A survey of 73 dairy farms in south-western Victoria was conducted to assess the cost and mean herd incidence of foot lameness for the period from calving to the end of November, 1985, and to identify the herd, management and environmental factors associated with foot lameness. The mean herd size was 125 cows (range 82 to 220). Lameness occurred in 64 (88%) herds, and the mean herd incidence was 7.0% (range 0.0 to 30.9%). The main clinical signs associated with lameness were the presence of overworn and/or bruised soles, or stones lodged in the interdigital cleft. Factors associated with lameness were: property and herd size, age of cow, bail feeding, voluntary entry into the bails, and features of the farm track including its length, the presence of steep slopes, the type of surface material, presence and treatment of broken sections and maintenance including rolling history. The association of these factors with specific clinical signs was examined. The mean cost was estimated to be $42.90 per lame cow due to loss of production, treatment, the culling or death of lame cows, and extra man hours spent managing lame cows. It was concluded that the site, construction, maintenance and use of the farm track were of major importance to the incidence of lameness in herds in this area and recommendations for reducing lameness are made.  相似文献

The distribution and morphology of fibrous astrocytes in the cervical spinal cord of normal horses and horses with chronic compressive myelopathy were demonstrated using immunohistochemical staining for glial fibrillary acidic protein. In the spinal cord from normal horses, astrocytes with stellate cell bodies and short processes were irregularly distributed in grey matter. In the white matter, their cell bodies were small and angular in areas adjacent to grey matter and larger and more stellate-shaped in the subpial area. Astrocyte processes were fine, and evenly distributed in a predominantly radial pattern in transverse sections of cord. Gliosis was marked in the spinal cords of horses with cervical compressive myelopathy. In the grey matter at the level of compression astrocytes were often enlarged and rounded, with short, blunt processes, but the gliosis was generally mild. In the white matter, gliosis was obvious in areas of nerve fibre swelling and degeneration at the level of compression and in areas of ascending and descending Wallerian degeneration. The fine radial pattern of astrocyte fibres was replaced by a dense, irregular arrangement. Gliosis persisted in the cords of chronically affected horses after active nerve fibre degeneration had subsided. The areas of gliosis coincided with the areas of Marchi staining for degenerating myelin and with areas of myelin loss in osmium tetroxide post-fixed tissue. Histological observations were consistent with astrocytes replacing areas of extracellular space that remained after nerve fibre degeneration. it is concluded that astrocytic gliosis is a prominent and persistent alteration of the spinal cord of horses with chronic cervical compressive myelopathy.  相似文献

Data from species other than cattle indicate that ghrelin and GH secretagogue receptor (GHS-R) could play a key role in fat deposition, energy homeostasis, or glucose metabolism by directly affecting liver and adipose tissue metabolism. Beef steers (n = 72) were used to test the hypothesis that plasma ghrelin and leptin concentrations and abundance of the GHS-R in liver, muscle, and adipose tissues differ in steers exhibiting differences in composition of gain. At trial initiation (d 0), 8 steers were slaughtered for initial carcass composition. The remaining 64 steers were stratified by BW, allotted to pen, and treatment was assigned randomly to pen. Steers were not implanted with anabolic steroids. Treatments were 1) a low-energy (LE) diet fed during the growing period (0 to 111 d) followed by a high-energy (HE) diet during the finishing period (112 to 209 d; LE-HE) or 2) the HE diet for the duration of the trial (1 to 209 d; HE-HE). Eight steers per treatment were slaughtered on d 88, 111, 160, and 209. Carcass ninth, tenth, and eleventh rib sections were dissected for chemical composition and regression equations were developed to predict compositional gain. Liver, muscle, and subcutaneous adipose tissues were frozen in liquid nitrogen for subsequent Western blotting for GHS-R. Replicate blood samples collected before each slaughter were assayed for ghrelin and leptin concentrations. When compared at a common compositional fat end-point, the rate of carcass fat accretion (g·kg of shrunk BW(-1)) was greater (P < 0.001) in HE-HE steers whereas the rate of carcass protein accretion (g·kg of shrunk BW(-1)) was less (P < 0.001) compared with LE-HE steers. When compared at a common compositional fat end-point, plasma leptin, ghrelin, and insulin concentrations were greater (P < 0.05) for HE-HE compared with LE-HE steers. Abundance of the GHS-R, to which ghrelin binds, increased over time in liver and adipose tissue but did not differ as a result of treatment. Plasma ghrelin concentrations were increased for cattle continuously fed the HE diet as they became increasingly fatter; however, abundance of the GHS-R in liver, muscle, and subcutaneous adipose tissue was not different between treatment groups. The role of ghrelin in cattle metabolism warrants further investigation as it could have a significant effect on composition of BW gain, feed efficiency, and metabolic disorders such as ketosis and fatty liver.  相似文献

