The effects of laser incising on the surface of heat-affected zones (HAZs) and liquid uptake of wood were examined. Deep pin holes were incised on the tangential section of the wood with a laser beam of 1 kW in power with varied pulse widths, and the HAZs of the holes were investigated using scanning electron microscopy. The length of liquid uptake passing through HAZs surfaces was then measured. It was observed that the HAZs were unable to maintain normal porous structure, and smooth surfaces such as that of wood charcoal were also not observed. This is because the composite of the middle lamella completely melted, which led to a decrease in the length of liquid uptake. In addition, the release of high gas-pressure and thermal energy during the laser-incising process did not damage the tylose and the aspirated pit in the longitudinal direction. Because of this, the liquid movement remained blocked. However, the incising holes provided additional liquid-intake points, resulting in greater intake capacity. The present study aimed to clarify changes of oxidative stress and antioxidative
functions in treadmill-exercised Thoroughbred horses (n=5, 3 to 7 years old), using
recently developed techniques for measurement of serum d-ROMs for oxidative stress, and
BAP for antioxidative markers. Also, the effect of nasogastric administration of
hydrogen-rich water (HW) or placebo water preceding the treadmill exercise on these
parameters was examined. Each horse was subjected to a maximum level of treadmill exercise
in which the horses were exhausted at an average speed of 13.2 ± 0.84 m/sec. Blood samples
were taken 4 times, immediately before the intake of HW or placebo water at 30 min
preceding the treadmill exercise, immediately before the exercise (pre-exercise),
immediately after the exercise (post-exercise) and at 30 min following the exercise. In
all horses, both d-ROMs and BAP values significantly increased at post-exercise. The
increase in d-ROMs tended to be lower in the HW trial, as compared to the placebo trial at
pre-exercise. The increase in BAP was considerable at approximately 150% of the
pre-exercise values in both the HW and placebo treatment trials. The BAP/d-ROMs ratio was
significantly elevated at post-exercise in both treatment trials, while a significant
elevation was also observed at pre-exercise in the HW trial. BAP, d-ROM, and the BAP/d-ROM
ratio tended to decline at 30 min after the exercise, except BAP and BAP/d-ROMs in the
placebo trial. These results demonstrate that the marked elevation of oxidative stress and
anitioxidative functions occurred simultaneously in the intensively exercised horses, and
suggest a possibility that HW has some antioxidative efficacy. 相似文献

Two complementary DNA (cDNA) libraries were constructed from phyllosomas and hemocytes of adult Japanese spiny lobster Panulirus japonicus and a total of 2,673 expressed sequence tags (ESTs) were obtained. After assembly and clustering, 450 and 458 unique sequences
were found from the phyllosoma and hemocyte cDNA libraries, respectively. Of these, 114 and 220 ESTs showed significant homologies
with known genes in the National Centre for Biotechnology Information (NCBI) database. The remaining sequences were of unknown
function. Immune-related genes found in this study include lectin, proteinase inhibitor, prophenoloxidase, heat-shock protein,

We investigated the possibility to reduce the usage of Blasin®Flowable (BF), a disinfectant inhibiting the germination and appressorium formation of Pyricularia oryzae Cavara conidia, by using carbon dioxide microbubbles (CO2MB). Germination was significantly inhibited by 10 000-fold diluted BF solution containing CO2MB generated by the decompression-type generator compared to CO2 millibubbles (CO2MMB) and CO2MB generated by the gas-water circulating-type generator. Appressorium formation in the 10 000-fold diluted BF solution containing both CO2MBs was less than that in CO2MMB. Scanning electron microscopy showed wrinkles and dents on the surface of conidia treated with 5 000-fold diluted BF solution containing both CO2MBs. Via transmission electron microscopy, we observed the expansion of the vacuole and the intracellular space and bloated or absent lipid granules in the conidia treated with BF solution containing both CO2MBs. Our results show that inhibition of the conidium germination and appressorium formation of P. oryzae Cavara by 10 000-fold diluted BF solution could be achieved by using the decompression-type CO2MB. 相似文献

