Heartwood of pernambuco (Guilandina echinata Spreng, synCaesalpinia echinata Lam.), which has been used as material for violin bows, was extracted by soaking in water, and the obtained extractives were analyzed. The main components of the extractives were identified to be protosappanin B and brazilin. In particular, protosappanin B occupied about 40% of the pernambuco extractives. The loss tangent (tan) of spruce wood impregnated with protosappanin B decreased markedly, the same as that of specimens impregnated with extractives before being purified. Canine mast cell tumors (MCTs) are the most common cutaneous tumors in the dog. They have a wide range of behaviour, which can make these tumors challenging to treat. Recently, mutations in c-kit proto-oncogene have been identified in several canine MCTs. Imatinib is the first member of a new class of agents that act by inhibiting particular tyrosin kinase enzymes, including KIT which is a product of the c-kit. In this study the efficacy of imatinib to reduce or abolish canine MCT [CMC-1] using xenografted MCT in severe combined immunodeficient [SCID] mice was evaluated. Imatinib was administered at doses of 200 mg/kg and 100 mg/kg once a day for one week. The antitumor responses in SCID mice with CMC-1 xenografts following treatment with imatinib were observed. Significant tumor regression occurred with 100 mg/kg on days 7, 10, 14 and 21, and 200 mg/kg on all days. Our results indicate that imatinib is effective against canine mast cell tumor in mouse xenograft models. Canine MCTs might be a potential target for imatinib therapy. 相似文献

A rapid and efficient DNA extraction method was developed for detecting mastitis pathogens in milk. The first critical step involved cell wall disruption by bead-beating, as physical disruption using beads was more effective for DNA extraction from Gram-positive bacteria, such as Staphylococcus aureus, than enzymatic disruption using proteinase K. The second critical step involves the use of acetic acid and ammonium sulfate in the purification process, as these reagents effectively and efficiently remove the lipids and proteins in milk. Using these methods, DNA suitable for loop-mediated isothermal amplification was obtained within 30 min. Also, the rapid and sensitive detection of S. aureus in milk was possible at levels as low as 200 cfu/ml. 相似文献

Epsilon-polylysine micro particles (SGEPL) and polyethyleneimine micro particles (SGPEI) were developed by the addition of a hydrophobic group and the immunological characterization of these micro particles and aluminum hydroxide (ALUM) was investigated. BALB/c mice were injected intraperitoneally with ovalbumin (OVA) as an antigen and SGEPL, SGPEI or ALUM as an adjuvant. The results showed that the mice injected with SGEPL produced a significant portion of anti-OVA antibody subclass IgG2a in the sera and suppressed interleukin (IL)-4 and IL-5, but enhanced IL-12 and interferon-gamma (IFN-gamma) from the spleen cells. Similar results relating to cytokines were also obtained, even without OVA. Direct stimulation with SGEPL to na?ve BALB/c mouse spleen cells induced IL-12 and IFN-gamma. Both spleen and purified B cells produced IgG1 and IgE after stimulation with IL-4 and the anti-CD40 monoclonal antibody. With the addition of SGEPL, the IgE production from the cells was suppressed as a result of enhanced IFN-gamma production. Furthermore, IgE production was also suppressed in the purified B cells without the influence of IFN-gamma or IL-12. Thus, we suggest SGEPL drives cytokine production to Th1 profile. It will be a novel promising adjuvant based on this viewpoint. 相似文献

We investigated the growth of juvenile common snook (Centropomus undecimalis) reared at 25°C and 28°C and salinities of 0.3, 15, and 32 g L?1. Total length, weight, RNA/DNA, and protein/DNA ratios were determined after 90 days of experiment. Higher growth was observed at 28ºC compared with 25°C, at the same salinity. At 28°C and 15 g L?1 salinity, the weight (25.14 g) of juveniles was twice that of the juveniles reared at the lower temperature. At different salinities, only higher temperature affected growth, with higher weight values obtained at 15 g L?1 in comparison with 0.3 and 32 g L?1. Length was similar at 0.3 and 15 g L?1. The RNA/DNA ratio was greater in juveniles reared at a salinity of 15 g L?1 when compared with 0.3 and 32 g L?1. This study shows that the combination of higher temperature and intermediate salinity promotes better growth of common snook juveniles. 相似文献