We determined the effects of short-term fasting and refeeding on temporal changes in plasma concentrations of leptin, insulin, insulin-like growth factor- 1 (IGF-1), growth hormone (GH), glucose, and nonesterified fatty acids (NEFA), in early lactating cows, non-lactating pregnant cows, and postpubertal heifers. In experiment 1, Holstein cows in early lactation were either fed ad libitum (Control, n=5) or feed deprived for 48 h (Fasted, n=6). Plasma leptin, insulin, and glucose concentrations rapidly declined (P<0.05) within 6h, and IGF-1 by 12h, but all these variables sharply returned to control levels (P>0.10) within 2h of refeeding. Plasma NEFA and GH concentrations were elevated (P<0.05) by 4 and 36 h of fasting and returned to control levels (P>0.10) by 8 and 24h after refeeding, respectively. In experiment 2, four ruminally cannulated pregnant non-lactating Holstein cows were used in a cross-over design and were fasted for 48 h (Fasted) or fasted with partial evacuation of rumen contents (Fasted-Evac). The plasma variables measured did not differ (P>0.10) between Fasted and Fasted-Evac cows. Plasma leptin, insulin, and IGF-1 concentrations were reduced by 10, 6, and 24h of fasting, respectively, in Fasted-Evac cows; and these variables were reduced by 24h in Fasted cows (P<0.05). Plasma glucose levels were reduced (P<0.05) by 48 h of fasting in both groups of fasted animals. Plasma NEFA and GH levels were increased (P<0.05) by 12 and 48 h of fasting, respectively. In experiment 3, postpubertal Holstein heifers were either fed ad libitum (Control, n=4) or feed deprived for 72 h (Fasted, n=5). Concentrations of leptin, insulin, IGF-1, and glucose in plasma were reduced (P<0.05) by 24, 10, 24, and 48 h of fasting, respectively. Plasma NEFA concentrations increased (P<0.05) by 4h, of fasting while GH levels were not significantly (P>0.10) affected by fasting. Collectively, our data provide evidence that plasma leptin concentrations are reduced with short-term fasting and rebound on refeeding in dairy cattle with the response dependent on the physiological state of the animals. Compared to the rapid induction of hypoleptinemia with fasting of early lactation cows, the fasting-induced hypoleptinemia was delayed in non-lactating cows and postpubertal heifers.  相似文献

We tested the hypothesis that recombinant ovine leptin would attenuate the acute effects of neuropeptide Y (NPY) on secretion of GH and gonadotropins (LH and FSH) in cows. Ovariectomized cows (n=6) fitted with third ventricle guide cannulas were assigned randomly to each of three groups in a Latin square arrangement: (1) control; saline treatment only, (2) NPY; saline followed by NPY, and (3) L-NPY; leptin pretreatment followed by NPY. Treatments were: s.c. injection of saline or leptin (30 microg/kg BW) at time 0, i.v. injection of saline or leptin (30 microg/kg BW) at 70 min, and intracerebroventricular (i.c.v.) injection of saline or NPY (500 microg) at 90 min. Plasma leptin was elevated (P<0.01) at least four-fold throughout the experiment in the L-NPY group. Mean plasma concentrations of LH declined within 1 h and were lower (P<0.03) than controls in both the NPY and L-NPY groups beginning 2 h after NPY injection. An acute increase in plasma concentrations of GH was observed within 1 h after NPY in the NPY group and mean values were greater (P<0.01) than controls. However, in the L-NPY group, leptin pretreatment attenuated the NPY effect on GH. Treatments had no effect on FSH secretion. Results confirm suppressive and stimulatory effects of NPY on LH and GH secretion, respectively, and indicate that leptin can attenuate the acute effects of NPY on GH secretion in cattle.  相似文献

The influences of body condition score (BCS) at calving and postpartum nutrition on endocrine and ovarian functions, and reproductive performance, were determined by randomly allocating thin (mean BCS = 4.4 +/- 0.1) or moderate condition (mean BCS = 5.1 +/- 0.1) Angus x Hereford primiparous cows to receive one of two nutritional treatments after calving. Cows were fed to gain either 0.45 kg/d (M, n = 17) or 0.90 kg/d (H, n = 17) for the first 71 +/- 3 d postpartum. All cows were then fed the M diet until 21 d after the first estrus. A replication (yr 2; M, n = 25; H, n = 23) was also used to evaluate reproductive characteristics. Concentrations of IGF-I, leptin, insulin, glucose, NEFA, and thyroxine were quantified in plasma samples collected weekly during treatment and during 7 wk before the first estrus. Estrous behavior was detected by radiotelemetry, and luteal activity was determined based on concentrations of progesterone in plasma. All cows were bred by AI between 14 and 20 h after onset of estrus, and pregnancy was assessed at 35 to 55 d after AI by ultrasonography. Cows that calved with a BCS of 4 or 5 had similar endocrine function and reproductive performance at the first estrus. During treatment, H cows gained BW and increased BCS (P < 0.01), and had greater (P < 0.05) concentrations of IGF-I, leptin, insulin, glucose, and thyroxine in plasma than M cows. However, during the 7 wk before the first estrus, plasma concentrations of IGF-I, leptin, insulin, glucose, NEFA, and thyroxine were not affected by time. Cows previously on the H treatment had a shorter (P < 0.01) interval to first postpartum estrus and ovulation, and a larger dominant follicle (P < 0.01) at first estrus, than M cows, but duration of estrus and the number of mounts received were not influenced by nutrient intake. Pregnancy rate at the first estrus was greater (P < 0.03) for H (76%, n = 38) than for M (58%, n = 33) cows. Increased nutrient intake after calving stimulated secretion of anabolic hormones, promoted fat deposition, shortened the postpartum interval to estrus, and increased pregnancy rate at the first estrus. Concentrations of IGF-I and leptin in plasma were constant during 7 wk before the first estrus, indicating that acute changes in these hormones are not associated with the resumption of ovarian function in primiparous beef cows.  相似文献