The difference in yields of cultivars may be causing difference in soybean yield between Japan and the USA. The objective of this study was to identify the effect of the cultivar on dry matter production and to reveal the key factors causing the differences in yield by focusing utilization of solar radiation in recent Japanese and US soybean cultivars. Field experiments were conducted during two seasons in Takatsuki, Japan (34°50′), and in a single season in Fayetteville (36°04′), AR, USA. Five Japanese and 10 US cultivars were observed under near-optimal conditions in order to achieve yields as close to their physiological potential as possible. The seed yield and total aboveground dry matter (TDM) were measured at maturity as long as radiation was intercepted by the canopy. The seed yield ranged from 3.10t ha?1 to 5.91t ha?1. Throughout the three environments, the seed yield of US cultivars was significantly higher than that of Japanese cultivars. The seed yield correlated with the TDM rather than the HI with correlation coefficients from .519 to .928 for the TDM vs. .175 to .800 for the HI, for each of the three environments. The higher TDM of US cultivars was caused by a higher radiation use efficiency rather than higher total intercepted radiation throughout the three environments. The seasonal change in the TDM observed in four cultivars indicated that dry matter productivity was different between cultivars, specifically during the seed-filling period. 相似文献

MytiLec; a novel lectin isolated from the Mediterranean mussel (Mytilus galloprovincialis); shows strong binding affinity to globotriose (Gb3: Galα1-4Galβ1-4Glc). MytiLec revealed β-trefoil folding as also found in the ricin B-subunit type (R-type) lectin family, although the amino acid sequences were quite different. Classification of R-type lectin family members therefore needs to be based on conformation as well as on primary structure. MytiLec specifically killed Burkitt''s lymphoma Ramos cells, which express Gb3. Fluorescein-labeling assay revealed that MytiLec was incorporated inside the cells. MytiLec treatment of Ramos cells resulted in activation of both classical MAPK/ extracellular signal-regulated kinase and extracellular signal-regulated kinase (MEK-ERK) and stress-activated (p38 kinase and JNK) Mitogen-activated protein kinases (MAPK) pathways. In the cells, MytiLec treatment triggered expression of tumor necrosis factor (TNF)-α (a ligand of death receptor-dependent apoptosis) and activation of mitochondria-controlling caspase-9 (initiator caspase) and caspase-3 (activator caspase). Experiments using the specific MEK inhibitor U0126 showed that MytiLec-induced phosphorylation of the MEK-ERK pathway up-regulated expression of the cyclin-dependent kinase inhibitor p21, leading to cell cycle arrest and TNF-α production. Activation of caspase-3 by MytiLec appeared to be regulated by multiple different pathways. Our findings, taken together, indicate that the novel R-type lectin MytiLec initiates programmed cell death of Burkitt’s lymphoma cells through multiple pathways (MAPK cascade, death receptor signaling; caspase activation) based on interaction of the lectin with Gb3-containing glycosphingolipid-enriched microdomains on the cell surface. 相似文献

Previously, we reported α(2)-macroglobulin (α(2)M) to be a novel marker characteristic of rat hepatocellular preneoplastic and neoplastic lesions negative for hitherto well-established markers. In the present study, we further examined other candidate markers with specificity for the same type of lesions. Glutathione S-transferase-placental form (GST-P)-negative hepatocellular altered foci (HAF) were generated using a two-stage (initiation and promotion) carcinogenesis protocol with N,N-diethylnitrosamine (DEN) and either Wy-14,643 or clofibrate, two peroxisome proliferators. Microarray analysis using total RNAs isolated from laser-microdissected GST-P-negative HAF (amphophilic cell foci) and adjacent normal tissues was conducted along with immunohistochemistry and real-time RT-PCR. Staining for glucose-regulated protein 78 (GRP78) was detected in GST-P-negative HAF and hepatocellular adenomas, and slightly increased GRP78 mRNA expression was observed in the lesions by real-time RT-PCR analysis. Thus, an early increase of GRP78 expression in hepatocarcinogenesis is likely a feature of the amphophilic subset of HAF. 相似文献

In this study, to summarize the changes of thermal-softening behaviors of wood and acetylated wood due to differences in the kinds of swelling liquids, the following measurements were conducted. Untreated and acetylated wood samples were swollen by various liquids and the temperature dependences of the dynamic viscoelastic properties were measured after the heating and cooling histories were unified among the samples. The results obtained are as follows. Untreated samples swollen by high-polarity liquid had lower peak temperature of tanδ, however acetylated samples had higher peak temperature of tanδ than those of untreated wood. On the other hand, untreated wood samples swollen by low-polarity liquid had higher peak temperature of tanδ, however acetylated samples had lower peak temperature of tanδ than those of untreated wood. The amount of swelling is determined by interaction between wood and liquid due to proton-accepting power and molar volumes of liquid and so on, therefore the peak temperature of tanδ and degree of reduction in dynamic elastic modulus (E´) with increasing temperature were corresponded to the amount of swelling. 相似文献
