples aged from 0.1 years to over 1000 years. The micropore size distribution was obtained using the Horvath-Kawazoe method. Micropores smaller than 0.6 nm in wood decreased in number with elapsed time in the environment, and a negative correlation was found between cumulative pore volume for pores smaller than 0.6 nm and elapsed time in the environment. Cumulative pore volume in the 1000-year sample was almost half of that in the 0.1- year sample. Micropores smaller than 0.6 nm in wood with a few decades or more of elapsed time increased in number after rewetting and drying. Consequently, microstructures of wood with longer time elapsed in the environment were considered to be more stable, because of longer-term thermal motion and possibly more repeated moisture adsorption and desorption and/or temperature variation in the environment.  相似文献

The influences of heating history, cooling method, and cooling set on microstructures and the mechanical properties of water-swollen wood were studied by measuring viscoelastic properties and dimensional changes while elevating temperatures between 20°C and 90°C. Both the viscoelastic properties and dimensional changes of waterswollen wood in the first heating process were quite different from those in the other heating processes. The results revealed that the molecular state of green wood around room temperature was stabilized and could not return to this state if drying or heating was carried out. Cooling methods greatly affected the viscoelastic properties, while they hardly affected dimensional changes when the temperature was elevated. Localized stress in the microstructures of water-swollen wood produced by quenching affected the mechanical properties in the heating process, while external stress less than the proportional limit caused by a cooling set had no effect. This revealed that much greater localized stress linked to the instability of waterswollen wood than the external stress in relation to the cooling set occurred. We investigated the temporal changes in creep and stress relaxation behavior in both microscopic crystalline cellulose and macroscopic strain of wood specimen using Japanese cypress (Chamaecyparis obtusa Endl.) to understand the viscoelastic properties of wood cell walls. Specimens 600 µm in thickness were observed by the X-ray diffraction and submitted to tensile load. The crystal lattice strain of (004) plane and macroscopic strain of specimen were continuously detected during creep and stress relaxation tests. It was found that the creep compliance based on macroscopic strain showed a gradual increase after instantaneous deformation due to loading and then the parts of creep deformation remained as permanent strain after unloading. On the other hand, crystal lattice strain showed a different behavior for macroscopic strain; it kept a constant value after instantaneous deformation due to loading and then increased gradually after a certain period of time. These differences between macroscopic and microscopic levels were never found in the stress relaxation tests in this study. Relaxation modulus at the macroscopic level only showed a decreasing trend throughout the relaxation process. However crystal lattice strain kept a constant value during the macroscopic relaxation process. In addition, the microfibril angle (MFA) of wood cell wall has a role of mechanical behavior at microscopic level; crystal lattice strains were smaller with increasing MFA at both creep and relaxation processes. Creep compliance and stress relaxation modulus at the macroscopic level decreased and increased with increasing MFA, respectively. Our results on the viscoelastic behavior at microscopic level evidenced its dependency on MFA.  相似文献

To investigate micropores and mesopores in the cell walls of dry wood, CO2 gas and N2 gas adsorption onto dry wood were measured at ice-water temperature (273 K) and liquid nitrogen temperature (77 K). CO2 gas adsorption isotherms obtained were used for determining micropore volumes smaller than 0.6 nm by the HK method (Horvath-Kawazoe method), and N2 gas adsorption isotherms obtained were used for determining the mesopore volume between 2 nm and 50 nm by the Barrett-Joyner-Halenda (BJH) method. Micropores and mesopores existed in cell walls of dry wood, and the cumulative pore volume was much larger for micropores than for mesopores. Micropores in the cell wall of dry wood decreased with elevating heat treatment temperature, and the decreased micropore was reproducible by wetting and drying. Mesopores did not decrease so much with elevating heat treatment temperature. Micropore volumes for the softwood Hinoki and the hardwood Buna were compared. A larger amount of micropores existed in hardwood Buna than in softwood Hinoki, and this relationship was considered to correspond to the difference in thermal softening properties for lignin in water-swollen Hinoki and Buna. This result probably indicates that micropores in the cell walls of dry wood relate to the structure of lignin.  相似文献

Effective preselection of sex has been accomplished in several species of livestock and also in humans using the flow cytometric sperm sorting method. A guaranteed high sorting accuracy is a key prerequisite for the widespread use of sperm sexing. The standard validation method is flow cytometric remeasurement of the DNA content of the sexed sperm. Since this method relies on the same instrument that produced the original sperm separation, it is not truly independent. Therefore, to be able to specifically produce either male or female offspring in the dog, we developed a method of direct visualization of sex chromosomes in a single sperm using fluorescence in situ hybridization (FISH) as a validation method. Denaturation of canine spermatozoa by immersion in 1 M NaOH for 4 min yielded consistent hybridization results with over 97% hybridization efficiency and a good preservation of sperm morphology. There was no significant difference between the theoretical ratio (50:50) and the observed ratio of X- and Y-chromosome-bearing spermatozoa in any of the three dogs. In addition, the mean purities of flow-sorted sex chromosomes in spermatozoa of the three dogs were 90.8% for the X chromosome fraction and 89.6% for the Y chromosome fraction. This sorting was evaluated by using the dual color FISH protocol. Therefore, our results demonstrated that the FISH protocol worked reliably for both unsorted and sexed sperm samples.  相似文献

Lions (Panthera leo) breed well under captivity, so contraception has been commonly conducted for population management, leading to a demand for a less invasive and reversible contraceptive approach in lions. In this study, we examined the efficacy of a commercial gonadotropin-releasing hormone vaccine as a method of suppressing reproductive activity in a sexually matured female lion. Under behavioral restraint, the vaccine was injected twice (days 0 and 109). After the initial vaccination, ovarian activity is still observed. After the second vaccination, contraceptive effect was confirmed for 246 days until restart of estrous cycles. We confirmed only a slight swelling around the injection site after the second vaccination. This study may suggest an alternative option for a contraceptive method in lions.  相似文献

Intrathecal (IT) immunization involves injecting antigens directly into the intraventricular or subarachnoid spaces, or brain, to induce antigen-specific antibodies (Ab) in the cerebrospinal fluid (CSF). In the present study, rabbits were immunized IT with inactivated rabies virus to investigate the origins of CSF Ab. The time course of Ab induction and tumor necrosis factor-alpha expression suggested the possibility that the CSF Ab originated in the serum. In addition, Ab-producing cells infiltrated around the blood vessels of the brain, suggesting local production of Ab within the central nervous system (CNS). Furthermore, subcutaneous (SC) immunization prior to IT immunization induced a rapid and magnified Ab response in the CSF compared with IT immunization alone. These results were confirmed by the fact that mice immunized SC prior to IT were more resistant to intracerebral challenge with rabies virus than mice immunized via the IT route alone. Taken together, these results suggest that combined SC and IT immunization is a more effective vaccination protocol for prophylaxis and treatment of rabies.  相似文献