OBJECTIVE: Green tea has been widely acknowledged in Japan to induce a pleasurable mental feeling. Recent laboratory studies have suggested positive psychological effects as a result of consuming green tea. The present study examined whether green tea consumption in everyday life in Japan is associated with positive mental health. DESIGN: A cross-sectional study was performed in February-March 2002. SETTING AND SUBJECTS: The subjects of the study consisted of a general population of 600 Japanese aged 20-69 years. Responses of 380 subjects, obtained by home-visit interview, were analysed. The questionnaire inquired about consumption of brewed green tea and other beverages, perceived mental health status, lifestyle and others. The 12-item General Health Questionnaire (GHQ 12) was used for the assessment of mental ill-health (GHQ score >or=4). RESULTS: After adjustments for age, area, perceived mental stress, lifestyle and daily caffeine intake, the consumption of brewed green tea was not statistically associated with any decrease in risk of mental ill-health among either males or females (odds ratio (OR)=0.78, 95% confidence interval (CI)=0.47-1.29 for males; OR=0.77, 95% CI=0.51-1.14 for females). Daily caffeine intake (100 mg) inclusive of green tea, black tea, coffee and other caffeine-containing beverages was associated with a higher risk of mental ill-health among females (OR=1.26, 95% CI=1.01-1.56). CONCLUSIONS: The results provide population-based evidence on the consumption of brewed green tea in everyday life and mental health, together with information on consumption patterns of various beverages and lifestyles. 相似文献

The tonsils of cattle, including palatine tonsils, pharyngeal tonsils, tubal tonsils and lingual tonsils, are designated as specified risk materials (SRM). However, the detailed distribution of lingual tonsils in cattle is unknown. We therefore histologically examined their distribution in 198 tongue specimens from cattle. The examinations confirmed that the presence of lingual tonsils was limited to the tissue of the lamina propria on the dorsal and lateral aspects of the tongue, not reaching the muscular layer below. More than 90% of the lingual tonsils were located between the distribution center of the vallate papillae and the radix linguae (root of the tongue). However, they were also found in the area extending from the lingual torus to the rostral-most vallate papilla in an individual, suggesting that the complete removal of the lingual tonsils requires elimination of the lamina propria extending from the lingual torus to the radix linguae. 相似文献

采用不同浓度来自70%甲醇溶液的麦冬块根提取物对2种产自日本的稻瘟病菌生理小种菌株MAFF305480和MAFF101002的菌丝生长及其接种诱发水稻幼苗稻瘟病发生的抑制效果进行评价.试验表明:含有麦冬块根提取物的原液1/10,1/20,1/40以及1/80浓度都能明显抑制稻瘟病菌小种菌株(MAFF305480和MAFF101002)]菌丝生长,抑制率均56.4%以上,对接种诱发稻瘟病的水稻幼苗处理发现,幼苗植株发病级数为明显下降,抑制率均38.4%以上,并且都随着处理浓度提高抑制效果明显提高.研究结果表明,麦冬块根提取物中含有抑制稻瘟病菌生长的化学物质,并且能直接抑制稻瘟病菌发生和生长. 相似文献

Electroretinography (ERG) is an effective method for the diagnosis of retinal disease. In the dog, dependable ERG recording is difficult without the use of an expensive device like a Ganzfeld full-field stimulator. The International Society for Clinical Electrophysiology of Vision has defined the standard flash stimulus condition (SF) and evaluation of the retina using the b/a ratio in humans. In dogs, evaluation using the b/a ratio has not been reported, whereas the intensity of SF has been defined. In this study, we performed a convenient ERG recording method using a contact lens electrode with a built-in light source (LED-electrode), and confirmed SF as reported previously. ERG recordings were performed on 15 healthy beagle dogs under sedation. We performed bilateral ERG at 12 different intensities after 30 min dark adaptation. After 10 min light adaptation, we recorded single flash cone and flicker cone response using the SF determined in this study. In this study, SF of 3.0 cd/m(2)/sec (6,000 cd/m(2), 0.5 msec) resulted in b/a=2. The intensity for rod response that recorded only the b-wave was 0.0096 cd/m(2)/sec (80 cd/m(2), 0.12 msec). We could achieve ERG for each response easily and smoothly under sedation, and without general anesthesia. Using an LED-electrode, we could perform more quantitative and reproducible ERG examinations than with traditional methods.
